A comparative histological investigation of the thymus, spleen and inguinal lymph nodes has been performed in the rats flown for 18.5 days on board the biosatellite "Cosmos-936" under the conditions of weightlessness and artificial gravitation (acceleration 1 g) imitating terrestrial magnetism. It has been stated that in the animals that were under the conditions of weightless ness during the flight and were sacrificed 4.5--13 h after they have landed the Earth, accidental involution of lymphoid organs is noted with morphological signs in them of an acute stress in the form of massive degeneration of the thymus lymphocytes and neutrophilic infiltration of the spleen. In rats that during the flight were subjected to the effect of artificial gravitation there was noted neither involution of the lymphoid organs nor morphological signs of acute stress in them. One of the main cause of acute stress in the rats subjected to weightlessness during the space flight is supposed to be transition to the terrestrial gravitation.

Structural-functional organization of the lymphoid organs and functional state of the adrenals have been studied in animals, subjected to cold in early postnatal period, as well as changes of the parameters mentioned to a short and prolonged cooling in mature rats. For the animals increase in the thymus mass and in reproduction rate of lymphocytes in the thymus, spleen and lymph nodes is specific against the background of high corticosteroid secretion. When the control animals are kept in cold for a long time, after the phase of an acute stress, accompanied with hypercorticism and a pronounced lymphatic effect, during the period of an increased cold stability, the high secretion of glucocorticoid hormones is accompanied with a certain activation of thymus-dependent zones in the peripheral lymphoid organs. In the mature rats, subjected to cold at early ontogenesis both stress-reaction to cooling and rearrangement in the regulatory systems studied does not develop at adaptation to cold.

A study was made of the content of corticosterone, thyroxine and triiodothyronine in the blood, of the thymic weight and cytomorphological characteristics of rat lymph nodes during prolonged cold (4-5 degrees C) adaptation. Interrelated alterations in the characteristics under study were revealed at different stages of adaptation. The morphofunctional recovery of the lymphoid tissue occurs in the phase of the increased specific stability in spite of the fact that the action of cold is continued and the blood corticosterone concentration is elevated. A new type of the endocrine-lymphoid relationships is supposed to be formed during lengthy adaptation, with this type being different from that both in the starting state and acute stress.

The histological study of the thymus, spleen and inguinal lymph nodes of monkeys exposed to head-down tilt (-6 degrees) for 7 or 19 days showed that the exposure caused an acute stress that resulted in the thymus involution and hypoplasia of the lymphoid tissue of the spleen and lymph nodes. The exposure produced fluid redistribution due to which the spleen blood content decreased and the mass of inguinal lymph nodes increased. This was induced by partial pooling of the lymph flowing from the lower extremities. The exposure also led to autoimmune developments.

The weight and histological appearance of the thymus from patients with myasthenia gravis were studied. Nine patients treated with corticosteroids were compared with 10 patients not treated with corticosteroids before thymectomy. The thymuses of corticosteroid-treated patients were found to have significantly increased fat and connective tissue, decreased germinal centers, and poorer corticomedullary differentiation than those of untreated patients. The glandular weight and degree of myoepithelial stroma were the same in both groups. The changes following corticosteroid treatment mimic those following aging or acute stress, with retention of the myoepithelial stroma but depletion of thymic lymphoid elements. In myasthenic patients not undergoing thymectomy, the possibility remains that persistent myoepithelial tissue can adversely affect the prognosis.

Previous studies have demonstrated that acute stress leads to the appearance of T-lymphocytes in skin and bone marrow. In order to examine the effects of adrenergic stimulation on the traffic of immune lymphocytes to the lung, C57BL/6 mice were injected with epinephrine subcutaneously, and changes in lymphocyte representation in lung, hilar lymph node, spleen, and blood were examined at 30 min. Cytofluorimetric lymphocyte subset analysis showed that epinephrine increased "memory" CD4+ lymphocytes in enzymatic digests of lung but decreased their representation in spleen. NK1.1+ lymphocytes were concomitantly increased in lung and decreased in spleen. Next, lymphocytes in spleen or popliteal lymph nodes were labeled with the Hoechst lipophilic red dye PKH26 in vivo 48 h prior to epinephrine injection. PKH26+ lymphocytes were identified in lung tissue at 30 min postepinephrine but not in controls. Lymphocyte traffic was inhibited by propanolol. We conclude that epinephrine promotes rapid shifts in immune lymphocytes from lymphoid tissues into lung and blood. Directed traffic of immune lymphocytes may be a feature of the adaptive immune response to stress at mucosal barrier surfaces.

Stress is a potential factor causing increased susceptibility of fish to pathogens. In this study, stress-induced immunological changes that may contribute to a decreased immune status were investigated. A 3 h drop in ambient water temperature of 9 degrees C was used as a relative mild and acute stress model for carp. Effects of this stressor on the dynamics of leucocyte populations were determined with specific monoclonal antibodies. The relative number of circulating B-lymphocytes in the total leucocyte population decreased significantly within 4 h after the onset of single or multiple cold shocks. This decrease was reversible, as B-lymphocyte numbers were restored within 24 h. Most probably, a redistribution of B-lymphocytes contributed to this phenomenon. In head kidney, an increase was measured in the relative number of B-lymphocytes. Granulocyte numbers showed opposite reactions: the percentage of granulocytes in the total leucocyte population nearly doubled in circulation and decreased significantly in the head kidney. This demonstrates that in vivo, a mild stressor differentially alters the distribution of leucocytes. In stressed carp, the percentage of apoptotic lymphocytes in blood is significantly higher compared with the unstressed animals. B-lymphocytes as well as Ig- lymphoid cells contributed to this increased apoptosis. Labelling of blood lymphocytes with a polyclonal antiserum against the glucocorticoid receptor also showed, besides B-lymphocytes, part of the Ig- lymphoid cell population to be glucocorticoid receptor positive. As the distribution of B-lymphocytes was substantially affected, the effect of temperature stress on T-lymphocyte-independent (trinitrophenyl-lipopolysaccharide) and T-lymphocyte-dependent (dinitrophenyl-keyhole limpet hemocyanin) humoral antibody responses was determined. Kinetics of the primary antibody response to the T-lymphocyte-independent antigen showed lower antibody titres in stressed carp during the onset of the immune response, implying a slower development of the antibody response against the T-lymphocyte-independent antigen.

The number of common lymphoid progenitors (CLP) and their pre-pro-B and pro-B cell progeny is reduced in old mice, but the age-related changes responsible for these declines have not been fully elucidated. The aim of this study was to provide additional insights into the impact of senescence on early B cell development by analyzing the CLP and pro-B cell compartments under steady-state conditions and after cytoablation with 5-fluorouracil. 5-Fluorouracil subjects the hemopoietic system to acute stress and has the advantage of revealing defects in progenitors that may otherwise be subtle. The data demonstrate significant, age-related defects in the proliferative potential of early B cell precursors and suggest that the ability of CLP to differentiate into pre-pro-B cells is also compromised by senescence. These age-related changes in early B lymphopoiesis do not result from a general defect in HSC or the bone marrow microenvironment that impairs development in all hemopoietic lineages. Instead, data demonstrating that myeloid progenitor number and developmental potential do not decline with age indicate that B lymphopoiesis is particularly sensitive to defects that accumulate during senescence.

Influences of psychological stress on the acquired immune system have not consequently been investigated. We found acute psychological stress to cause an increase in CD56+ and CCR5+ effector T cells in the peripheral blood of healthy human subjects (N=22), while skin-homing CLA+ T cells decreased. At the same time, we observed a stress-induced decrease in CD45RA+/CCR7+ naive and CD45RA-/CCR7+ central memory T cells, while CD45RA-/CCR7- effector memory and CD45RA+/CCR7- terminally differentiated T cells increased. This T cell redistribution translated into an increase in T cells expressing perforin/granzyme B and in Epstein-Barr virus-specific, cytomegalovirus-specific and influenza virus-specific CD8+ T cells. Thus, acute stress seems to promote the retention of less mature T cells within lymphoid tissue or skin while effector-type T cells are mobilized into the blood in order to be able to rapidly migrate into peripheral tissues.

We studied the effect of interleukin-1beta on functional activity of lymphoid structures in the gastrointestinal tract of rats with various behavioral parameters during stress of simultaneous immobilization and electrocutaneous stimulation. Morphofunctional characteristics of lymphoid tissue were estimated by studying elimination of intraperitoneally injected Chinese ink particles into the mesenteric lymph nodes and wall of the jejunum. Intraperitoneal injection of interleukin-1beta (5 mug/kg, 10(8) U/mg) was accompanied by accumulation of Chinese ink in the mesenteric lymph nodes of unstressed passive and active rats. The observed changes reflect an immunostimulatory effect of this cytokine. Acute stress was followed by an increase in the number of ink particles in the mesenteric lymph nodes and wall of the jejunum in behaviorally active rats. Under these conditions, the number of ink particles was elevated only in the mesenteric lymph nodes of passive specimens. As differentiated from passive animals, pretreatment of active rats with interleukin-1beta before acute stress was followed by the increased elimination of Chinese ink (antigenic material) from the abdominal cavity to the lymph nodes and through the wall of the jejunum. These data illustrate specific features of immune mechanisms for the stress response in mammals with various behavioral characteristics.
