The individual and combined antistress effects of the fruit of Schizandra chinensis and the radix of Scutellaria baicalensis were evaluated using a mouse
acute stress
model. Stress consisted of immobilization and electric foot shocks over 5 days. Mice were treated with herbal extracts for 7 days before exposing the animals to stress. Before each stressor presentation, the mice were treated with each herbal extract. Reduced locomotor activity and the percentage of time spent in the open arms of an elevated plus-maze under stress were recovered by treatment with the extract containing equal amounts of S. chinensis and S. baicalensis (CB11) at 200 and 400 mg/kg (p < 0.05). The effects of CB11 were greater than the effects of S. chinensis or S. baicalensis alone. CB11 treatment (100, 200 and 400 mg/kg) significantly reduced serum corticosterone levels (p < 0.05).
Spleen
size and the serum interleukin-2 level decreases induced by stress were prevented by CB11 (200 mg/kg) (p < 0.05). Taken together, these results suggest that S. chinensis and S. baicalensis in equal amounts could be used to treat stress disorders, in part, by preventing corticosterone and IL-2 level changes and ameliorating stress-related behavior parameters.

It has been demonstrated that a short-duration stress (
acute stress
) may result in immunopreparatory or immunoenhancing physiological conditions. The aim of the present study was to investigate whether exposure to prenatal restraint stress (PRS) influences the impact of
acute stress
on the T-cell response in the adult life. We found that female mice exposed to PRS (PS mice) did not exhibit changes in the T-cell-dependent IgG antibody production with respect to prenatally non-stressed mice (no-PS mice). However, no-PS mice exposed to
acute stress
showed an increase of antibody production after antigen stimulation. In contrast, PS mice exhibited a decreased response after an acute situation.
Spleen
catecholamines and plasma corticosterone levels were increased in
acute stress
in both PS and no-PS mice. Nevertheless, lymphocyte response to hormones was altered in PS mice. Particularly, inhibitory effect of corticosterone was higher on lymphocytes from PS mice. In addition, an increase in protein levels and mRNA expression of glucocorticoid receptor was found in lymphoid cells from PS mice. These results show that prenatal stress alters the immune intrinsic regulatory mechanism that in turn induces an increased vulnerability to any stressful situation able to modify immune homeostasis.

Psychological stress has great impacts on the immune system, particularly the leukocytes distribution. Although the impacts of
acute stress
on blood leukocytes distribution are well studied, however, it remains unclear how chronic stress affects leukocytes distribution in peripheral circulation. Furthermore, there is no report about the role of spleen in the blood leukocytes distribution induced by stress. Here we show that spleen contributes to the alteration of restraint stress induced blood leukocytes distribution. Our data confirmed that restraint stress induced anxiety-like behavior in mice. Furthermore, we found that restraint stress decreased the CD4/CD8 ratio and elevated the percentages of natural killer cells, monocytes and polymorphonuclear myeloid-derived suppressor cell. We demonstrated that activation of hypothalamic-pituitary-adrenal axis (HPA) and sympathetic nervous system (SNS) contributes to restraint stress induced alteration of blood leukocyte distribution. Interestingly, we found that splenectomy could reverse the change of CD4/CD8 ratio induced by restraint stress. Together, our findings suggest that activation of HPA axis and SNS was responsible for the blood leukocyte subsets changes induced by restraint stress.
