The effect of ruminal or abomasal starch infusion on milk yield and glucose metabolism of early lactation cows was measured. Four cows were continuously infused in the rumen or abomasum with partially hydrolyzed starch (1500 g/d) or were not infused (control) for three 14-d periods during wk 4 to 12 postpartum. Milk yield averaged over 40 kg/d throughout the experiment. Milk and milk
lactose
yields tended to increase when starch was infused and DMI was decreased, regardless of the site of infusion. Starch infusion increased mean insulin concentration and tended to decrease the concentration of serum nonesterified fatty acids. Ruminal starch infusion did not affect glucose irreversible loss rate but tended to increase
glucagon
concentration and decrease glucose oxidation. The increased milk yield that occurred when starch was infused ruminally relative to the milk yield of control cows could be a result of increased microbial protein supply or increased energy availability. Compared with ruminal starch infusion, abomasal starch infusion tended to increase the irreversible loss rate of glucose and to increase glucose oxidation. Abomasal infusion tended to increase plasma insulin concentration and to decrease the nonesterified fatty acid concentration relative to ruminal infusion. production, glucose oxidation, and the possible storage of glucose as body fat, which indicates that the early lactation dairy cow has a greater capacity for glucose metabolism than is provided by voluntary feed intake of average diets, but that not all available glucose is partitioned to the mammary gland. These data should be useful in testing current concepts and equations in nutritional and metabolic models of dairy cattle.

and SCFA infusions increased the number of TLESRs as well as the proportion of TLESRs associated with acid reflux episodes, but saline solutions did not. The postprandial fall of LESP was enhanced by previous SCFA infusion. Plasma PYY and OLI increased similarly after all colonic infusions. Colonic fermentation of

markedly affected LES function, and this effect was reproduced by SCFA infusion. Whether the mechanisms of this feedback phenomenon are of hormonal nature, neural nature, or both remains to be determined.

Veal calves fed by bucket often develop postprandial insulin resistance, hyperglycemia, and glucosuria during fattening. Automatic feeding systems allow feed intake for 24 h, and small ingested portions are expected to decrease postprandial glucose loads. We have studied metabolic and endocrine traits in calves that were either 1) fed identical daily amounts of whole milk plus milk replacer by a computer-programmed automatic feeder (> or =6 portions from 0800 to 2400 h) (GrA) or 2) fed by bucket at 0800 and 1630 h (GrB). Calves started at a body weight of 118 kg, and the experiment lasted for 3 wk. During wk 3,

was supplemented to stress postabsorptive glucose homeostasis. Feed intake and average daily gains in GrA and GrB were similar. Plasma concentrations during an 8-h period of glucose (in part), lactate, urea, and somatostatin (in wk 3), and of
glucagon
and insulin (wk 2 and 3) were smaller in GrA than in GrB, whereas growth hormone, insulin-like growth factor I, insulin-like growth factor binding protein-1 (wk 2), and prolactin concentrations (wk 2 and 3) were higher. Lactose supplementation in wk 3 enhanced transient postprandial hyperglycemia and hyperinsulinemia. Thus, there were marked metabolic and endocrine differences when calves sucked their feed in six or more portions during a 16-h period from an automatic feeder compared with twice daily drinking from a bucket. Ingestion of small portions by calves avoided marked hyperglycemia and lactate increments, and lower plasma urea concentrations mirrored enhanced nitrogen utilization, possibly mediated by the altered growth hormone, IGF-I and insulin status.

The effect of intestinal glucose supply on whole body rate of glucose appearance (WBGRa) and mammary utilization of glucose was studied in four lactating dairy cows. Glucose (0, 443, 963 and 2398 g/d) was continuously infused in the duodenum over 14-d periods using a Latin square design. A grass silage-based diet was formulated so that treatments were isoenergetic and isonitrogenous and contained 100 and 110% of energy and protein requirements according to INRA (1989). The WBGRa was measured by the [6,6-(2)H2]glucose dilution technique, and mammary glucose balance by arteriovenous differences and blood flow measurements. Duodenal glucose infusion increased arterial glucose concentrations linearly, whereas arterial concentrations of insulin, growth hormone, and
glucagon
were not changed. The WBGRa increased linearly with increasing glucose loads. The increase represented 42% of the intestinal glucose supplement. Mammary blood flow dramatically increased (up to 45%) and was associated with a significant increase of arterial insulin-like growth factor-1 concentrations. Mammary gland rate of glucose disappearance ([6,6-(2)H2]glucose measurement) increased linearly, whereas net mammary balance of glucose,
lactose
, and milk yields increased quadratically. Net mammary balance of glucose accounted for 60% of WBGRa, except for the greatest dose (47.6%). The decrease in milk yield with 2398 g/d of glucose may be explained by an imbalance in intracellular intermediate concentrations. The milk ratio of glucose-1-phosphate to glucose-6-phosphate decreased significantly at the greatest infusion of glucose. In conclusion, exogenous glucose supply to a grass silage-based diet increased WBGRa, mammary utilization of glucose and milk synthesis.

Longitudinal studies indicate that milk and fermented milk products lower basal plasma cholesterol concentrations, despite their high content of saturated fat, and therefore have favourable health effects. However, there have been few studies on the postprandial effects of milk products. The present study compared the effect of whole milk with a fermented milk, A-38, on postprandial carbohydrate and lipid metabolism, gastric emptying and appetite. Eight healthy young men participated. On the two test days, they arrived fasting for collection of baseline values before consuming the meals, which for a 75 kg subject consisted of 1.4 litre milk or fermented milk, plus 165 mg [13C]acetate (for later determination of gastric emptying by a [13C]acetate breath test). Lactose (15 g) was added to the A-38 meal to equalize the
lactose
content. Postprandially the A-38 meal resulted in a slower gastric emptying rate than milk (P<0.001). Furthermore, the A-38 meal resulted in a greater increase and a quicker decrease of the triacylglycerol content in all lipoprotein fractions (LDL-fraction, P<0.05; other fractions, P<0.001) and of the gastrointestinal hormones (cholecystokinin and peptide YY, P<0.05; gastric inhibitory polypeptide and
glucagon
-like polypeptide-1, P<0.001). There were no significant differences in appetite sensations (measured by visual analogue scale) or in the glucose and insulin response (P>0.10). The slower emptying rate of the liquid phase after the A-38 meal is probably due to the higher viscosity of A-38. The lower and more prolonged triacylglycerol response after the milk meal might be caused by coagulation of milk in the stomach.

Gluconeogenic capacity may be an important factor regulating dry matter intake (DMI) in lactating dairy cows. To determine whether increased glucose demand affects feed intake and hepatic gene expression, lactating Holstein cows were treated with phlorizin or vehicle (propylene glycol) for 7 d. Multiparous cows (n = 12; 269 +/- 65 d in milk, mean +/- SD) were randomly assigned to treatment sequence in a crossover design and were adapted to a common diet for 7 d before the beginning of the experiment. Phlorizin injected s.c. at 4 g/d caused glucose excretion in urine at the rate of 474 g/d. Although phlorizin decreased
lactose
synthesis and milk production (both P < 0.01), DMI and 3.5% fat-corrected milk production were not altered by treatment. A net deficit of 383 g glucose/d in milk and urine for phlorizin (relative to control) was likely replaced partially through increased gluconeogenesis. The molar insulin:

ratio was decreased 17% by phlorizin (P < 0.001) and hepatic phosphoenolpyruvate carboxykinase, glucose-6-phosphatase, and pyruvate carboxylase mRNA abundance increased (all P < 0.05). Late-lactation dairy cows adapted quickly to an increase in peripheral glucose demand; adaptation mechanisms likely included enhanced gluconeogenic capacity, whereas DMI was not altered.

-like peptide-2 (GLP-2) during chronic TPN improves intestinal growth and morphology, it is uncertain whether GLP-2 enhances absorptive function. We placed catheters in the carotid artery, jugular and portal veins, duodenum, and a portal vein flow probe in piglets before providing either enteral formula (ENT), TPN or a coinfusion of TPN plus GLP-2 for 6 days. On postoperative day 7, all piglets were fed enterally and digestive functions were evaluated in vivo using dual infusion of enteral ((13)C) and intravenous ((2)H) glucose, in vitro by measuring mucosal lactase activity and rates of apical glucose transport, and by assessing the abundances of sodium glucose transporter-1 (SGLT-1) and glucose transporter-2 (GLUT2). Both ENT and GLP-2 pigs had larger intestine weights, longer villi, and higher

The present study examined, at identical daily nutrient intakes, the impact of separating protein and
lactose
intakes across two daily meals on the metabolic and endocrine status in heavy veal calves. Calves were assigned to one of six degrees of separating protein and
lactose
over the two meals (termed nutrient synchrony, SYN 1-6; 6 calves/treatment). They were fed the protein-rich (P-)meal and the
lactose
-rich (L-)meal at 06:00 and 18:00h, respectively, or vice versa. At SYN 1, calves were fed with 50% of the daily protein and 50% of the daily
lactose
intake in each meal. Protein and
lactose
were iso-energetically exchanged between the two daily meals from SYN 1 to 6. At SYN 6, 85% of the daily protein and 20% of the daily
lactose
was fed in the P-meal and the remainder in the L-meal. Blood samples were collected hourly during 24h. Mean 24h glucose concentrations increased and insulin concentrations decreased from SYN 1 to 6. Postprandial 5h areas under concentration curves (AUC(0-5h)) of glucose increased with increasing meal

Feeding rumen-protected fat (RPF) is an alternative to increase energy density of the diet and therefore energy intake in dairy cows. To investigate metabolic and endocrine changes in dairy cows fed either a diet containing RPF (FD) or a control diet with an increased amount of cornstarch (SD), 3 Holstein cows (83 +/- 1 d in milk) were fitted with catheters in the portal vein, a mesenteric artery, and 2 mesenteric veins. Cows were fed consecutively SD and FD for 3 wk, respectively. In FD, cornstarch [92 g/kg of dry matter (DM)] was replaced by 50 g of RPF/kg of DM (mainly C16:0 and C18:1). Tracer infusions of NaH(13)CO3 and D-[U-(13)C6]glucose were performed into a jugular vein to measure rate of appearance and oxidation of glucose. Arterial and portal blood samples were collected to measure concentrations of glucose, lactate, volatile fatty acids, nonesterified fatty acids, beta-hydroxybutyrate, triglycerides, AA, insulin, and
glucagon
. Concomitantly, para-aminohippurate was infused into a mesenteric vein for measurement of portal plasma flow. Although DM intake was slightly lower in FD, protein and energy intakes were unaffected by diets. Milk and
lactose
yields were higher in FD than SD. Arterial plasma glucose concentration was lower with FD than SD, whereas nonesterified fatty acid and triglyceride concentrations were higher in FD.
