This study was designed to understand the uterine activity (UA) in normal labor. 43 nulliparous women(33 in normal course of labor and 10 in abnormal course of labor) were in the active phase of spontaneous labor with cephalic presentation for vaginal delivery. An open-end fluid filled intrauterine catheter connected to a pressure transducer was used to measure the uterine contraction intensity, frequency and duration. UA was quantified by means of Montevideo Unit (MU) and Uterine activity integral(UAI). The receiver operator characteristic(ROC) curve defined the cut-off points of UA between normal labor and abnormal labor. The results showed that MU and UAI were correlated with the rate of cervial dilatation in the active phase of labor (r = 0.3734 and 0.3502 respectively, P < 0.05). There was a strong correlation between MU and UAI (r = 0.7173, P < 0.001). The mean of UA in normal labor as MU = 226(s = 68.45) mmHg/15 min. UAI = 901(s = 258.02) kPa/15 min. The minimum of UA to normal labor in MU was 170 mmHg/15 min, of UAI 650 kPa/15 min. The study suggests that quantification of UA is very useful in the management of labor, especially in the proper use of oxytocin in cases abnormal labor.

The properties of recombinant Aeromonas punctata prolyl endopeptidase(apPEP) were studied using specific substrate and peptides. Results show that the optimum catalytic temperature and pH was 34 degrees and 8.4, the stability of the apPEP was in the range of 4-32 degrees and pH 6.0-10.0, and its K(m) was 0.03 mmol/L based on the Z-Gly-Pro-betaNA. The apPEP was not sensitive to PMSF, TLCK, TPCK, Trypsion inhibitor, EDTA, tetrathionate and some metal ions, but was sensitive to SDS and Zn(2 ), and was completely inhibited by DFP. Oxytocin and calcitonin could be specifically hydrolyzed by apPEP at the carboxyl site of proline residue, but the hydrolysis efficiency of calcitonin by the enzyme was less than for oxytocin and for Z-Gly-Pro-betaNA.

Reverse transcription(RT)-PCR method was used to examine the cholinergic neuron derivative factor (CNDF)mRNA level changes in rat brain after the administration of ZNC(C)PR or its analogs. Glyceraldehyde-3-phophate dehydrogenase (GAPDH)mRNA was co-amplified as an internal control. The CNDF mRNA expression was significantly increased and its peak was reached at 18 h after the administration of ZNC (C)PR (hippocampus, 3.02 fold cortex, 5.33 fold compared to control, P < 0.01). NLPR, agonist of ZNC(C)PR receptor, enhanced the CNDF mRNA transcription to a higher level than ZNC(C)PR. ZDC(C)PR, antagonist of ZNC(C)PR receptor, partially blocked the CNDF transcription when co-administered with ZNC(C)PR. AVP increased the CNDF mRNA expression to lower level than ZNC(C)PR, while oxytocin had no effect on the CNDF transcription. The results suggest that the CNDF gene may be one of the targets of ZNC(C)PR, and its enhancement effect on CNDF mRNA expression was mediated by the ZNC(C)PR receptor.
