In rat hypothalamus intraventricularly injected with colchicine, the same neurons of the ventral region are stained with I.S. against alpha and beta-endorphin, (1-24) and (17-39) ACTH, alpha and beta-MSH, and beta-LPH. They are distinct from those producing LH-RH, somatostatin, neurophysin, and dopamine. These results suggest that the same neurons elaborate peptides identical with or immunologically related to endorphins, ACTH, alpha-MSH and beta-LPH, probably issued from a common precursor.

In rat brains intraventricularly injected with colchicine, the same discrete neurons of the arcuate and ventromedial nuclei can be stained with antisera against alpha- and beta-endorphins, (1-24)ACTH, (17-39)ACTH, alpha- and beta-MSH, and beta-LPH, as demonstrated by comparative studies in consecutive serial sections. These neurons are strongly reactive with anti-(17-39)ACTH, anti-beta-endorphin, anti-alpha-MSH and anti-beta-MSH, and more faintly stained with anti-alpha-endorphin, anti-beta-LPH and anti-(1-24)ACTH. Exceptionally, neurons reactive with anti-(17-39)ACTH and anti-beta-endorphin are poorly stained or completely negative with anti-alpha-MSH and anti-beta-MSH. Immunoreactive fibers end in the lateral median eminence and in the arcuate nucleus proper, or form ascending pathways along the third ventricle. Comparative studies with other antisera or with the Falck and Hillarp technique show that these neurons differ from the elements producing LH-RH, somatostatin, neurophysin, oxytocin, vasopressin and dopamine. These results suggest that the same neurons of the rat hypothalamus synthesize several neuropeptides identical with or immunologically related to endorphins, ACTH, alpha-MSH and beta-LPH, probably arising from a common precursor molecule similar to that found in the corticotropic cells of the pituitary. These neuropeptides of a common cellular and molecular origin might be involved in basic processes of the central nervous system as neurotramsmitters or neuromodulators.

Injection of posterior pituitary powder induces an intense mitotic stimulation in the zona glomerulosa of the adrenal gland of young rats. This effect is much more pronounced in females than in males. It is maximal at two days treatment. Longer periods result in a hypertrophied zona glomerulosa and lower mitotic activity. A search for the hormone responsible for the stimulation shows that vasopressin, and to a lesser extent oxytocin, are mitogenic. ACTH, alpha-MSH, beta-MSH and the pineal hormones have no effect. Renin (but not angiotensin) induces a significant stimulation. It is concluded that vasopressin exerts a potent influence on the glomerulosa. This is in contrast with the prevalent view that the glomerulosa is little affected by the hypophysis.

Previous work indicated that brain contains 3 types of lipolytic-melanotropic peptide: (1) in adenohypophysis: ACTH, alpha-MSH, beta-MSH, peptide I, peptide L', beta-lipotropin and gamma-lipotropin; (2) in neurohypophysis: peptide 7D6, also termed neurophysin I, peptide II or Wuu-Saffran peptide; (3) in extrahypophyseal regions: peptide IIF. Bovine and human neurophysin I prepared by R. Walter has now been found devoid of lipolytic and melanotropic activities. Porcine and bovine peptide 7D6, closely similar or identical to bovine neurophysin I in electrophoretic mobility and amino acid composition, were therefore reexamined to determine whether their lipolytic-melanotropic property resided in a contaminating factor. When peptide 7D6 was analyzed in 100 transfer counter current distribution (1 butanol/0.1M NH4 HCO3), the neurophysin was recovered in tubes 1-9 (7D6-alpha) representing 95% of 7D6. 7D6-alpha was inactive in lipolytic and melanotropic assays. The biologic activities of 7D6 were recovered instead in tubes 50-70 (labeled 7D6-beta), representing 5% of 7D6. 7D6-beta proved to be a peptide with MW 1000-3000, closely similar to peptide IIF in amino acid composition, MW, and Rf values in 4 systems of paper chromatography.

The neurotransmitter histamine participates in the neuroendocrine regulation of pituitary hormone secretion by an indirect action at a hypothalamic level where histaminergic neurons are abundant. The effect of histamine is caused by activation of postsynaptic H1- or H2-receptors. Histamine stimulates the secretion of ACTH, beta-endorphin (mediated by CRH and AVP), alpha-MSH (mediated by dopamine and peripheral catecholamines), and PRL (mediated by dopamine, serotonin and AVP), and participates in the stress-induced release of these hormones and possibly in the suckling- and estrogen-induced PRL release. The release of GH and TSH is predominantly inhibited by histamine; however, uncertainty exists regarding its role and the hypothalamic factors involved. Histamine increases the secretion of LH in females (mediated by GnRH), and may be involved in the mediation of the estrogen-induced LH surge. AVP and oxytocin are stimulated by histamine, probably by an effect in the supraoptic and paraventricular nuclei of the hypothalamus.

A dense peptidergic innervation has been demonstrated in the substantia innominata region in postmortem specimens of human brain using immunocytochemical techniques. A peptidergic innervation of the nucleus of Meynert - the prominent nucleus of this area containing the cholinergic cell bodies which innervate the cerebral cortex - has been demonstrated by immunostaining with antisera against the following eight neuropeptides: somatostatin, substance P, cholecystokinin octapeptide, vasoactive intestinal polypeptide, met-enkephalin, ACTH, alpha-MSH and oxytocin. Other immunocytochemical features of the substantia innominata region include a dense band of peptide immunoreactivity beneath the medial aspect of the anterior commissure and islands of somatostatin and substance P terminal immunoreactivity in the rostral part of the substantia innominata. Somatostatin immunostained cell bodies have been located in a discrete area of the bed nucleus of the stria terminalis and in the rostral portion of the substantia innominata, nucleus accumbens and the ventral part of the putamen. The dense band of peptide immunoreactivity beneath the medial aspect of the anterior commissure consists of ribbon-like processes stained with antisera against somatostatin, substance P, cholecystokinin octapeptide, vasoactive intestinal polypeptide and met-enkephalin. Less intense immunostaining of ribbon-like elements is also present in the globus pallidus. The presence of a peptidergic innervation to the nucleus of Meynert suggests a possible important modulatory role in cortical cholinergic function.

Neuropeptides and biogenic amines known to be present in neurons or afferent terminals in the paraventricular nucleus (PVH), supraoptic nucleus (SON) and/or lateral hypothalamus (LH) were added to small areas of these structures obtained by micropuncture and cyclic adenosine monophosphate (cAMP) levels were measured. cAMP accumulation occurred in PVH, SON and LH in response to neuropeptides of the secretin family, such as vasoactive intestinal peptide (VIP) and in response to catecholamines. Bradykinin, alpha-melanocyte-stimulating (alpha-MSH), luteinizing hormone-releasing hormone (LH-RH), oxytocin and carbamylcholine stimulated cAMP accumulation selectively in one or two of the above structures. Glucagon, cholecystokinin (CCK), somatostatin (SRIF), corticotropin-releasing factor (CRF), thyrotropin-releasing hormone (TRH), adrenocorticotropin (ACTH), melanocyte-stimulating hormone (MSH), methionine enkephalin (Met-Enk), beta-endorphin, neurotensin, bombesin and angiotensin II did not effect cAMP levels while leucine enkephalin (Leu-Enk), arginine vasopressin and gamma-aminobutyric acid (GABA) elicited regionally selective decreases in basal levels of cAMP. When interactions between some of these compounds were measured, VIP and norepinephrine exerted a more than additive effect on cAMP elevation in the PVH, while the effect on cAMP of the SON and LH was additive.

We have previously demonstrated that intracerebroventricular (ICV) administration of oxytocin (OXY) enhanced grooming behaviors in male and female rats at a 1 microgram dose. In the present study female rats were injected ICV with 1 microgram OXY or equimolar doses of other peptides. At this dose arginine-vasopressin (AVP), arginine-vasotocin (AVT) and lysine-vasopressin (LVP), as well as alpha-MSH, were as effective as OXY in increasing grooming behavior. At equimolar doses, ACTH1-10, tocinoic acid (the ring structure of OXY) and Pro-Leu-Gly-NH2 (the tail structure of OXY) had no significant effect on grooming behavior. The potency of AVP and AVT was determined across a 0.05-5 microgram dose range. Grooming scores increased in an apparent linear manner across a similar OXY dose range. Both AVP and AVT, however, manifested an inverted U grooming response curve. Maximum grooming scores resulted from a 0.1 microgram dose of AVT or a 0.5 microgram AVP dose. Analyses of the aspects of grooming separately found that nonapeptides OXY, AVP and AVT all elevated body grooming, washing, and scratching. Because AVT and AVP administration resulted in grooming scores significantly higher than OXY at lower doses, we concluded that the CNS is more sensitive to the effects of AVT and AVP on grooming behavior than OXY.

Oxytocin (OXY) and arginine-vasopressin (AVP) are widely distributed within the brain and have a number of behavioral effects resulting from central administration. We have previously found that central OXY administration accelerated the onset of maternal behavior in ovariectomized (OVXed) estrogen-treated nulliparous rats. We now report that intracerebroventricular (ICV) injections of OXY enhance lordosis behavior in OVXed estrogen-treated rats. After treatment with 0.15, 0.20, or 0.25 micrograms EB IM for three days, OXY (800 ng) infusion ICV on the fourth day produced a significant increase in lordosis behavior between 20 and 90 minutes after administration. Doses of OXY between 0.8 and 5 micrograms injected ICV significantly increased lordosis behavior in animals pretreated with 0.5 micrograms EB for three days. In other OVXed rats treated with 0.5 micrograms EB for three days, ICV injections of 1 micrograms OXY or an equimolar dose of AVP significantly increased lordosis while equimolar doses of ACTH1-10, ACTH4-10, arginine-vasotocin, GnRH and alpha-MSH did not significantly increase lordosis behavior over saline vehicle levels. ACTH1-24 significantly lowered lordosis quotients. We have concluded from these data that central administration of OXY (and possibly AVP) enhance female sexual receptivity. This effect is estrogen dependent, dose related and under our test conditions, specific to OXY and AVP.

The intracerebroventricular (i.c.v.) injection of oxytocin, in doses ranging from 5 to 90 ng (5-90 pmol) induced penile erection and yawning in male rats. Such response was not induced by doses of the peptide higher than 100 ng, nor by equimolar doses of i.c.v. [Arg8]vasopressin, ACTH-(1-24), alpha-MSH, rat corticotropin-releasing factor (rCRF), delta sleep-inducing peptide, neurotensin or substance P. Oxytocin-induced penile erection and yawning were prevented by atropine and morphine, but not by methylatropine or the opiate antagonist naloxone. Haloperidol, a dopamine receptor antagonist, was ineffective at low doses; it partially prevented penile erection but not yawning at high doses. Since oxytocin is present not only in the neurohypophysis but also in other brain areas, our results suggest that oxytocin is implicated in the regulation of penile erection and yawning, and provide further evidence that oxytocin acts as a neuropeptide in the central nervous system.
