People who refuse to eat meat animal products mostly adhere to vegetarianism, veganism, crudivorism or macrobiotism, But these food habits are only one part of life-style chosen for spiritual, ethic or hygienic and healthy motivations. Except vitamin B12 deficiencies these regimens do not produce other deficiencies if they are correctly followed and if the energy intake is in agreement with the RDA'S. They reduce the risks of metabolic diseases, coronaropathies, arterial hypertension, colon cancer, diverticular disease of the colon, kidney and gallstones. Nevertheless crudivorism and macrobiotism are associated with high risks of deficiencies especially in children and pregnant women.

The 10th edition of the RDA discusses carbohydrates in the context of dietary fiber and digestible carbohydrates, which provide energy, and recommends that more than half the energy requirement beyond infancy be provided by carbohydrates and that a desirable intake of fiber be achieved by consumption of fruits, vegetables, legumes, and whole-grain cereals. The recommendations on digestible carbohydrates are primarily based on the desirability of limiting the intake of fat. An additional consideration is the suggestion that excess calories from carbohydrate vs. fat are metabolized and stored with different efficiencies. The scientific basis for recommending an appropriate intake of dietary fiber is not clearly evident in the current RDA. The association between a high plant food, and thus fiber intake, and lower incidence of cardiovascular disease, colon cancer, and diabetes is of interest but difficult to use to form the basis of specific recommendations for an appropriate amount of fiber to include in the diet. The importance of fiber for normal gastrointestinal function provides the best scientific basis for developing recommended intake levels. Dietary fiber, as well as non-digested starch, are the only known dietary components that increase stool weight and are the primary substrates for the microflora in the large intestine. Metabolism of this microflora, including production of short chain fatty acids that are used by epithelial cells, appears to be important for intestinal health. However, the challenge we still face is to define the parameters of gastrointestinal function that are compatible with health.

Deficiencies of the vitamins B12, B6, C, E, folate, or niacin, or of iron or zinc mimic radiation in damaging DNA by causing single- and double-strand breaks, oxidative lesions, or both. The percentage of the population of the United States that has a low intake (< 50% of the RDA) for each of these eight micronutrients ranges from 2% to 20+ percent. A level of folate deficiency causing chromosome breaks occurred in approximately 10% of the population of the United States, and in a much higher percentage of the poor. Folate deficiency causes extensive incorporation of uracil into human DNA (4 million/cell), leading to chromosomal breaks. This mechanism is the likely cause of the increased colon cancer risk associated with low folate intake. Some evidence, and mechanistic considerations, suggest that vitamin B12 and B6 deficiencies also cause high uracil and chromosome breaks. Micronutrient deficiency may explain, in good part, why the quarter of the population that eats the fewest fruits and vegetables (five portions a day is advised) has about double the cancer rate for most types of cancer when compared to the quarter with the highest intake. Eighty percent of American children and adolescents and 68% of adults do not eat five portions a day. Common micronutrient deficiencies are likely to damage DNA by the same mechanism as radiation and many chemicals, appear to be orders of magnitude more important, and should be compared for perspective. cDNA representational difference analysis (cDNA RDA) is a PCR-based subtractive enrichment procedure for the cloning of differentially expressed genes. In this study, we have further developed the procedure to take advantage of solid-phase technology, and to facilitate the use of RDA when starting material is limited. Several parameters of the PCR-based generation of cDNA representations were investigated, and a solid-phase based purification step was introduced to simplify removal of digested adapter-ends and uncleaved fragments. The use of magnetic particles increased the speed of the method, and also eliminated the risk of carry-over contamination between iterative steps of subtraction and PCR amplification. The modified protocol was evaluated in monitoring differences in gene expression in (i) a rat system consisting of livers with and without growth hormone treatment, and in (ii) a human system consisting of normal colon and colon cancer.

Several studies have demonstrated low dietary fiber intake in elderly people, which increases the risk of diseases such as constipation, colon cancer and diverticulosis. A spaghetti formula enriched with lupin fibre was developed to increase the dietary fibre intake in elderly people, as spaghetti are frequently consumed in this age group. Sweet lupin bran (Vitafiber) was used as fibre source and gluten was used as improving additive. Response surface methodology with a two variable composite rotatable design was applied to optimize the formulations. The independent variables were lupin bran (7.14-14.29%) and gluten Vital (0.1-2.0%). The dependent variables were the responses of a trained 10-member sensory panel who evaluated the sensory quality parameters color, shape, aroma, flavor and texture by the Karlsruhe 9-point test. The optimized formula was prepared with 66.7% semoline, 7.14% lupin bran, 1.05% gluten and 24.7% water, enriched with 0.019% of a vitamin premix (A. E, D, B2, B12 and folic acid) and with 0.41% of a mineral premix (Ca, Fe, Zn), in order to meet 30% of the RDA for the elderly per 100 g dry spaghetti. The dietary fibre content of the optimized product was 11.05 g/100 g. The study showed that fibre-enriched spaghetti formula is a good way to increase dietary fibre intake in elderly people, as it is a common food, simple to prepare and easy to eat.

Although exposure to ultraviolet light is often viewed as pathogenic owing to its role in the genesis of skin cancer and skin aging, there is growing epidemiological evidence that such exposure may decrease risk for a number of more serious cancers, may have a favorable impact on blood pressure and vascular health, and may help to prevent certain autoimmune disorders - in addition to its well-known influence on bone density. Most likely, these health benefits are reflective of improved vitamin D status. Increased synthesis or intake of vitamin D can be expected to down-regulate parathyroid hormone (PTH), and to increase autocrine synthesis of its active metabolite calcitriol in certain tissues; these effects, in turn, may impact cancer risk, vascular health, immune regulation, and bone density through a variety of mechanisms. Presumably, a truly adequate supplemental intake of vitamin D - manyfold higher than the grossly inadequate current RDA - could replicate the benefits of optimal UV exposure, without however damaging the skin. Diets moderately low in bioavailable phosphate - like many vegan diets - might be expected to have a complementary impact on disease risks, inasmuch as serum phosphate suppresses renal calcitriol synthesis while up-regulating that of PTH. A proviso is that the impact of dietary phosphorus on bone health is more equivocal than that of vitamin D. Increased intakes of calcium, on the other hand, down-regulate the production of both PTH and calcitriol - the latter effect may explain why the impact of dietary calcium on cancer risk (excepting colon cancer), hypertension, and autoimmunity is not clearly positive. An overview suggests that a vegan diet supplemented with high-dose vitamin D should increase both systemic and autocrine calcitriol production while suppressing PTH secretion, and thus should represent a highly effective way to achieve the wide-ranging health protection conferred by optimal UV exposure.
