The tumor stem cell clonogenic assay was utilized to investigate the autocrine growth response of small cell lung cancer (SCLC) to bombesin (BN) and
beta-endorphin
(beta-E).
Mycoplasma
contamination was detected in the human SCLC cell line NCl-H345 by a nucleic acid hybridization assay which detects
mycoplasma
ribosomal RNA. Clonogenic assays of
mycoplasma
(+) cells were compared to assays of the same cell line following treatment for
mycoplasma
. Concentrations of beta-E ranging from 0.1nM to 25nM or BN (0.1nM-100nM) were added to cells, media and agarose and applied to prepared base layers. Following incubation for 12-14 days at 37 degrees C, the degree of clonal growth stimulation was determined by colony counts greater than or equal to 42 mu. The non-infected cell population grew in the presence of 25nM BN up to 69% over control growth. The infected cells, however, did not grow more than 27% above control. In the presence of 10nM beta-E, colony counts of non-infected cells exceeded the control values by up to 187% whereas the
mycoplasma
(+) colonies did not grow more than 20% over the control values. These results indicate a marked reduction in the response of SCLC cell lines to the peptides BN and beta-E when infected with
mycoplasma
. Since infecting
mycoplasma
typically adhere to cellular membranes, these adherent
mycoplasma
may interfere with membrane receptors or alter signal transduction, thus, inhibiting the development of the autocrine response.

Field studies have suggested that 'stressors', such as transportation and mixing, might interfere with the immune competence of pigs. Therefore, an experimental model was established to study the influence of elevated concentrations of circulating cortisol on the immune capacity in swine. Three experimental groups, with six pigs in each, were immunized twice, 4 weeks apart, with
Mycoplasma
hyopneumoniae antigen. Endogenous production of cortisol was induced by intramuscular injection of
adrenocorticotropic hormone (ACTH)
twice daily. One group received ACTH during the week before and after the second immunization, one group during the week after the second immunization only, while one group served as untreated controls. The treatment with ACTH induced high, but physiological, concentrations of cortisol in plasma. Simultaneously, the number of lymphocytes per milliliter blood decreased while the neutrophil number increased. The elevated concentrations of cortisol also coincided with reduced proliferation and interleukin-2 production by blood lymphocytes stimulated with the mitogens concanavalin A and phytohemagglutinin in vitro, while the responses to pokeweed mitogen were less affected. The suppression of mitogen responses was more pronounced in cultures of whole blood than in cultures of purified peripheral blood mononuclear cells (PBMC). Antibody production, induced by M. hyopneumoniae in cultures of purified PBMC was also inhibited by ACTH treatment. Both the rate of increase and the magnitude of the antibody production induced by the primary immunization were reduced. In contrast, no effects of ACTH treatment were recorded for the response to the second immunization or on the serum levels of antibodies to M. hyopneumoniae.(ABSTRACT TRUNCATED AT 250 WORDS)

Fibromyalgia and widespread pain were common in Gulf War veterans with unexplained illness referred to a rheumatology clinic. Increased tenderness was demonstrated in the postmenstrual phase of the cycle compared with the intermenstrual phase in normally cycling women but not in users of oral contraceptives. Patients with fibromyalgia had high levels of symptoms that have been used to define silicone implant-associated syndrome. Tender points were found to be a common transient finding associated with acute infectious mononucleosis, but fibromyalgia was an unusual long-term outcome. The common association of fibromyalgia with other rheumatic and systemic illnesses was further explored. A preliminary study revealed a possible linkage of fibromyalgia to the HLA region. Patients with fibromyalgia were found to have an impaired ability to activate the hypothalamic pituitary portion of the hypothalamic pituitary adrenal axis as well as the sympathoadrenal system, leading to reduced
corticotropin
and epinephrine response to hypoglycemia. Much interest has been expressed in the literature on the possible role of autonomic dysfunction in the development or exacerbation of fatigue and other symptoms in chronic fatigue syndrome.

The effectiveness of cognitive behavioral therapy and behavior therapy for chronic pain in adults was emphasized. A favorable outcome of fibromyalgia and chronic fatigue syndrome in children and adolescents was reported.
