Homogenization of guinea pig liver in isotonic sucrose solution followed by the separation of the subcellular fractions by differential centrifugation releases the liver L-asparaginase (L-asparagine amidohydrolase, EC 3.5.1.1) activity into the supernatant fraction. Electron micrographs of the liver L-asparaginase-antibody complexes, precipitated from the clear supernatant phase by addition of L-asparaginase-specific antiserum, show membrane-liek structures and some amorphous material. The attachment of L-asparaginase to the membrane-like structures is indicated by the
ferritin
-labeled antibody technique. The immunoprecipitates possess low activities of
5'-nucleotidase
, alkaline phosphodiesterase I, NADPH cytochrome c reductase, glucose-6-phosphatase, and acid phosphatase. This observation suggests that L-asparaginase found in the liver supernatant fraction is associated with cytomembrane components. Analysis of guinae pig serum L-asparaginase-antibody complexes is polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate gives three distinct protein bands. These bands correspond to heavy and light chains of rabbit immunoglobulins and the L-asparaginase subunits. Analysis of the liver L-asparaginase-antibody complexes by the above procedure shows similar but more diffuse protein bands.

A Sewall Wright strain-2 guinea pig model producing malignant ascites after injection of a diethylnitrosamine-induced hepatocellular carcinoma cell suspension (Line-10) was used to demonstrate the multilayered settling of tumor cells on the peritoneal surface, frequently followed by the formation of papillary projections and the early invasion in a proliferating submesothelial tissue. At the border of tumor cells and the desmoplastic tissue the malignant cells changed their shape and generally two categories were recognized. Often multilayering, atypical flat cells covered the stromal tissue, while mostly rounded ones invaded using their branched penetration processes, being devoid of cationized

, which was only present on the luminal sides of all cellular elements. Flattened malignant cells, penetrating processes and invading cells lost their microvillous surface pattern. The infiltrating cells were often only detectable with the monoclonal antibody 10 TL 40 and the anti-cytokeratin OV TL 12-5, demonstrating the need for immunohistochemistry in diagnosing solitary invading malignant cells in light microscopy. It appeared that still numerous mesothelial cells were found scattered deeply within the desmoplastic tissue. A method using low concentrations of formaldehyde and dithiothreitol was applied to obtain 'right-side out' luminal plasmalemma-derived vesicles from bovine aortic endothelial cells (EC) in culture, and from human umbilical vein and bovine or porcine aortas perfused ex vivo with the vesiculation solution. Vesicle formation and shedding were examined by phase-contrast microscopy and by transmission (TEM) and scanning electron microscopy (SEM). Vesicles showed the characteristic trilaminar pattern of the unit membrane and did not contain cellular organelles. As detected in freeze-fracture preparations, vesicle membrane displayed intramembrane particles and filipin-detectable cholesterol. Plant lectin concanavalin A conjugated with

(Con A-F) injected i.v. was used for the detection of the specific monosaccharide residues (alpha-D-mannosyl and alpha-D-glucosyl) on the luminal surface of endothelial cells (ECs) in brain micro-blood vessels (MBVs). Both normal mice and animals with mechanically damaged blood-brain barrier (BBB) were used in this study. In addition, the activity of

(5'N), the putative receptor for Con A, was studied cytochemically. Various methodologic experiments indicated that the reaction product formed on the luminal plasmalemma of ECs after incubation of samples in the cytochemical medium for the detection of 5'N activity results from the action of unspecific phosphatase hydrolyzing both specific and nonspecific substrates. The abluminal side of the wall of MBVs seems to be a major location of 5'N activity. Thus, no correlation between cytochemically demonstrable 5'N activity and Con A receptor sites on the luminal surface of ECs was noted. After damage of the BBB, extensive internalization of the luminal plasmalemma forming the limiting membranes of pinocytotic vesicles, vacuoles, and endothelial channel-like structures was observed. This process was represented by a relatively rapid translocation of Con A receptors from luminal surface into the interior of the ECs and to the abluminal side of the vessel wall.

Plasma membranes of vertebrate lens fiber cells contain large numbers of gap junctions that may provide pathways for metabolic cooperation. Characterization of fiber cell gap junctions is thus necessary to understand this function. In this study, plasma membrane fractions were isolated from bovine lens according to established techniques, but without urea, detergents, or proteolytic enzymes. Electron microscopy indicated that isolated plasma membranes with gap junctions form double-membrane vesicles, and gap junctions comprised approximately 35% of the total membrane area in the crude fraction. These vesicles were impermeable to cationized
ferritin
, suggesting that they were sealed, and may be useful for permeability studies. Treatment of the crude fraction with 2.5% beta-mercaptoethanol or dithiothreitol caused reversible separation of junctional membranes, suggesting that disulfide bonds may be important in maintaining gap junction structure. Fractions with varying proportions of gap junctions were isolated using linear sucrose density gradient centrifugation. The proportional area of gap junction membrane versus total membrane in the various fractions ranged from 10% to at least 51%. Cytochemical localization of
5'-nucleotidase
(AMPase) has been investigated in the parotid acinar cells of rats at various stages of exocytic secretion induced by an administration of isoproterenol (IPR). In the resting stage, the acinar cells show AMPase activity located on the baso-lateral and luminal plasmalemma, and in the earliest secretory stage the luminal plasma membranes are devoid of the enzymatic activity. However, these particular regions exhibit AMPase activity during the advanced stages of secretion, and the AMPase positive membranes become absorbed into the cytoplasm by endocytic activity. The absorbed membrane components then seem to be degraded by the action of lysosomes. The intracellular fate of the endocytic vacuoles has been examined by the aid of
ferritin
particles introduced retrogradely through ductal lumina. Ferritin containing vacuoles are distributed in the cytoplasm, and these droplets change into secondary lysosomes. No tracer particles are recognized in the internal space of the Golgi lamella and its associated vesicles. The results suggested that in the exocytic secretion of parotid acinar cells, AMPase originating from plasma membrane intermingles with the membranes derived from secretion granules, and is translocated into cytoplasm by an endocytic mechanism. The internalized membrane components are, at least partly, degraded by lysosome action.

This study determined whether a single 60-mg dose of ferrous sulfate interferes with fractional zinc absorption (FZA) at 7-9 wk of lactation. In a crossover design, 5 exclusively breast-feeding women were given either a single 60-mg iron supplement or no supplement. FZA was measured by analyzing zinc stable isotope tracers ((70)Zn and (67)Zn) in urine samples collected for 7 d after isotope dosing. A 0.7-micromol intravenous (IV) infusion of (70)Zn as ZnCl(2) in saline was followed by a 0.03-mmol oral dose of (67)Zn as ZnCl(2) given with a standardized meal. After a 7-d wash-out period, the supplement given was reversed and a second FZA measurement was taken. 5'-nucleotidase
deficiency is a rare autosomal recessive disorder characterized by haemolytic anaemia, marked basophilic stippling and accumulation of pyrimidine nucleotides within the erythrocytes. The gene encoding for this enzyme (P5'N-1) has been cloned recently, and seven mutations have so far been identified in 11 unrelated families. We describe the haematological and molecular characteristics of six unrelated Italian patients affected by pyrimidine 5'-nucleotidase deficiency (one from northern and five from southern Italy). The sequence of the complete P5'N-1 gene showed the presence of four different new mutations: a missense mutation AAT-AGT at codon 190 (Asn-Ser), one splicing mutation (IVS9-1 g-c) and two frameshift mutations, DelG576 and InsGG743. Although the molecular defect was homozygous in all patients but one, parents' consanguinity could be confirmed in only one case. InsGG743 was detected in two cases, and DelG576 was found in three patients originating from southern Italy, suggesting a possible geographical distribution of the genetic defect. Haematological data showed the presence of peripheral spherocytosis in all cases, although only one had a concomitant membrane defect. An increase in serum
ferritin
levels was observed in the splenectomized patients, suggesting that the iron status of these subjects should be monitored and that they should be investigated for potential additional risk factors for iron accumulation.
