The effects are reported of a combination therapy of i.v. C. parvum and cyclophosphamide on the survival time and immune responses of patients with inoperable squamous-cell carcinoma of the bronchus. The immune status of the patients was evaluated by determining the antibody response to C. parvum, the E and
EAC
rosettes, the PHA response of blood lymphocytes, the skin-test reactivity to Candida and PPD, the response to DNCB and the chemotaxis and NBT-dye reduction capacity of neutrophil leucocytes. The survival time of patients treated with the combination therapy was found to be significantly shorter than that of untreated patients and of those receiving cyclophosphamide only. Severe side effects were observed after C. parvum infusions, with no decrease on repeated administration. The effect of C. parvum on the different immune parameters of cyclophosphamide-treated patients was negligible, though there was a normal antibody response to C. parvum.

-26, an extended-spectrum beta-lactamase has been characterized in clinical isolates of Klebsiella pneumoniae and Escherichia coli derived from patients on the Paediatric Oncology Unit of St James's University Hospital, Leeds. The nucleotide sequence of this beta-lactamase gene (blaTEM26b) was determined, and compared with the nucleotide sequences of other
TEM
-type beta-lactamases. The blaTEM26b gene was found to differ from blaTEM12b by a single nucleotide. This difference causes the substitution of glutamic acid in blaTEM12b for lysine in blaTEM26b at position 102 in the predicted amino acid sequence. The blaTEM12b gene was first described in an isolate of Klebsiella oxytoca from a patient nursed on the same unit that yielded the strains that carry blaTEM26b. However, the blaTEM26b gene differs at no less than six nucleotides from the nucleotide sequence encoding the
TEM
-26 beta-lactamase that was first described in isolates from

patients nursed in the Children's Hospital, Stanford, California, USA. This indicates that the genes encoding

We have established eleven T cell hybridoma cell lines to investigate mechanisms controlling interaction of T lymphocytes with endothelial cells as well as extracellular matrix (ECM) proteins at the clonal level. T cell hybridomas were characterized and subdivided into four groups on the basis of their interaction behavior with high endothelial venules (HEV). Group 1 (G1) exhibited strong adhesiveness. The binding was temperature- and divalent cation-dependent. Group 2 exhibited both adhesiveness and transendothelial migration (

has been limited by the development of drug resistance. Verapamil and ligustrazini were found to show significant potentiating effect on the cytotoxicity of ADR in vitro and decreased the IC50 of ADR by 84.85% and 30.82% of the resistant line, respectively, and no effect was found on the IC50 in the sensitive line. The Glutathione-S-transferase activity of ADR-resistant and sensitive mouse

The excess risk of tumours exposed to asbestos were previously compared with the results of rat inhalation experiments. It could be demonstrated that humans at the workplace suffer from a tumour risk at fibre concentrations which are 300 times lower than those needed in the rat inhalation model to produce the same risk. However, the estimation of human risk was based on the study of workers at a chrysotile textile factory, whereas animal experimental results were related to exposure to amphiboles. Since for this comparison the risk of
cancer
due to exposure to amosite or crocidolite fibres at the workplace is of interest, quantitative exposure-response relationships for lung cancer and mesothelioma for the white workforce of South African amosite and crocidolite mines were discussed. On comparing the risk of lung cancer in this study with the risk of lung cancer for chrysotile textile workers, it can be concluded, that the risk of lung cancer and mesothelioma from crocidolite and amosite was higher than in the chrysotile textile factory. It could be also demonstrated, on the basis of a study of the lung burden of mesothelioma cases and of controls, that a significantly increased odds ratio of about 5 was established at amphibole concentrations of between 0.1 and 0.2 f micrograms-1 dry lung (WHO fibres longer than 5 microns from
TEM
analysis). On the other hand, carcinogenic response was observed at a fibre concentration 6000 times higher in animal inhalation experiments with crocidolite asbestos (SEM analysis of WHO fibres). As a result of these findings, it has been concluded that inhalation studies in rats are not sufficiently sensitive for the detection of hazards and risks to humans exposed to man-made fibres.

This report evaluates 74 fine-needle aspiration biopsies processed for transmission electron microscopy with subsequent surgical procedure. The specificity of diagnosis obtained by cytology alone was compared to that obtained by cytology and electro microscopy, using histologic diagnosis as the gold standard. When cytology gave a diagnosis of
malignancy
but could not give tumor category or type, electron microscopy could correctly give both. When cytology could give tumor category but not type, electron microscopy correctly identified type in the majority of cases. When cytology gave tumor category and type, electron microscopy confirmed the diagnosis. Transmission electron microscopy is very helpful when the cytopathologist can diagnose
malignancy
but cannot give tumor category and/or type. When the cytopathologist is specific in his/her diagnosis,

is not as helpful.

Toxoplasma gondii, an intracellular protozoan infecting many kinds of eukaryotic cells, has been used to experimentally infect macrophages, epithelial cells, fibroblasts, and various
cancer
cells, but rarely T and B lymphocytes or granulocytes. The present study was performed to determine the susceptibility of murine (BALB/c or CBA) splenic T and B lymphocytes, and granulocytes to infection with T. gondii RH tachyzoites. The ultrastructure of the infected host cells was observed by
TEM
, and the degree of intracellular parasite proliferation was quantified using 3H-uracil uptake assay. At 24 hrs post-culture, the host cell cytoplasm was found to contain 1 or 2, or a maximum of 7-8 tachyzoites. Infected T lymphocytes demonstrated a peripherally displaced nucleus, a parasitophorous vacuole enveloping the parasite, and an increased number of mitochondria. In B lymphocytes infected with tachyzoites, RER was not well developed compared to uninfected B lymphocytes. Uninfected granulocytes contained many electron-dense granules, but T. gondii-infected granulocytes demonstrated a decreased number of granules. Based on the 3H-uracil uptake assay, the susceptibility of T and B lymphocytes, and granulocytes, to infection with T. gondii tachyzoites was fairly high irrespective of cell type and strain of mouse. This strongly suggests deterioration in the functioning of infected host immune cells.

The antineoplastic activity of a platinum complex K4 (Pt Cl2 ATP) (Pt-ATP) has been investigated in transplantable tumors, both in an ascitic and solid (Ehrlich ascites carcinoma) tumor model. It has been observed that the drug at the total dose of 10 mg/kg body weight successfully inhibited the tumor burden both in ascitic and solid tumor system and subsequently increased the host's life span. An assessment of the in vitro (3H) thymidine incorporation into TCA precipitable material of

tumor cells done in the presence of Pt-ATP indicates that the drug inhibits (3H) thymidine incorporation in tumor cells. The drug has no appreciable toxic effect on the peripheral blood cells as well as bone marrow and spleenic cellularity.
