Arthrogryposis, frequently associated with cleft palate,
scoliosis
and kyphosis, has been investigated in pure and crossbred Charolais calves. During a 3-year study, a total of 76 congenitally malformed arthrogrypotic calves from 41 purebred and commercial Charolais herds was investigated. Investigations of the management, feeds,
water
and poisonous pasture plants in 6 herds reporting deformed calves did not reveal any factors which could be associated with the appearance of the defect. Analysis of pedigrees and matings which produced arthrogrypotic calves revealed that the arthrogryposis syndrome in the Charolais breed and its crosses in Canada is genetic in origin and caused by an autosomal recessive gene with complete penetrance in the homozygous state.

The prevalence of abnormalities and associated tissue selenium residues were assessed for the fish population of Belews Lake, North Carolina, and two reference lakes in 1975, 1978, 1982, and 1992. Teratogenic defects identified included lordosis, kyphosis,
scoliosis
, and head, mouth, and fin deformities. Many fish exhibited multiple malformations and some were grossly deformed and distorted in appearance. Other abnormalities observed were edema, exophthalmus, and cataracts. Whole-body tissue residues of selenium in the fishes of Belews Lake were up to 130 times those in the reference lakes and the incidence of abnormalities was some 7 to 70 times greater. Teratogenic defects increased as selenium levels rose between 1975 and 1982 and fell with declining selenium levels between 1982 and 1992 as selenium inputs into Belews Lake were curtailed. The relationship between selenium residues and prevalence of malformations approximated an exponential function (R2 = 0.881, P < 0.01; cubic model) for centrarchids over the range of 1-80 micrograms/g dry wt selenium and 0-70% deformities. This relationship could be useful in evaluating the role of teratogenic effects in warm-
water
fish populations suspected of having selenium-related reproductive failure. Unique conditions may have existed in Belews Lake which led to the high frequency and persistence of deformities in juvenile and adult fish. In other, less-contaminated locations competition and predation may eliminate malformed individuals in all but the larval life stage. Teratogenesis could be an important, but easily overlooked phenomenon contributing to fishery reproductive failure in selenium-contaminated aquatic habitats.

More than 235 patients were imaged. One hundred sixty-five of these were imaged with a 1.5-T unit and 3-D reconstruction MRI. Coronal, transverse (axial), oblique transverse, and sagittal plane T1-weighted, selected T2-weighted, and fast spine echo pulse sequences were obtained, 4- to 5-mm slice thickness, 40 to 45 cm full field of view, 512 x 256 matrix and 2 NEX. Compromise of the neurovascular supply seemed to be one etiology that could be demonstrated. The clinical history, technique, and anatomic bilateral brachial plexus imaging is stressed to improve patient care. The cervical rib is one of the compromising brachial plexopathies selected for this presentation.

Tadpoles of Rana perezi were kept for 14 weeks in
water
containing two sublethal levels of the carbamate insecticide ZZ-Aphox or the organophosphate Folidol. Approximate concentrations of their active ingredients were 0.25 and 1 mg/L. Resulting malformations were studied by skeletal analysis and histological and histochemical investigation of te rear limbs of the tadpoles. The pesticides caused the animals to have malformations of the spinal column (
scoliosis
) and/or limbs (short and thick long bones with the epiphyses grossly twisted). Histochemical study showed differences in the composition of the connective matrix, and microscopic examination of the long bones indicated alterations in the thickness of the uncalcified bone matrix (osteoid) and the presence of abundant vascularised connective tissue in the region of the periosteum. The results confirmed changes in the composition of the connective tissue matrix as the cause of the defects observed in bone formation which are also discussed in relation to vitamin D absorption and calcium homeostasis.

An Hologic QDR 1000-W dual energy X-ray absorptiometer (DXA) was used to make a densitometric evaluation and to optimize ex vivo scanning and analysis conditions for excised sheep bones. Articulated lumbar spines and femora, removed at autopsy from eight 1-2-year-old cross-bred merino ewes, were studied using standard lumbar spine and hip protocols (version 4.20). Spine data were analysed with the ultra-high resolution (UHR) protocol (version 4.25). Preliminary hip measurements also were made using the lumbar spine (version 4.20) and
scoliosis
protocols (version 4.25). Bone mineral density (BMD) precision for the total hip (n = 5) was 0.4% using version 4.20, and 0.8% using the
scoliosis
protocol (version 4.25) but was 1.2-3.4% for femoral sub-regions. Precision for the lumbar spine was 0.8% (version 4.20). There was considerable variation in BMD and bone mineral content (BMC) at the same site between animals. Right and left hip BMD were highly correlated (r = 0.98, P < 0.00001). Measurement variables evaluated to determine the optimal conditions included: Hologic scanning protocols, depth of

The morphology and composition of the intervertebral disc and also of the cartilage end-plate were studied in patients with idiopathic or congenital
scoliosis
. The cartilage end-plate was investigated because of its function as an epiphyseal plate in humans and the association between growth and progression of the scoliotic curve. The proteoglycan and
water
contents were reduced in both structures in specimens from scoliotic patients, particularly toward the concavity of the curve, compared with autopsy material. The distribution of some collagen types differed in tissue from scoliotic patients and autopsy tissue. Calcification of the cartilage end-plate, and sometimes of the adjacent disc, occurred in all but three scoliotic patients, whereas there was minimal calcification in the autopsy specimens. We suggest that, although these changes are probably a secondary response to altered loading in the scoliotic patients, they may be highly significant to the progression of the scoliotic curve.

Spinal braces such as the Soft Boston Orthosis (SBO) help stabilize
scoliosis
and improve sitting, positioning, and head control in individuals with cerebral palsy. However, their impact on pulmonary mechanics in this population has not been studied. We examined the effect of a Soft Boston Orthosis on the pulmonary mechanics and gas exchange in 12 children and young adults (5-23 years of age) with severe cerebral palsy. Pulmonary resistance, compliance, tidal volume, minute ventilation, work of breathing, oxygen saturation, and end-tidal CO2 tension were measured with the subjects seated both with and without the orthosis and in the supine position without the orthosis. There were no significant differences in the measured parameters when comparing subjects with and without their orthoses in the sitting or in the supine position. As would be expected in individuals with severe cerebral palsy, pulmonary resistance was increased (7.33 cm
H2O
/L/s) and compliance was decreased (0.12 L/cm
H2O
) compared to reported normal values. Work of breathing was greatest in the sitting position without the orthosis (1.2 dynes/cm), suggesting that the improved positioning achieved with the orthosis may decrease the work of breathing. We conclude that the application of a Soft Boston Orthosis does not impact negatively on pulmonary mechanics and gas exchange in young people with severe cerebral palsy.

Bipyridylium herbicides produce embryotoxic and teratogenic effects in dipteran, amphibian, avian, and mammalian organisms. Diquat dibromide, a bipyridylium compound, is commonly used as an aquatic herbicide. Mallard (Anas platyrhynchos) eggs were exposed to diquat by immersing the eggs for 10 s in solutions of 0.88, 3.5, 7, 14, or 56 g/L on either d 4 or 21 of incubation. Application of diquat on d 4 yielded an estimated LC50 of 19.5 g/L through 18 d of incubation, and 9.6 g/L through hatching. Body and organ weights, and bone lengths of hatchlings did not differ between control and treatment groups with the exception of a slight increase in brain weight in the 14 g/L group. Malformations in diquat-treated embryos included defects of the brain, eye, bill, limb, and pelvis; skeletal
scoliosis
; and incomplete ossification. Subcutaneous edema was also present. Significant manifestations of oxidative stress were apparent in hatchlings and included increased hepatic thiobarbituric acid-reactive substances (TBARS) (lipid peroxidation) and decreased brain reduced glutathione (GSH). Brain protein-bound sulfhydryls (PBSH) increased. Diquat applied on d 21 of incubation yielded an estimated LC50 of 12.6 g/L through hatching. Exposure at this late stage of development did not produce deformities. Body and organ weights and, bone lengths of hatchlings did not differ between control and treatment groups. Significant manifestations of oxidative stress in hatchlings included decreased brain GSH, increased oxidized glutathione (GSSG), and ratio of GSSG to GSH. depth of the application area, would probably have little impact on mallard embryos. However, concentrations applied above ground to weeds and cattails along ditches could adversely affect the survival and development of mallard embryos, and presumably other avian species nesting in such habitats.

We have conducted studies with juvenile yellow perch (Perca flavescens) over a period of 20 weeks to address the question of the interaction between
water
- and lipid-soluble antioxidant vitamins. Fish (2.25+/-0.14 g) were divided into twelve groups, and triplicate groups were fed one of four casein-based, semi-purified diets formulated to contain low or high vitamin E levels of either 5 or 160 mg/kg without or with vitamin C supplementation (250 mg/kg). Diets were designated as -C-E, -C+E, +C-E, or +C+E, respectively. The fish fed the +C+E diet showed significantly higher weight gain, feed intake, and feed efficiency than the groups fed vitamin C-deficient diets. Total ascorbate concentrations of liver were significantly higher in fish fed vitamin C-supplemented diets than in fish fed the vitamin C-deficient diet after 16 and 20 weeks. The liver alpha-tocopherol concentrations were increased by supplemental vitamin C in vitamin E-deficient dietary groups which indicates a sparing or regenerating effect of vitamin C on vitamin E. Fish fed vitamin C-deficient diets (-C-E and -C+E) exhibited severe deficiency symptoms, such as

, lens cataracts, anorexia, and haemorrhages. The cumulative mortality was significantly higher in the -C-E groups. The thiobarbituric acid-reactive substances value was significantly higher in blood plasma of fish fed a diet unsupplemented with both vitamins. The findings in the present study with yellow perch support the hypothesis that vitamin C regenerates and/or spares vitamin E in vivo.

To evaluate the use of morphological abnormalities for standard testing of endocrine-disrupting substances (EDS), we tested the hypothesis that developmental abnormalities are a sensitive indicator of exposure to waterborne estrogenic and antiestrogenic EDS during embryonic, larval, and juvenile stages in the common estuarine killifish, the mummichog (Pisces: Cyprinodontidae). Static exposures with daily renewal were carried out with 10 to 10,000 ng/L of the estrogen agonist 17alpha-ethynylestradiol (EE2) or antagonist ZM189,154 (ZM) for the first 25 or 60 d of life. Incidence of skeletal abnormalities (
scoliosis
, lordosis, head, craniofacial, jaw, fin) and soft tissue abnormality (anal swelling) were significantly increased by EE2 but only at high concentrations (1,000 or 10,000 ng/L). Sixty-day exposure produced more severe abnormalities than 25-d exposure and in a higher proportion of fish. Within the longer exposure, 10,000 ng/L EE2 produced more abnormal fish than 1,000 ng/L (100% vs 51.6%) and more abnormalities per abnormal fish (5.73 vs 1.47). Fish reared to 12 months in clean
water
after exposure for 60 d to 10,000 ng/L EE2 survived at a lower rate than controls, retained abnormalities with the exception of anal swelling and, like fish exposed to other concentrations of EE2 and ZM, showed increased weight at length at 6 and 12 months. Sixty-day exposure to ZM increased the incidence of
scoliosis
(1,000 ng/L) but decreased the overall incidences of abnormal fish and lordosis (10 and 10,000 ng/L). No impacts of EE2 or ZM were observed before hatch, and clearing and staining of larvae demonstrated that expression of vertebral abnormalities coincided temporally with ossification. We conclude that morphological abnormalities in mummichogs are not a sensitive indicator of exposure to estrogenic or antiestrogenic waterborne EDSs at environmentally relevant concentrations.
