The influence of Ca2+ and the possible action of hormone blockers on the activation of glycogenolysis by methotrexate were investigated.
Methotrexate
was inactive on glycogenolysis and oxygen uptake when the liver, depleted of intracellular Ca2+, was perfused with Ca(2+)-free medium. The action of methotrexate in calcium-depleted hepatocytes could be restored by the addition of extracellular Ca2+. When Ca2+ was absent in the extracellular medium, but the intracellular stores were not depleted, methotrexate produced transient and progressively attenuated increases in glycogenolysis and oxygen uptake. Like many agonists, methotrexate produced transient increases in Ca2+ efflux. The action of methotrexate was not blocked by the antagonists of norepinephrine, phenylephrine, isoproterenol,
vasopressin
and angiotensin II. It was concluded that methotrexate acts through a Ca(2+)-dependent mechanism, which is similar to that of the Ca(2+)-dependent agonists. This action, however, seems not to be receptor mediated.

We report a case of AML with diabetes insipidus (DI). A 68-year-old female was admitted to our hospital because of fever and leukocytosis. The WBC was 197,000/microliter with 98% blasts positive for myeloperoxidase, CD33, CD34 and HLA-DR. While, on admission, urine volume was more than 6 liters daily. Blood

level was 0.3 microgram/ml. The patient was diagnosed as having AML with DI. By chemotherapy consisting of BHAC, DNR, 6-MP and PSL and intrathecal administration of AraC,
MTX
and PSL, and nasal drip of DDAVP, complete remission was attained and the urine volume was reduced to normal. Finally DDAVP became unnecessary. Although the exact cause of DI cannot be ascertained, rapid increase of leukemic blasts and leukostasis in small vessels might be associated with hypothalamus-pituitary system damage. Reportedly, DI is a rare complication of leukemia and administration of DDAVP could be halted in only two patients with leukemia and DI.

Frogs ingest little water orally, but absorb the majority of the water needed for normal physiological performance through a specific region of the ventral skin, the pelvic patch. We observed non-stimulated (basal water absorption, BWA) water flux through the isolated pelvic patch in vitro in Japanese tree frog (Hyla japonica). We found that water flux through non-stimulated skin from the pelvic patch was greater in males than females. This water flux was confirmed as BWA by observing no effect following the in vitro administration of propranolol and [adamantaneacetyl(1), O-Et-D-Tyr(2), Val(1), aminobutyryl(6), Arg(8, 9)]

The gonad appears essential for observed sexual dimorphism in BWA, and we hypothesize that sex hormones regulate the release of PRL, that in turn influences BWA indirectly. E2 is known to exert a specific stimulatory effect on PRL secretion. In addition, we have observed that some endocrine disrupting contaminants also eliminate the sexual dimorphism in BWA observed in the Japanese tree frog.
