Changes in rheological properties of the blood were produced by intravenous injection of a high-molecular weight dextran and
lysin
-
vasopressin
. The animals were decapitated in one hour. Oxygen absorption by mitochondria of the heart in oxidation of 2.5-10 mM of the succinate increased by 90-120%, as compared to control. Stimulation of respiration by ADP was decreased 1.5-2 times. Simultaneous administration of the succinate and glutamic acid normalized the respiration and phosphorylation. A possibility of inhibition of succinic-dehydrogenase by the oxalo-acetic acid was suggested. Switching of respiration to succinic acid and limiting of the SDG activity can be considered as adaptive factors under conditions of changes in rheological properties of the blood, and are directed to the maintenance of cardiac activity, this being evidenced by the absence of changes in the ATP-asic activity and in the myosin content of the heart.

The present study shows that in a group of 6 euadrenal patients, previously treated by complete adrenalectomy for pituitary dependent Cushing's syndrome, the stress stimulus of insulin induced hypoglycaemia is followed by a plasma ACTH response which is of similar magnitude as the response obtained with
lysin
-
vasopressin
. Both observations indicate that the central nervous system-pituitary axis is basically normal in pituitary dependent Cushing's syndrome as assessed by insulin induced hypoglycaemia. It is concluded that non-responsiveness of the pituitary-adrenocortical system to insulin induced hypoglycaemia in untreated patients with pituitary dependent Cushing's syndrome does not represent a fundamental defect of the stress mechanism, but is due to hypercorticism per se.

The aim of this study was to investigate the qualitative and quantitative changes of ACTH-cells in the rat after application of a specific and a non-specific stimulus. A CRF-analog (
lysin
-
vasopressin
) and a prostaglandin (prostaglandin E1) were used. 40 rats were injected
lysin
-
vasopressin
or prostaglandin E1, respectively, for 4 weeks. The pituitary glands were investigated by means of light microscopy, electron microscopy and morphometry. Activation of the ACTH-cells could be observed after use of both substances, the effect of
lysin
-
vasopressin
being more intense than that of prostaglandin E1. Enlargement of the nucleus, the cytoplasm and the organelles involved in hormone-production and -transport were found and verified by morphometry. Additionally an increase in number of the cells could be demonstrated. Prostaglandin influenced not only ACTH-cells, but also other cells of the anterior pituitary.

8 children with precocious puberty were treated with cyproterone acetate (CPA). During treatment there were no definite clinical signs of depressed adrenocortical function. The plasma cortisol concentrations were grossly depressed and the diurnal cortisol rhythm was abolished. Two months after discontinuation of CPA treatment the adrenocortical function had greatly improved. The
lysin
-
vasopressin
stimulation test revealed in one child a normal, in another child an exaggerated ACTH response during CPA therapy. Fasting plasma ACTH concentrations were elevated compared with normal controls, but they were very low compared with patients with Addison's disease. The results suggest that CPA has a twofold effect leading to adrenocortical insufficiency: i.e., inhibition of cortisol secretion by the adrenals themselves and inhibition of ACTH secretion at the hypothalamopitiuitary level.

Pre- and postoperative evaluation of hypothalamic-pituitary function was performed in six children, aged 5.5 to 13.3 years with craniopharyngiomas. Before surgery growth hormone deficiency (GHD) was documented in four, hypothalamic hypothyroidism in three, and secondary ACTH-deficiency and hyperprolactinaemia in one patient. Diabetes insipidus was absent in all patients. After neurosurgical treatment GHD was present in all, hypothyroidism in five, ACTH-deficiency in three, hyperprolactinaemia in three, and diabetes insipidus in four children. The study shows that all endocrine functions tested may be defective even before surgery, although diabetes insipidus seems to be a rare preoperative complaint. Surgical intervention, however, often leads to additional endocrine disorders. From the data presented here one may suggest that TRH stimulation tests, evaluation of serum prolactin, and
lysin
-
vasopressin
stimulation tests are the most useful investigations to distinguish between hypothalamic and primary pituitary disorders.

The now nine years old girl with growth retardation, started to be ill with otitis and then diabetes insipidus of central origin at 1974. A treatment with
lysin
-
vasopressin
is prescribed. The PNEG in May 1976 shows a little, pea like, suspicious, supra-sellar nodule who is not surgically explored since they was no ophtalmologic symptoms and because a normal CT scan. Corticoid and thyroid substitutive therapy is added until September 1977 when the general status becomes impaired and vomiting starts. Also because some visual loss, a new neuroradiological study is performed showing a supra-sellar tumour and a fourth ventricle mass. The CT scan asserts the double intracranial expansive process and a posterior fossa craniotomy is done with subtotal resection of a vermian tumour and Torkildsen drainage. The histology is : Immature Dysembryoma (seminoma type) or germinoma. The follow-up was good under hormonal care. X Rays Therapy over the posterior fossa, the suprasellar region, the brain and the spinal channel was instaured. Four months later, the CT scan shows normal sized ventricles and no tumour mass at all. This case gives the authors opportunity for comments and to study the concerned literature.

In two nonpregnant women with cranial diabetes insipidus, myometrial activity under different hormonal conditions was studied by intrauterine pressure recording. Recordings were performed when the women were under the influence of their regular treatment with desaminocys1-D-arg8-
vasopressin
(DDAVP), and when they had fully developed symptoms of their disease. Uterine activity was similar to that found in normal women under the same hormonal conditions, and generally did not change when symptoms of lack of
vasopressin
appeared. Both DDAVP (15-20 microgram), given intranasally, and
lysin
vasopressin
(0.2 IU), given intravenously, stimulated uterine activity, particularly in late secretory menstrual phase. It is suggested that endogenous
vasopressin
is of minor importance for the induction of spontaneous uterine activity in nonpregnant women.

The effects of
vasopressin
and of its analogues on liver function, and their possible mechanisms of action, are poorly understood. This study was designed to assess the effect of triglycyl-
lysin
-
vasopressin
on liver function, evaluated by two quantitative tests independent of liver blood flow, i.e., indocyanine green intrinsic hepatic clearance and galactose elimination capacity. Indocyanine green intrinsic hepatic clearance and galactose elimination capacity were determined before and after administration of 2 mg triglycyl-
lysin
-
vasopressin
to (respectively) 10 and 12 patients with cirrhosis. Eighteen additional patients with cirrhosis were studied before and after infusion of placebo. No significant variation in either test was observed in placebo-treated patients. A significant decrease in indocyanine green intrinsic hepatic clearance, averaging 22%, was observed in patients receiving the drug (p = 0.04). Conversely, galactose elimination capacity remained unchanged after the drug. These results are compatible with the hypothesis that the drug produced a preferential decrease in perfusion in functioning areas of the liver, with relative maintenance of blood flow in non-functioning areas.

The effect of
arginine-vasopressin
(
AVP
) and its analogues upon the endocochlear potential (EP) was examined in the guinea pig.
AVP
decreased the EP reversibly and dose-dependently like
lysin
-
vasopressin
(LVP). V1 antagonist failed to block the effect of
AVP
upon the EP. V2 agonist reversibly decreased the EP like
AVP
. The results suggest that V2 receptors mediate the action of
vasopressin
on the EP.

Plasma concentrations of adrenocorticotrophin (ACTH) and cortisol were measured during insulin-induced hypoglycemia and
lysin
-8-
vasopressin
(LVP) test in 60 healthy subjects, non-smokers and habitually smokers of 10 or more cigarettes/24 hours. A marked and statistically significant rise of both hormones was found in non-smoker subjects, whereas smokers showed poor and not significant modifications. These results suggest that continuous chronic inhalation of nicotine may act as a powerful stimulus on the hypothalamo-hypophyseal structures that control the hypothalamic CRF and/or ACTH production and release. Central nervous mechanisms of hormonal regulation may become less sensitive and efficient when an acute rise of ACTH is required, as during stimulating tests. Our investigation confirms that cigarette smoking is heavily responsible of endocrine disorders, in particular of hypophyseal-adrenal axis.
