The chemotherapeutic action of 5-fluorouracil monotherapy on HRS-

in mice was compared with those of 5-fluorouracil nucleoside combinations (thymidine, cytidine or guanosine). The curative action of 5-fluorouracil was potentiated without increasing its toxicity, when cytidine or guanosine were applied at definite intervals before or after 5-fluorouracil.

The biological activity of a hot-water extract from delipidated BCG, designated as HSA (Hot-water Soluble Adjuvant), was investigated. The HSA did not induce hyperreactivity to bacterial endotoxin. The hot-water extract from which nucleic acids had been removed by streptomycin (SM-HSA) was found to enhance the delayed-type hypersensitivity as evidenced by the results of footpad reaction of mice. The HSA and SM-HSA could be injected to mice by an intraperitoneal route for 20 consecutive days without undesirable side effects. A comparative study was made on the effects of HSA in relation to the duration and doses of treatment with HSA using the ddI mice inoculated with
Sarcoma
-180. The most remarkable effect was observed when 0.25 mg of HSA had been injected for 20 consecutive days. Also SM-HSA was found to exert antitumor activity when applied in the same manner as above. These results suggest that the presence of nucleic acids is not related to the biological and antitumor activities of the hot-water extract.

-180. As well as a marked increase in the relaxation time of the tumour itself, mice injected with
Sarcoma
-180 showed a rise in the kidney relaxation time, along with blood relaxation times which were often either above or below the range found in normal mice. Correlation coefficients for the relaxation times of the various organs were calculated and found to be very sensitive to the physiological state of the animal. For healthy mice, the correlations were reasonably high, but even mild stress decreased the correlations, whilst the presence of cancer almost completely destroyed the correlations. These findings are all consistent with the hypothesis that cancer affects the water regulatory system of the animal as a whole.

We studied the effect of thyroxine treatment on tumor growth and metastases resulting from tumor implants on the hind feet of mice in two syngeneic systems. In control, untreated A/Jax mice, tumor

1 at Day 14 after implantation had average tumor weight of 582 +/- 60 (S.D.) mg and showed an incidence of 57% metastases to regional popliteal nodes and 5% metastases to thymus. In contrast, the thyroxine-treated group (40 microgram/mouse s.c., 5 times/week for 1 month) had an average tumor weight of 808 +/- 56 mg (p less than 0.001), and metastases to popliteal nodes and thymus were 90 and 35%, respectively. In another syngeneic tumor system, Lewis fibrosarcoma was implanted in C57BL/6J mice, and the tumor weight and metastatic index (derived from the number and size of the pulmonary tumor foci) were determined at Day 28. Again, the synthetic L-thyroxine treated group showed a significant enhancement tumor growth and metastatic index. The mean tumor weight in the treated group was 385 +/- 26 mg (control, 694 +/- 25 mg; p less than 0.005) and metastatic index was 84 +/- 29 (control, 30 +/- 25; p less than 0.001). Induced hypothyroidism (treatment with 131I, 100 microCi/mouse i.p.) showed the reverse effect on both tumor systems. These results suggest that both tumor systems are dependent on thyroid hormones for their growth and spread.

Anti-tumour activity of an aromatic retinoic acid analog has been tested on
