The 5S rRNAs from Sulfolobus solfataricus and Pyrodictium occultum were digested to nucleosides and analyzed using directly-combined HPLC/mass spectrometry. P. occultum 5S rRNA contains two modified nucleoside species, N4-acetylcytidine (ac4C) and N4-acetyl-2'-O-methylcytidine (ac4Cm). Oligonucleotides were generated from P. occultum 5S rRNA by RNase T1 hydrolysis, and their molecular weights were determined using electrospray mass spectrometry and compared with those predicted from the P. occultum 5S RNA gene sequence. Deviation in mass between expected and observed molecular weights permitted ac4Cm to be located at position 35, in the nonanucleotide

-CACC[ac4Cm]G, and the ac4C in one or both of two (C,U)G trinucleotides. 2'-O-Methylcytidine is unambiguously characterized in S. solfataricus 5S rRNA, confirming earlier tentative assignments at the analogous sequence position (Stahl, D.A., Luehrsen, K.R., Woese, C.R., and Pace, N.R. (1981) Nucleic Acids Res., Vol. 9, pp. 6129-6137;

Root nodules infection of different faba bean (Vicia faba L.) cultivars by bean yellow mosaic virus (BYMV) and the effect of the disease on the specific activity of the nodule are reported. The disease reduced the fresh weights of tops, roots, root nodules and induced premature nodule decay and/or nodule drop. Six local cultivars: Barekat, Iranshahri, Saraziri, Aljazayeri, Shakhbozi and Zohre of faba bean were selected and inoculated with BYMV under greenhouse conditions. ELISA test (
DAS
-ELISA) with specific BYMV antibody (DSMZ AS0471) demonstrated that nodules from faba bean plants which had been inoculated with BYMV contain the virus too. Susceptibility of different faba bean cultivars was analyzed by ELISA. The relative accumulations of BYMV in the nodules were evaluated by mean ELISA values (OD405) of BYMV. There was significantly difference in cultivars. Cultivars went more susceptible from Barekat to Iranshahri, Saraziri, Aljazayeri, Shakhbozi and Zohre. High susceptibility of Zohre was confirmed in a second experiment including visual evaluation and
DAS
ELISA. Analysis by IC-RT-PCR revealed the presence of the virus in all nodules and amplified a 970 bp fragment with specific designed primers (Forward primer (5'-CT(AC) CA(AG) ATG GAG AA(CT) CC(CT) GC 3') and Reverse primer (5'-CCA AAG TTC
CAA
TCA CCA CC 3').

Background
: The differentially expressed proteins (DEPs) involved in the effect of hydrogen-rich water on myocardial ischemia reperfusion injury (MIRI) and their biological processes and signaling pathway were analyzed.
Methods
: 20 Wistar rats were randomly and equally divided into a control and a hydrogen-rich group.
Hearts
were removed and fixed in a Langendorff device. The control group was perfused with K-R solution, and the hydrogen-rich water group was perfused with K-R solution + hydrogen-rich water. Protein was extracted from the ventricular tissues, and GSR-
CAA
-67 was used to identify the DEPs between two groups. DEPs were analyzed through bioinformatic methods.
Results:
Compared with the control group, in the treatment group, the expression of 25 proteins was obviously decreased (
P
<0.05). For the DEPs, 359 biological processes, including the regulation of signaling pathways, immune reaction and formation of cardiovascular endothelial cells, were selected by GO enrichment analysis. Five signaling pathways were selected by KEGG pathway enrichment analysis.
Conclusions:
25 proteins that are involved in hydrogen-water reducing MIRI were selected by high-throughput GSR-
CAA
-67. The biological processes and metabolic pathways involved in the DEPs were summarized, providing theoretical evidence for the clinical application of hydrogen-rich water.
