SOC, a fungal growth medium composed of Solryth, oxgall, and caffeic acid, was evaluated as a medium to provide rapid, differential identification of Candida albicans and Cryptococcus neoformans. Using a variety of common isolation media to produce the yeast inocula, the germ tube methods tested ranked in the following order of decreasing sensitivity: SOC (97% +/- 1), serum (92% +/- 5), rabbit coagulase plasma with EDTA in combination with tryptic soy broth (89% +/- 5), TOC (89% +/- 6), and rabbit coagulase plasma with EDTA (83% +/- 4). In chlamydospore production, SOC also proved to be the most sensitive after 24 h incubation: SOC (96% +/- 2), TOC (80% +/- 2), and cornmeal-Tween 80 agar (14% +/- 3). Other medically important yeasts showed normal patterns of growth within 24 h on SOC, thus assisting in their identification. Eighty strains of Cryptococcus neoformans showed characteristic brown pigmentation on SOC and TOC within 18 h, while all other species of the genus Cryptococcus and 229 Candida isolates did not show a change in pigmentation.

The effect of thymalin and epithalamin on dissemination of tumors treated by pulsed laser radiation was studied in C57B1 female mice bearing subcutaneously transplanted melanoma B16 and Lewis carcinoma. Radiation was delivered from TOC-1001 neodymium laser, wave-length of 1,060 nm, pulse duration--1 msec and fluence--350-400 J/cm2. Drug dosage was 1 mg. Thymalin and epithalamin treatment potentiated the preventive effect of laser radiation on tumor dissemination. Application of the said agents was followed by an increase in antibody-producing cells (as measured by Jerne's procedure) in the spleen of irradiated melanoma B16--bearing mice.

Six different cultural media (corn meal agar, rice extract agar, chlamydospore agar, PCB, Tween 80-oxgall-caffeic acid and diluted milk) were compared for chlamydospore production by 224 yeastlike fungi isolates. Candida albicans formed chlamydospores to a variable degree in all of the media, as did C. stellatoidea to a lesser extent. C. tropicalis, C. parasilopsis, C. guilliermondii and C. krusei did no produce chlamydospores in any of the media tested. Statistically, the most productive media were the milk and TOC media. Milk medium is particularly useful because of its simplicity and economy.

Swimming pool water covers made of different plastics were investigated in laboratory experiments. The possible influence on the chemical and the bacteriological water quality (test bacterium Ps. (fluorescens) was observed. Strong-PVC-plastics proved to have no respectively hardly any chemical or bacteriological influence on the water quality. PE-soft-plastics showed clearly higher chlorine consumption and TOC counts. The influence on bacterial growth was here either inhibition or of nutrient character. A direct colonisation of the plastic was not to be seen--neither in vivo nor in vitro. There are no principal objections against the use of plastics swimming pool covers from hygienical point of view. Some limitating aspects are discussed.

Because the chlamydospore formation test for the identification of C. albicans is frequently utilized in most clinical Mycology laboratories, a comparative study of the different methods employed for the test was carried out. A total of 108 strains of yeast forming organisms belonging to C. albicans, C. tropicalis, C. stellatoidea, C. pseudotropicalis, C. parakrusei, C. guillermondii, C. neoformans, C. laurentii, C. albidus, C. candidum, T. glabrata, R. rubra, T. cutaneum, and S. carlsbergensis was used. The following culture media were compared: corn meal agar, rice agar, chlamydospore agar, PCB, and the recently described TOC medium (tween 80, oxgall and caffeic acid). Chlamydospore formation was positive, respectively, in 54.2%, 66.6%, 71.4%, 82.8%, and 91.4% and pseudomycelia formation, in 85.7%, 80%, 71.4%, 94.2%, and 91.4%. Only Cryptococcus neoformans produced in the TOC medium a characteristic coffee brown pigmentation which distinguished it from the other fungi. The results demonstrate the usefulness of the TOC medium in comparison with other culture media for the chlamydospore formation test for the diagnosis of C. albicans, and for the diagnosis of C. neoformans because of the characteristic pigmentation it produces.

Chronic administration of amiodarone (AMIO), widely used by clinicians for the treatment of therapy-resistant cardiac arrhythmias, is frequently associated with serious side-effects. AMIO and its main metabolite desethylamiodarone (DEA) are known to induce phospholipidosis in vivo and in cultured cells presumably by inhibition of lysosomal phospholipid degradation. D-alpha-Tocopherol = vitamin E (alpha-TOC) was able to reduce AMIO and DEA toxicity in cell cultures. Results from the present study showed that alpha-TOC reduced phospholipidosis in cultured human skin fibroblasts chronically exposed to micromolar concentrations of AMIO and DEA and inhibited cumulative uptake of the drugs in a dose-dependent manner. A linear correlation was observed between cellular AMIO levels and phospholipid accumulation suggesting a stoichiometric relationship. alpha-TOC was also effective in clearing previously accumulated phospholipids after discontinuation of the drug treatment. The results can best be explained by an interference of alpha-TOC (a) with drug-phospholipid complex formation responsible for both phospholipid storage and drug accumulation, and (b) with pre-existing drug-phospholipid complexes, accelerating their dissociation and rendering phospholipids to substrates for lysosomal phospholipases. The finding raises hope that side-effects of AMIO and DEA can be prevented or made reversible by the administration of alpha-TOC. It must, however, be proven that the antiarrhythmic drug will still be effective.

TOC-39, a new parenteral cephalosporin, is a hydroxyimino-type cephem antibiotic with vinylthio-pyridyl moiety at the 3 position. TOC-39 was evaluated for antibacterial activity against various clinically isolated strains. TOC-39 had excellent activity, stronger than that of methicillin, oxacillin, the cephalosporins tested, imipenem, gentamicin, minocycline, tobramycin, ofloxacin, and ciprofloxacin against methicillin-resistant Staphylococcus aureus (MRSA) and had an MIC comparable to that of vancomycin (the MICs of TOC-39 and vancomycin for 90% of the strains tested were 3.13 and 1.56 micrograms/ml, respectively). Against Enterococcus faecalis strains, which are resistant to cephalosporins, TOC-39 was twice as active as ampicillin. Against methicillin-susceptible S. aureus, coagulase-negative Staphylococcus spp., and Streptococcus pneumoniae, TOC-39 was twice as active as or more active than cefotiam, ceftazidime, flomoxef, and cefpirome. Against Streptococcus pyogenes, TOC-39 was superior to cefotiam, ceftazidime, and flomoxef and was similar to cefpirome. In addition, the activity of TOC-39 was equal to or greater than that of cefotiam, ceftazidime, flomoxef, and cefpirome against Haemophilus influenzae, Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis, and Morganella morganii. In terms of bactericidal effect against MRSA, TOC-39 was superior to vancomycin. No mutant resistant to TOC-39 or vancomycin was obtained from susceptible MRSA strains. In murine systemic infection models, TOC-39 showed potent activity against S. aureus and E. coli. Against highly MRSA, the activity of TOC-39 was comparable to that of vancomycin.

The effect of d-alpha-tocopherol on the progression of renal dysfunction was investigated in rats injected with adriamycin (ADR), a model of progressive glomerulosclerosis associated with the nephrotic syndrome. Treatment with d-alpha-tocopherol was started 1 day before or 1 day after ADR injections (BE-TOC or AF-TOC rats). When compared to rats without d-alpha-tocopherol treatment (ADR-CON rats), the serum total cholesterol and triglyceride levels were significantly lower in the BE-TOC and AF-TOC groups. In week 16, the LDL cholesterol level and the atherogenic index were both significantly lower in BE-TOC and AF-TOC rats than in ADR-CON rats. The urinary protein, serum creatinine, blood urea nitrogen, malondialdehyde, and systolic blood pressure levels as well as the glomerulosclerosis score were high in ADR-CON rats, and reduced in BE-TOC or AF-TOC rats. There were no significant differences in body weight and serum albumin between the three groups in week 16. It is concluded that d-alpha-tocopherol can improve hyperlipidemia and ameliorate glomerulosclerosis in rats with ADR-induced progressive renal failure. Thus, d-alpha-tocopherol may have the potential for clinical application to treat focal glomerulosclerosis.

Nitrogen and phosphorus removal in a methane fermentation plus activated sludge method type pig farm (M-III) with an intermittent aeration process (IAP) was evaluated in comparison with a continuous aeration process (CAP) based on the full-scale and bench-scale experiments. Operation conditions for the treatment system were the same except for the aeration program (in the CAP), a consecutive 24-hr aeration was used, whereas in the IAP, the aeration and non-aeration periods were alternated at intervals of 3:1 hr. BOD and TOC removal efficiencies with the intermittent aeration were as high as those with the continuous operation (92-98%). In the removal of nitrogen and phosphorus, large differences between IAP and CAP were observed. At an influent T-N/TOC ratio of 0.1, removal efficiencies for T-N in the bench-scale IAP was 70%, and for T-P was 22%, respectively. At an even higher influent T-N/TOC ratios of 0.3-1.0, the removal efficiencies for T-N were decreased to about 59-61%, whereas that for T-P were -0.3-13%. In a full-scale plant, removal efficiencies for T-N with IAP and CAP were 42% and -0.09%, respectively. The results of this study show the successful performance of a simple IAP for piggery wastewater: simultaneous, one-sludge denitrification with nitrification in single-activated sludge reactor in a pig farm.

A process on crystallized precipitation of ammonium by adding magnesium salt and phosphate was carried out to improve C/N ratio in swine wastewater. After completion of crystallized precipitation of ammonium, an intermittent aeration process with aeration and nonaeration periods alternated at interval of 1:1 hr day-1 is used for the improved swine wastewater (T-N/BOD = 0.14: BOD = 8200 mg/liter and T-N = 1166 mg/liter). The results obtained from the experiment show that the removal ratios of T-N and NH4-N are 91% and 99%, respectively. T-P is not removed, while the removal ratio of PO4-P is 60% as 3% of CaCl2 liquid is added. The results also indicate that dilution with water is effective to improve the removal of phosphorus even if raw swine wastewater contains high concentrations of T-N, T-P, BOD, and TOC.
