The effect of increased concentration of vasopressin and
oxytocin
in the cerebrospinal fluid on the excitability of the hypoglossal nerve nucleus was investigated. The experiments were carried out on rats under chloralose anaesthesia. Retractory jerks of the outstretched tongue were evoked by supra- or infraorbital nerve stimulation during perfusion of the cerebral ventricles with McIlwain-Rodnight solution. The solution contained synthetic arginine vasopressin 0.05 U/ml or synthetic
oxytocin
0.05 U/ml. Perfusion of the ventricles with vasopressin increased and perfusion with
oxytocin
decreased the evoked tongue jerks.

1. Two enzymes acting on the linear portion of
oxytocin
: carboxamidopeptidase (releasing Gly . NH2) and prolyl peptidase (releasing Leu-Gly . NH2) were identified in the cytoplasmic fraction of chicken liver. 2. Carboxamidopeptidase was purified 134-fold with a 23% yield, and prolyl peptiase 71-fold with a 20% yield. The specific activity of the final preparations was 181 and 96 microU/mg protein, respectively. 3. The optimum pH for carboxamidopeptidase was 6.0--6.5 and for prolyl peptidase, 7.5. Carboxamidopeptidase activity was inhibited by Mn2+, Zn2+, Ca2+, Co2+, and stimulated by EDTA; the activity of prolyl peptidase was inhibited by Zn2+ and Mn2+. The Km value of both enzymes for
oxytocin
was 1.5--2.4 microM.

Release of

Under general urethane and chloralose anaesthesia the ventral surface of the hypothalamus and the pituitary gland were exposed by a transpharyngeal approach and the anterior pituitary lobe was entirely removed. The posterior lobe with the remaining neural and partially vascular connections with the hypothalamus was incubated in situ.
Oxytocin
and Substance P-like peptides were assayed biologically in six 20-min samples of the fluid outflow from the posterior pituitary lobe incubated in situ. At the beginning of collection of the fourth sample a hypertonic solution was infused into the third ventricle through a micropipette. This infusion increased significantly the release of
oxytocin
and insignificantly that of Substance P-like pepides from the posterior pituitary lobe into the incubation fluid.

The purpose of this work was to compare the oxytocic activity of plasma in oestrous cows during milking and for the first and second days following oestrous. Oxytocic activity of blood plasma was measured by the in vitro rat mammary cube assay of van Dongen and Hays. During oestrous, signigicantly lower amounts of
oxytocin
are released into the bloodstream of cows following milking as compared with the control values.

., 1977, 28 (6): 497-504. Rats received one infusion of 200 microgram 6-hydroxydopamine with 25 microgram of ascorbic acid into the lateral cerebral ventricle. After 57 days some rats were deprived of water for 4, 8 or 12 days. Then, the animals were sacrificed by decapitation.

was determined in extracts from the posterior pituitary lobe and hypothalamus by the method of Van Dongen and Hays, while the vasopressin content was determined by the method of Dekanski. It was found that 6-hydroxydopamine injection into the cerebral ventricles causes a rise in

Using the method of incubation in situ of the posterior pituitary lobe and washing the hypothalamic end of the divided pituitary stalk the activity of neurosecretory neurons was investigated directly. The oxytocic activity of the fluid incubating in situ the posterior pituitary lobe or washing the hypothalamic end of the cut pituitary stalk was determined on the rat myometrium. The oxytocic activity disappeared from this fluid after inactivation with sodium thioglycolate. The samples of the fluid incubating the posterior pituitary lobe obtained at 30 min intervals showed alternatingly increased or decreased oxytocic activity. In the fluid washing the cut pituitary stalk the activity was highest from 31 to 60 minutes after division of the stalk.

activity of the hypothalamus and neurohypophysis; carbachol did not influence the
oxytocin
potency of the hypothalamus but augmented it in the neurohypophysis. During long-term dehydration both carbachol and atropine intensified the depletion of

The study group consisted of 82 primigravid and 55 multiparous women with post term pregnancy, preeclampsia, intrauterine growth retardation, insufficiency of placenta and diabetes mellitus have induced labor. Prepidil (Upjohn) in dosage 0.5 mg was given into uterine cervix of 46 patients (PG group) and
oxytocin
was infused to 42 patients in dosage ranged from 5 mU/min to 30 mU/min (Ox group). Induction of labor has been considered as successful, if after 12 hours of drug administration, regular contractions of uterus and dilation of cervix more than 3 cm were obtained. Significant improvement of cervix state, measured by Bishop score has been observed only in PG group, even if the induction of labor failed. Similar rates of caesarean sections and the same occurrences of late and variable decelerations have been observed in both study groups. Results obtained in both these groups suggest that induction of labor in such pregnancies after prostaglandins administration is more effective than
oxytocin
infusion.
