A TEM specimen preparation method is described, with the aid of which electron transparent foils can be obtained across the external surface of a specimen. After careful pre-treatment, steel specimens have been electrolytically coated with nickel. Conventional thinning in a plane cutting the substrate-coating interface, gave thin foils displaying the internal structure as a function of depth under the initial free surface. The method has also been applied to minute metal particles, of dimensions too small to allow manipulating and foil preparation by conventional methods. Image examples are shown, and the applicability of the method is discussed.

This paper presents some aspects of the authors' experimental research on blast injury in the past two years. The main results are as follows: (1) A new designed 39 meter-long shock tube for biological test has been built in the laboratory. Its maximal overpressure values are 215 kPa (in open condition) and 505 kPa (in closed condition). It may meet the need for inflicting blast injuries with various degree of severity. (2) A study of the effect of simulating gun muzzle blast wave on sheep indicated that in the single explosion, the threshold overpressure values inflicting the injury of internal organs were: Lung-37.27 kPa, G-I tract-41.0 kPa; the upper respiratory tract-negative until 73 kPa, while in the multiple (20 times) explosions, they were 23.7, 23.7 and 41.4 kPa, respectively. (3) Using TEM, SEM and some other special techniques, such as morphometry, freeze-fracture technique, labelled lanthanum nitrate technique, etc, it was demonstrated that in the lung with blast injury there were significant pathological changes in pulmonary capillary endothelium, alveolar epithelium and their intercellular junctions with apparent increase of permeability. (4) It has been shown that parallel superficial stripelike hemorrhage typical for lung blast injury is "Intercostal marking" instead of "Rib marking". (5) A new type of material (foamy nickel) for protection against blast wave is presented. It was proved that the material can effectively weaken or eliminate the effect of blast wave on human body.

Binding sites for Con A and WGA were detected on bovine spermatozoa during epididymal maturation. We used colloidal gold as an EM-marker. The spermatozoa were treated according to a two-step method for lectin and colloidal gold, then adsorbed to lysine-coated nickel grids and subsequently examined by TEM in toto. Using this method we rapidly got information about the topographic distribution of lectin-binding sites. Major differences exist for WGA between caput and cauda spermatozoa. Conceding that cell-thickness poses some limitation, we consider this method to be practical and especially useful in studies concerning topographic distribution of cell surface components in single cell systems.

Mesoporous silica gels modified due to pyrolysis of acetylacetone or acetylacetonates (AcAc) of zirconium [Zr(AcAc)(4)], titanium [titanyl TiO(AcAc)(2)], nickel [Ni(AcAc)(2)], zinc [Zn(AcAc)(2)], chromium [Cr(AcAc)(3)], and cobalt [Co(AcAc)(2)] were studied using nitrogen adsorption-desorption, TPD-DTG, TEM, XRD, and XRF methods. Grafted C/X phases consist of metal compounds (X denotes oxide, silicate, or metal crystallites) and pregraphite pyrocarbon, whose characteristics can be varied by changing the metal in M(AcAc)(n). The structural parameters of C/X/SiO(2), such as the specific surface area of micro- and mesopores, pore volume, pore size and fractal dimension distributions, and adsorptive ability, depend nonlinearly on the concentrations of the C/X deposit due to alterations in the topology of grafted matters with increased concentrations and a possible catalytic effect of the X phase on pyrolysis. Two beta-lactamases, penicillinase type I from Bacillus cereus and TEM-1 beta-lactamase from Haemophilus ducreyi, were immobilized on a Chelating Sepharose Fast Flow column loaded with Ni2+ in an active form. Flow-injection analysis of beta-lactams was performed by using an enzyme column reactor fitted into the enzyme thermistor. With both enzymes it was possible to monitor both penicillins and cephalosporins. Moreover, Michaelis constants of the TEM-1 beta-lactamase were markedly increased upon immobilization for all substrates, especially carbenicillin, cephaloridine, and cefoperazone.

Porous nickel and cobalt oxides were prepared using their alkoxides as inorganic precursors. The stabilization of the mesostructure is especially critical for divalent elements such as Ni and Co, which do not form any network structure, like silicates. The lack of a network-forming multivalent bond is the probable reason why no stable mesoporous oxides have been synthesized for divalent elements yet. Here we have reported our attempt to synthesize porous oxides of Ni and Co. Octadecylamine has been used as the organic structure-directing agent. The product obtained was put under solvent extraction and calcination at various temperatures to remove the surfactant, followed by characterization using XRD, TEM and BET measurements. The FT/IR and thermal analyses (TGA and DSC) were also carried out for supporting information, such as extent of removal of surfactant from the pores of the metal oxide. A relatively better surface area has been obtained for the Co oxide, but in Ni the surface area found is not as good. A possible reason for that has been discussed. The porous (solvent extracted) cobalt oxide has been used as a catalyst in the oxidation reaction of cyclohexane in mild conditions. The catalyst has shown relatively better conversion of cyclohexane into cyclohexanone and cyclohexanol than the nanostructured cobalt oxide catalyst of regular structure.

Results from aromatic aminations and Kumada couplings, together with spectroscopic analyses (TEM, EDX, ICP-AES, React-IR), reveal that catalysis using nickel-on-charcoal (Ni/C) is most likely of a homogeneous rather than heterogeneous nature. In the course of a reaction with Ni/C, nickel bleed from the support was calculated to be as high as 78%. However, the existence of an equilibrium for this homogeneous species between nickel located inside vs outside the pore system of charcoal strongly favors the former, thus leaving only traces of metal detectable in solution. This accounts for virtually complete recovery of nickel on the charcoal following filtration of a reaction mixture and allows for recycling of the catalyst. TEM and EDX data were used to explain different reactivity profiles of Ni/C, which depended upon the method of reduction used to convert Ni(II)/C to Ni(0) as well as the level of nickel loading on the support.

Ni2+ and Cu2+ are the major heavy metal ions in electroplating wastewater of Hong Kong. In the present study, Pseudomonas putida 5-x cell biomass was used to remove Ni2+ from electroplating effluent. Ni2+ adsorption capacity of P. putida 5-x cell biomass cultured in sulphate-limiting medium was found to be minimum in early logarithmic growth phase, and maximum of 28.1 mg g(-1) in late stationary growth phase. Pretreated cells by 0.1 mol L(-1) HCl could greatly enhance the Ni2+ adsorption capacity of cell biomass from 28.1 to 36.7 mg g(-1) and had no significant effect on biomass loss. The adsorption process of P. putida 5-x fresh cells and pretreated cell all could be expressed with Freundlich isotherm. TEM analyses indicated that acidic pretreatment degraded the superficial layer-capsule outside of the fresh cell to improve the adsorption capacity of cell to Ni2+. The Ni2+ bound by P. putida 5-x cell biomass could be efficiently recovered using 0.1 mol L(-1) HCl, and the cell biomass could be reused at least five cycles for Ni2+ removal and recovery with 93% above removal efficiency and 98% above recovery rate. Owing to the Cu2+ presented in electroplating wastewater inhibiting Ni2+ adsorption process by P. putida 5-x cell biomass, two-stage biosorption processes should be designed to remove and recover Cu2+ and Ni2+ sequentially from electroplating effluent.

Near-edge fine structures of the metal L(2,3) and O K-edges in transition metal-oxides have been studied with a transmission electron microscope equipped with a monochromator and a high-resolution imaging filter. This system enables the recording of EELS spectra with an energy resolution of 0.1eV thus providing new near-edge fine structure details which could not be observed previously by EELS in conventional TEM instruments. EELS-spectra from well-defined oxides like titanium oxide (TiO(2)), vanadium oxide (V(2)O(5)), chromium oxide (Cr(2)O(3)), iron oxide (Fe(2)O(3)), cobalt oxide (CoO) and nickel oxide (NiO) have been measured with the new system. These spectra are compared with EELS data obtained from a conventional microscope and the main spectral features are interpreted. Additionally, the use of monochromised TEMs is discussed in view of the natural line widths of K and L(2,3) edges.
