In teleosts, changes in swimming, exploring, general locomotor activity, and
anxious state
can be a response to stress mediated by the
corticotropin
-releasing hormone system activation and its effects on glucocorticoid levels. Zebrafish has been widely used to study neuropharmacology and has become a promising animal model to investigate neurobehavioral mechanisms of stress. In this report the animals were submitted to acute restraint stress for different time lengths (15, 60 and 90 min) for further evaluation of behavioral patterns, whole-body cortisol content, and
corticotropin
-releasing hormone expression. The results demonstrated an increase in the locomotor activity and an alteration in the swimming pattern during a 5-min trial after the acute restraint stress. Interestingly, all groups of fish tested in the novel tank test exhibited signs of anxiety as evaluated by the time spent in the bottom of the tank. Whole-body cortisol content showed a positive correlation with increased behavioral indices of locomotion in zebrafish whereas molecular analysis of
corticotropin
-releasing hormone showed a late reduction of mRNA expression (90 min). Altogether, we present a model of acute restraint stress in zebrafish, confirmed by elevated cortisol content, as a valid and reliable model to study the biochemical basis of stress behavior, which seems to be accompanied by a negative feedback of

Psychological stress plays an important role in the occurrence and development of temporomandibular joint disorder (TMD). The correlation between psychological factors and TMD has been clinically shown, but the influence of psychological stress on the temporomandibular joint (TMJ) structure still lacks direct evidence. Here, we used communication box to establish the rat model of psychological stress. The stress level of animals was estimated by the elevated plus maze (EPM) test and hormonal assays. The histomorphology and three-dimensional microstructure of the rat condyles were observed by hematoxylin-eosin (HE) staining and Micro-CT, respectively. Compared with control rats, the
anxious state
of the stressed rats was evidenced by higher plasma
adrenocorticotropic hormone (ACTH)
and corticosterone (CORT), as well as lower ratios of open arm entries and time and lower time spent in open arms after 1, 2, 3, 4 and 5 week(s) post-exposure to psychological stimuli. HE staining and histomorphometric data analysis showed decreased thicknesses of the central and posterior condylar cartilages in stressed rats at weeks 3, 4 and 5, with the most obvious changes in the posterior part characterized by debonding and thinned fibrous layer, thickened proliferative layer, thinned mature layer and hypertrophic layer. Moreover, Micro-CT scanning revealed local lesion of the subchondral bone in the posterior condylar cartilages of stressed rats at week 5. Our findings indicate that pathologic changes of the histomorphology and three-dimensional microstructure occur in the condyles of stressed rats, hinting us a potential link between psychological factors and the pathogenesis or progression of TMD.

Early life events have special importance in the development as postnatal environmental alterations may permanently affect the lifetime vulnerability to diseases. For the interpretation of the long-term consequences it is important to understand the immediate effects. As the role of vasopressin in hypothalamic-pituitary-adrenal axis regulation as well as in affective disorders seem to be important we addressed the question whether the congenital lack of vasopressin will modify the stress reactivity of the pups and will influence the later consequences of single 24h maternal deprivation (MD) on both stress-reactivity and stress-related behavioral changes. Vasopressin-producing (di/+) and deficient (di/di) Brattleboro rat were used. In 10-day-old pups MD induced a remarkable corticosterone rise in both genotypes without
adrenocorticotropin
(ACTH) increase in di/di rats. Studying the later consequences at around weaning (25-35-day-old rats) we found somatic and hormonal alterations (body weight reduction, dysregulation of the stress axis) which were not that obvious in di/di rats. The more
anxious state
of MD rats was not detectable in di/di rats both at weaning and in adulthood (7-12-week-old). The lack of vasopressin abolished all chronic stress and anxiety-like tendencies both at weaning and in adulthood probably as a consequence of reduced ACTH rise immediately after MD in pups. This finding suggests that postnatal stress-induced ACTH rise may have long-term developmental consequences.
