CSAp is an antigen originally identified in the GW-39 human colonic carcinoma xenograft, and also found in gastric and colonic cancers, fetal colon, normal and inflammatory adult colon, and in some ovarian tumors. However, it appears to be increased primarily in inflammatory, benign , malignant, and fetal human intestine, gastric cancer, and ovarian tumors, as determined by an hemagglutination-inhibition assay. Gel immunodiffusion patterns show that CSAp is immunologically distinct from CEA,
NCA
, AFP, BOFA, and human liver
ferritin
. CSAp thus appears to be a putatively new fetal substance with a high degree of specificity for gastric, colonic, and ovarian tissues.

ZGM was purified from both primary and metastatic colonic carcinomas demonstrably positive for ZGM by immunofluorescence microscopy. ZGM purification included preparative Pevikon electrophoresis, Sepharose 4B molecular exclusion chromatography and Con A-Sepharose affinity chromatography. ZGM had an alpha2 electrophoretic mobility, an estimated molecular weight by Sepharose 4B equal to or greater than 2 x 10(6), and did not bind to Con A-Sepharose, although having determinants with CEA-like activity. Its immunologic activity was resistant to trypsin or phospholipase A but not to neuraminidase. Antisera prepared to ZGM and absorbed with saliva, when tested by double immunodiffusion, formed a single precipitation line with saline extracts of colon tumors and did not cross-react with CEA, AFP, normal tissue extracts,

Immunofluorescence microscopy confirmed the presence of ZGM in 40 out of 45 adenocarcinomas of the GI tract staining primarily in tumors, the apical cytoplasm, and in grossly nonmalignant tissues, the deep crypts of the villi, while all of 22 non-GI tumors in the study were ZGM negative.

In an attempt to identify new tumor markers in human colon carcinoma, we produced antisera in rabbits tolerant to normal human tissue antigens and immunized with zinc glycinate-treated extracts of liver metastases from a colon carcinoma. These antisera reacted with carcinoembryonic antigen and with an additional component present in the tumor extracts but not detected in the extracts of normal tissues. The new component, the zinc glycinate marker (ZGM), had an alpha2 mobility on immunoelectrophoresis, was soluble in 1 M perchloric acid, and had a molecular weight of approximately 2X10(6), as indicated by its elution pattern on Sepharose 6-B chromatography. It differed from alpha fetoprotein, nonspecific cross-reacting antigens (
NCA
, NGP, or CCA III),
ferritin
-like molecules, and blood group substances A, B, H, Lewis a, and Lewis b. The ZGM was similarly identified in saline or zinc glycinate extracts of 11 of 23 carcinomas of the colon. With routine hematoxylineosin staining, no histologic differences were apparent between tumors bearing the antigen and those without it.

levels in the aspirate of thyroid cyst (Six yellow clear, 4 yellow turbid and 10 chocolate colored turbid) without apparent sings of malignancy were found. The mean concentration in the 3,000 X g supernatant of the fluid was 40,116 ng/ml, and the 3,000 X g precipitate was 11,147 ng/ml. All cases showed normal levels of serum

was distributed from low to high. These ferritins showed a molecular weight of approximately 450,000 which was the same as found in human spleen and liver. The continuous increase in
ferritin
levels in thyroid cyst fluid was found by a chronological study in some cases. When tumor markers such as CEA,

, CA 19-9 and alpha 1-acid glycoprotein (alpha 1-AG, acute phase reactant) were examined simultaneously, an increase in some of the cyst fluid was observed. However, the incidence and the rate of increase of these tumor markers and acute phase reactant were low compared to
ferritin
. Neither a correlation between
ferritin
and CEA, nor between
ferritin
and CA 19-9 was found. The increase in

in thyroid cyst fluids may be due to the increased synthesis, release and storage by the inflammatory cells.

Eleven groups of workers submitted a total of 21 bronchial tumour-associated antigen preparations and 19 antisera for comparative studies. Many of the antisera proved to be polyspecific despite absorption procedures. Most of the antigen preparations contained some material reactive towards a reference antiserum to normal human serum proteins. While it appeared that no participants were studying identical antigen-antibody reactions, several cross-reactivities were identified in the antisera. When immune reactions to CEA, AFP,

, lactoferrin, human pepsin and gastricsin, and the pregnancy proteins, SP1 and SP3 were excluded by use of reference antisera and electroimmunoprecipitation methods, there remained 5 antigen-antibody reactions defining unique antigens.
