A unique protein was detected in the plasma of rheumatoid arthritis patients by two-dimensional electrophoresis. We named it rheumatoid arthritis specific protein (RASP). According to the pattern and concentration of its spot, RA patients were classified into three groups as RASP+, RASP +/- and RASP-. RASP+ was detected only in RA and its occurrence was 45/120 (38%). The occurrence of RASP including RASP +/- was 84/120 (70%) in RA, 6/13 (46%) in SLE, 2/8 (25%) in
PSS
, 11/51 (22%) in liver disease and 4/44 (9%) in normal. RA patients with RASP+ indicated significantly high level of
ESR
, CRP and platelet counts comparing with RA patients with RASP-. These results suggested that RASP might be a good indicator for the diagnosis and evaluation of the activity of RA. RASP was purified with gel chromatography repeated three times. RASP was very similar with immunoglobulin G in its biochemical and immunological properties.

Currently there are no widely accepted criteria for the diagnosis of MCTD. In this work we attempted to define the clinical profile of a group of 68 patients with anti nRNP antibodies, detected by immunoprecipitation in 0.6% agarose. The diagnosis of each collagen vascular disease was established in every patient, who met with the strict diagnostic criteria either at clinical presentation or during the follow-up period. Twenty-eight patients had SLE, 9 had classical erosive RA, three had
PSS
and one had PM. The only distinctive features in the group of SLE with anti nRNP was an increased incidence of anti Sm antibodies (p less than 0.05). In the RA group there was a trend towards a high frequency of Raynaud's phenomenon and swollen hands. At clinical presentation twenty-seven patients did not fulfil enough criteria to be diagnosed of any of the well-defined collagen vascular disease. They presented an undifferentiated syndrome, characterized clinically by Raynaud's phenomenon (100%), swollen hands (88.9%) and joint symptoms (88.9%), with scarce tendency of developing severe systemic manifestations. The main laboratory abnormalities in this group were hypergammaglobulinemia, mildly increased
ESR
, abnormal levels of CIC, negative anti nDNA and anti Sm antibodies, and the virtual absence of hypocomplementemia. During a clinical course of 96 +/- 72.5 months only one patient evolved into another collagen disease (SLE). The clinical course in the remaining cases, was stable improving with low doses of prednisone and/or NSAID. We suggest considering this undifferentiated syndrome as a distinct entity, for which the already classical term of MCTD could be reserved.

Modern pig production needs new tools for fast, reliable, more effective breeding. In the present paper we present a chip containing 45 SNP (Single Nucleotide Polymorphisms) which enables the determining of 1 genetic disease (
PSS
-Porcine Stress Syndrome), 4 QTLs genes: PRKAG3, CAST, MC4R and
ESR
, which together with the remaining SNPs create a panel useful in marker-assisted selection and veterinary control. The SNPs were genotyped using the PCR-APEX (Arrayed Primer Extension) technique. Special attention is paid to evaluation of the 45 SNP chip as an alternative approach to parentage and identity control. Based on allele frequency estimations, for a sample of 88 individuals of commercial pig lines, the probabilities that a randomly chosen candidate parent would be excluded from paternity or maternity were estimated to be 99.9% when genotypes of both parents and a progeny were known, and 98% when the genotypes of only one parent and a piglet were available. The marker set presented here also reached a probability of identity in the order of 10(-16), which allows for unequivocal discrimination of animals or their products among billions of individuals. Further improvements for upcoming chip versions were also considered.

Macromolecular characteristics and morphology of water-soluble complexes between sodium poly(styrene sulfonate) (
PSS
) and tetradecyltrimethylammonium bromide have been followed as a function of surfactant-to-polymer charge ratio (S/P) to elicit possible changes in the complexation mechanism. As revealed by light scattering, shorter
PSS
(30 and 150 repeat units) yield multichain complexes while longer
PSS
(450 and 5000 repeat units) form single-chain species throughout 0 < S/P < 0.9. Irrespective of
PSS
chain length, the complexes exist in solution in a swollen coil conformation and undergo a compaction with S/P but never collapse into a globule. Even when the free
PSS
chain is too short to coil (30 repeat units), the complexes adopt a coiled conformation due to multichain aggregation. Morphological changes (manifested by a hypochromic shift in UV spectra of the complexes at S/P < 0.5 and an increase in the local surfactant mobility observed at S/P > 0.5 by
ESR
) strongly suggest a change in the formation mechanism of the complexes with a transition near S/P = 0.5.
