Isolated rat paraventricular (PVN) and supraoptic (SON) nuclei were perifused in vitro and
oxytocin
and vasopressin releases were measured by radioimmunoassay during rest and during electrical stimulation. Stimulations at a frequency of 10 Hz (10-s bursts, every 10 s for 5 min) and an intensity of 4 mA, induced significant hormone release only with long duration pulses (10 ms). Short pulses (1 ms) applied at various frequencies (10, 20, 40 or 80 Hz) and intensities (4, 5, 10 or 20 mA) had no effect. The electrically evoked release of both hormones was not affected by tetrodotoxin (TTX), a sodium channel blocker, but was blocked in low-calcium medium or in the presence of gallopamil hydrochloride (
D-600
), a calcium channel blocker. These results suggest that, following electrical stimulation,
oxytocin
and vasopressin are released locally within the magnocellular nuclei even when blocking action potentials. The possibility of dendritic release is discussed.

When rat myometrium that had been depleted of Ca by incubation with 3 mM EGTA for 50 min was challenged with 10(-2) unit/ml
oxytocin
, it showed sustained contraction in a medium with no added Ca (Ca-free contraction). It also showed Ca-free contraction of similar magnitude in the presence of 1 mM EGTA. The effects on this contraction of divalent cations (Co2+, Ni2+ and Mn2+) singly and in combination with
D-600
(3 X 10(-6) M) were investigated. Co2+ and Ni2+ potentiated Ca-free contraction concentration-dependently, and their effects were greater in the presence of
D-600
. In contrast, Mn2+ evoked a triphasic response; first transient potentiation, second relaxation, and third persistent increase in tension.
D-600
did not block the first or second, but blocked the third, resulting in persistence of the second phase of relaxation. The relaxing action of papaverine on Ca-free contraction was not affected by
D-600
. Isoproterenol and dibutyryl cyclic AMP also relaxed the contraction.

The oxytocic activity of the hot methanol extract (HME) of the leaves of Monechma ciliatum was compared with other uterine stimulants like ergometrine,
oxytocin
, 5-hydroxytryptamine (5-HT), acetylcholine (ACh) and prostaglandins (PGs) E2 and F2alpha (PGE2 and PGF2alpha) in the presence of some antagonists in an attempt to explain the mechanism of action of the extract. The effects of the reference drugs on uteri isolated from rats pretreated with HME for 2 weeks were also observed. Atropine blocked the effect of ACh and partially blocked those of HME while L-366-948 blocked only the effect of
oxytocin
. Indomethacin inhibited the effects of HME as well as all the other drugs, except the PGs and ACh.
D-600
blocked the effect of all the drugs including HME. Methysergide antagonised only the effect of 5-HT and partially blocked ergometrine. Prolonged treatment altered the uterine musculature and the activity profile of the reference drugs. These results suggest that the HME may be acting by more than one mechanism to contract the uterus and explains the mechanism of the anti-implantation activity of the plant.
