The analysis of 36 human skeletons (eight subadults, 13 males, 15 females) recovered during a cemetery relocation near Charleston, SC, provides data on health and disease for a 19th-century sample of Afro-Americans. The majority of the burials date from 1840-1870. Skeletal analysis verified some historical interpretations. Gender differential in mortality is evident with average age at death for males 35 and females 40 years. Females, besides living longer, had more missing and carious teeth but fewer abscesses. Both genders expressed childhood metabolic stress as indicated by linear enamel hypoplasias. Males, however, had a higher incidence (92%) than did females (70%). Age at occurrence was more widely distributed for females, but ages 2-4 were most critical for both genders. Postcranial indications of recovery from acute stress, Harris lines, occurred more frequently for males (45%) than for females (18%). Anemia, probably both genetic and acquired, was a significant health problem for both genders. Cribra orbitalia appeared in 35% of the adult crania, and 80% of the subadults had orbital lesions. Diplotic expansion of the cranial vault and infection were relatively common in the sample. Skeletal reaction to infections appeared in 69% of the males, 60% of the females, and 80% of the subadults. Skeletal changes associated with demanding physical labor were ubiquitous. The shoulder and hip were especially affected by degenerative changes, the cervical vertebrae frequently expressed osteophytosis, and males show a preponderance of Schmorl herniations and hypertrophy of the ulnar supinator crest. Skeletal trace elements indicate a relatively high exposure to lead, strontium concentrations indicative of a diet high in plant foods, and relatively low zinc and copper concentrations.

Erythropoietic stress occurs under conditions of tissular hypoxia, such as anemia. Functional relationships between erythroid bone marrow (BM) proliferation, differentiation, the expression of survival and apoptotic related proteins, as well as the features of the BM microenvironment upon acute anemic stress, are not fully elucidated. To achieve this aim, CF-1 Swiss mice were injected with a single dose of 5-fluorouracil (5-FU, 150 mg/kg ip) and a multiparametric analysis was conducted for 20 days. Apoptosis (TUNEL assay), BM architecture organization (scanning electronic microscopy), proliferation (DNA assay), differentiation (clonogenic cultures), expression of survival erythroid related proteins (EPO-R, GATA-1, Bcl-xL) as well as the expression of apoptotic- related proteins (Bax, activated Caspase-3) by Western blotting, were evaluated. Experimental data showed that apoptosis, arrest of cell proliferation and disruptions of BM architecture were maximal within the first period of acute stress (1-3 days). Bax and caspase-3 overexpressions were also coincident during this acute period. Moreover, from day 5 upon drug challenge BM responds to acute stress through the EPO-EPO-R system, prompting expressions of GATA-1 and Bcl-xL. Erythroid proliferation rates and red-cell-committed progenitors enhanced in a coordinated way to restore the size and function of the red cell compartment. A second overexpression wave of active caspase-3 was noticed during stress recovery. Together, these results indicate that in response to acute stress a dramatic increase in CFU-E (erythroid colony forming units) population is concomitant with upregulation of EPO-R, GATA-1 and Bcl-xL in the BM erythroid compartment, and that these concurrent processes are crucial for acquiring proper erythroid cell functionality without delayed response to tissular hypoxia.

Using the protocol outlined in The Backbone of History: Health and Nutrition in the Western Hemisphere (BBH) (Steckel and Rose. 2002a. The backbone of history: health and nutrition in the Western Hemisphere. Cambridge: Cambridge University Press), this project compares the Mark I Health Index (MIHI) scores of the Ipiutak (n = 76; 100BCE-500CE) and Tigara (n = 298; 1200-1700CE), two samples of North American Arctic Eskimos excavated from Point Hope, Alaska. Macroscopic examination of skeletal remains for evidence of anemia, linear enamel hypoplasias (LEH), infection, trauma, dental health, and degenerative joint disease (DJD) was conducted to assess differences in health status resulting from a major economic shift at Point Hope. These data demonstrate that despite differences in settlement pattern, economic system, and dietary composition, the MIHI scores for the Ipiutak (82.1) and Tigara (84.6) are essentially equal. However, their component scores differ considerably. The Ipiutak component scores are suggestive of increased prevalence of chronic metabolic and biomechanical stresses, represented by high prevalence of nonspecific infection and high frequencies of DJD in the hip/knee, thoracic vertebrae, and wrists. The Tigara experienced more acute stress, evidenced by higher prevalence of LEH and trauma. Comparison of overall health index scores with those published in BBH shows the MIHI score for the Ipiutak and Tigara falling just above the average for sites in the Western Hemisphere, adding support to the argument that the human capacity for cultural amelioration of environmental hardships is quite significant.
