The aim of the present study was to compare effects of intravenous infusion of
vasopressin
AVP and V1 receptors blockade on blood pressure and heart rate in normotensive (WKY) and spontaneously hypertensive (SHR) rats. A 20 min
vasopressin
infusion (1.2 ng/kg/min) elicited significantly greater increase in mean blood pressure (MP) in SHR than in WKY. Heart rate was significantly reduced in SHR while nonsignificantly in WKY. A 20 min dEt2 AVP (V1 antagonist) infusion (0.5 microgram/kg/min) elicited significant decrease in MP and increase in heart rate (HR) in SHR, but produced no effect in WKY. The data indicate that SHR are more susceptible to pressor and hypotensive effects of sustained elevation of AVP and AVP antagonist. The results support the hypothesis that AVP may contribute to pathogenesis of hypertension.

In 12 patients with acute non-inflammatory renal failure (ANRF) and in 21 healthy persons the relationship was studied between the activity of the renin-angiotensin-aldosterone (RAA) system and
vasopressin
secretion. The study was carried out during the so called bed rest test and during water immersion (WI) after captopril-induced blockade of the angiotensin-converting enzyme. The results of these experiments suggest the absence of a close functional relationship between the activity of the RAA system and
vasopressin
secretion in patients with ANRF and in healthy controls.

Influence of blockade converting enzyme on plasma renin activity (PRA), aldosterone and
vasopressin
secretion in 12 hemodialyzed patients with chronic renal failure and in 21 healthy subjects was observed. Our observation were provided during bed rest and water immersion tests. We didn't observe statistically significant influence PRA (increase after converting enzyme blockade) on
vasopressin
secretion in patients with chronic renal failure and in healthy subjects. Correlation between PRA and
vasopressin
secretion was absent.

Recently, it was shown that in LLC-PK1 kidney epithelial cells hormones such as
vasopressin
or oxytocin increase cyclic GMP in a receptor-mediated and L-arginine-dependent manner. In the present study, the possible existence of cross-tolerance to
vasopressin
and oxytocin was investigated in nitrate-tolerant LLC-PK1 cells. Pretreatment with 1 mM glyceryl trinitrate for 3 h decreased cyclic GMP stimulation by 1 microM
vasopressin
and 1 microM oxytocin by 49% and 54%, respectively. Under the same conditions, cyclic GMP stimulation at 1 microM sodium nitroprusside was diminished by 56% whereas the cyclic GMP response to 100 microM glyceryl trinitrate was virtually abolished. Our results demonstrate that a substantial degree of cross-tolerance to L-arginine-dependent guanylate cyclase activators occurs in nitrate-pretreated nonvascular cells which may be due to glyceryl trinitrate-induced desensitization of soluble guanylate cyclase.

The hypothalamo-
neurohypophyseal
neurosecretory system was investigated in 8-, 15- and 30-day-old rats subjected to three intragastric doses of CCNU - 12.5 mg/kg b. wt. each on the 3rd, 5th and 7th day after birth. Neurosecretory neurons in the hypothalamus were visualized in the paraffin sections by the immunoenzyme (PAP) technique using antibodies against neurophysin and by Gomori chrome-hematoxylin staining. Accumulation of neurophysin was observed in these cells after treatment with CCNU. Karyometric measurements showed an increase of the mean nuclear cross-section area in PVN neurons in 8-day-old rats exposed to CCNU. In four experimental rats disseminated intracerebral hemorrhagic foci were present. Plasma osmolality was far below the normal values on the 8th day, on the 15th day of life it shifted to hyperosmolality and returned to normal at the age of 30 days. Discussion of the results leads to the conclusion that the increase of the neurosecretory function observed in this experiment was secondary to vasogenic changes.

It is pointed out in this article that efficient cognitive performance depends on two independent parameters, one concerned with the intrinsic capacity of the nervous system, the other with motivational processes such as arousal. It is argued that a substance should be regarded as a true cognition enhancer only if it improves the former. There is evidence suggesting that some neuropeptides--for example
vasopressin
--act on motivational and arousal processes, and a model which offers a plausible mechanism is discussed. The need for techniques and designs which permit us to separate these different effects is highlighted.

Water immersion (WI) induced alterations of plasma
vasopressin
(AVP), plasma osmolality, plasma volume (PV) and plasma sodium and potassium concentration were examined in 12 patients with noninflammatory acute renal failure (NARF) at the anuric/oliguric phase, 20 patients with chronic renal failure (CRF) and 15 healthy persons. In all examined groups a significant increase of PV and decrease of plasma osmolality and AVP after WI was observed. In patients with NARF the increase of PV was significantly greater than in CRF and in healthy persons. In patients with NARF a significantly greater decrease of plasma sodium concentration after WI was also noted than in patients with CRF and in healthy persons. Only in healthy persons, but not in patients with acute or chronic renal failure a significant correlation between PV or decrease of plasma osmolality and WI induced decrease of plasma AVP level was stated. We conclude that patients with NARF and CRF are characterized by an inappropriate volumetric and osmotic regulation of AVP secretion.

Rats euhydrated or dehydrated for four days were given intracerebroventricular insulin once daily in a dose of 100 ng (not affecting blood sugar level). In euhydrated rats, insulin decreased significantly the neurohypophysial
vasopressin
content. In dehydrated animals the neurohypophysial content depleted by deprivation of water could be further reduced by intracerebroventricular treatment with insulin. These results may suggest a possible regulatory role of brain insulin in the mechanisms of
vasopressin
release.

Dekanski's method was used to estimate the pressor activity of the extracts of the posterior pituitary lobe in anaesthetized rats, after the infusion of 0.9% NaCl solution above the supraoptic nuclei and haemorrhage in the amount of 1.5% of body weight or after the infusion of Substance P solution above the supraoptic nuclei and haemorrhage. It has been found that the
vasopressin
content in the posterior pituitary lobe decreased about 20% after the infusion of 0.9% NaCl solution and haemorrhage. The infusion of Substance P above the supraoptic nuclei inhibits the loss of
vasopressin
from the pituitary caused by haemorrhage.
