The role of lysine-vasopressin and oxytocin, as well as of their analog and fragment (DGAVP, desglycinamide arginine vasopressin; PLG, prolyl-leucyl-glycyl-amine), microinjected into the ventral hippocampus, in the formation and extinction of active avoidance conditioned reflex, was investigated. It was established that the introduction of lysine-vasopressin, as well as of its analog, into the hippocampus promoted the improvement of the formation of the avoidance reaction, while an inhibitory effect was elicited in the period of extinction of the skill. The opposite effects on the active avoidance reaction were obtained with oxytocin and its fragments. It is hypothesized that the observed behavioral changes in the active avoidance reaction are associated with the influence of these peptides on the processes of learning and memory.

The influence of intraperitoneal injection of vasopressin (LVP), oxytocin (OXY) and their fragments (DGAVP, PLG) on the acquisition and extinction of conditioned food reflex was studied in rats. It was found that vasopressin and its fragments had a more pronounced specific effect on the higher nervous activity of the animals. This effect consisted in impairment of the performance of the conditioned food reflex while oxytocin had a tendency to improve its performance. On the ground of the obtained data it is suggested that administration of vasopressin may facilitate the memory function only under specific environmental conditions.

Earlier it was found that oxytocin (OXT) treatment inhibited the development of tolerance to ethanol. In the present study the possibility was investigated whether the effect of OXT on ethanol tolerance was related to peptide fragments derived from the C-terminal part of the molecule. The actions of different doses of the C-terminal tripeptide (prolyl-leucyl-glycinamide, PLG) and of a synthetic dipeptide derivative (Z-prolyl-D-leucine, Z-Pro-D-Leu) on the development of tolerance to the hypothermic effect of ethanol in CFLP mice were therefore investigated. Peptide treatment did not affect body temperature in ethanol-naive animals. The acute effects (hypothermia, sleeping time) of a single ethanol injection were also unaffected by these peptides. In contrast, both PLG and Z-Pro-D-Leu inhibited the development of tolerance to the hypothermic effect of ethanol. Accordingly, it might be speculated that a sequence active in affecting ethanol tolerance is located in the C-terminal part of the OXT molecule.

Maintenance of intravenous heroin self-administration and the degree of tolerance to the analgesic effect of self-injected heroin were simultaneously measured in heroin-tolerant rats. Subcutaneous injection of oxytocin (OXT-(1-9)) and of its behaviorally active fragments desglycinamide9-oxytocin (OXT-(1-8)) and [pGlu4,Cyt6]-oxytocin-(4-8) (OXT-(4-8)) decreased the amount of heroin self-injected. The C-terminal tripeptide of oxytocin (prolyl-leucyl-glycinamide, PLG, OXT-(7-9)) and desglycinamide9-[Arg]8-vasopressin (AVP-(1-8] were ineffective in this respect. In spite of the lower amount of self-injected heroin after pretreatment with oxytocin fragments, no differences in the antinociceptive effect of self-injected heroin, as assessed by the lick response using a hot plate device, were observed after pretreatment with placebo and oxytocin fragments. These findings suggest that oxytocin and some of its behaviorally active fragments attenuate heroin tolerance and that this effect may result in a diminished heroin intake in tolerant animals self-injecting heroin.

The comparative effects of thyroliberin (TRH), pentagastrin, C-terminal tripeptides of gastrin (MNF), luliberin (RPG) and its analog (RPF), oxytocin (PLG) and N-terminal tripeptide of angiotensin (NRV) on the glucose concentration in mouse blood, as well as the interrelationship between the peptides under study and insulin and somatostatin, were studied. It was found that all the peptides (except PLG) at doses 5, 50 and 100 mg/kg (intraperitoneally) exert a hypoglycemic effect comparable to that of insulin at the dose of 0,15 MU/kg (subcutaneously). The dose-dependent effect is observed only in the case of TRH. Somatostatin reverses the hypoglycemic action of TRH, MNF and NRV. TRH and RPF decrease the glucose-induced rise in insulin concentration measured by RIA. The structure-activity relationship and possibility of the formation of endogenous peptide hormone fragments as products of hormone molecules enzymatic cleavage and their glucoregulatory role are discussed.

Using YPLG (Tyr-Pro-Leu-Gly), a tetrapeptide, as the template, an imprinted monolithic column was prepared and applied to the selective recognition of oxytocin based on the epitope approach and capillary electrochromatography (CEC). By optimizing the polymerization solution in terms of functional monomer, cross-linking reagent, porogen, and imprinted template via CEC evaluations of synthesized columns, an imprinted monolith with good recognition capacity (the imprinting factors for YPLG and oxytocin were 4.499 and 4.013, respectively) and high column efficiency (theoretical plates for YPLG and oxytocin were 22,995 plates/m and 16,952 plates/m, respectively) was achieved. In addition, the effects of various experimental parameters on the recognition of oxytocin, including the organic modifier content, the buffer concentration, and the pH value, were studied systematically. Furthermore, a mixture of oxytocin and other proteins was analyzed using this monolithic CEC column, and oxytocin was eluted much more slowly than other large biomolecules, which demonstrated the high selective recognition ability of such an imprinted monolith for oxytocin with PLG (Pro-Leu-Gly) as the epitope.
