Cocrynebacterium parvum (CP) was injected around the tumor and tumor cells were inoculated subcutaneously with activated peritoneal macrophages. The effects on the growth and metastases of Lewis
lung carcinoma
in C57BL/6 mice were observed. Experimental results indicated that CP had significant inhibitory effect on the tumor growth and lung metastases. The cytotoxic effects of activated peritoneal macrophages in mice on target tumor cells (L5178Y) in vitro were studied with the technique of 3H-TdR assay. The proliferation of tumor cells was markedly inhibited by the activated peritoneal macrophages of CP. Experiments showed that the antitumor mechanism of CP was the mediation via activated cytotoxic macrophages. These results indicated that CP is an effective immunostimulant. These findings will provide a scientific basis for the clinical treatment of the primary and metastatic

We studied the fine features of stromal elements in 33 cases of human
lung carcinoma
by electron microscopy. Cancer cells showed the damages to the basement membrane and matrix. On the other hand, stromal elements showed changes responding to the invasion of cancer cells. Lymphocytes and macrophages showed the activity of killing cancer cells. Most cell directly contacts the myofibroblast and fibroblast. Matured myofibroblast, early myofibroblast, active fibroblast and resting fibroblast were present near the nest of carcinoma. We discussed the roles and significance of stromal reactions.

Primary pulmonary cryptococcal granuloma is not common in Sichuan. The diagnosis of this disease is difficult to make because the patient has no characteristic symptoms and the chest X-ray findings of the mass are not easily differentiated from
carcinoma of the lung
. The incidence of this disease is apparently increasing. Pulmonary cryptococcosis may be disseminated hematogenously to the meninges and cryptococcal meningitis is very difficult to treat. If the pulmonary lesion is localized, the patient's general condition is good with no evidence of systemic lupus erythematosis, diabetes, leukemia or lymphoma, partial resection of the lung is indicated. But, if the patient has a history of recent cryptococcal meningitis, surgery must be deferred. Four cases of primary pulmonary cryptococcal granuloma have been treated surgically supplemented with medical therapy in the First Affiliated Hospital from 1986 to 1987. Follow-up of more than one year showed good results in each case.

Epidemiological reports have indicated that occupational exposure to asbestos and smoking may greatly increase the incidence of
lung carcinoma
. In this study rats were injected intratracheally with crocidolite or benzo(a)pyrene [B(a)P] only, and crocidolite plus B(a)P in combination. The incidence of lung carcinomas in crocidolite group was 6.4% (3/47), in B(a)P group 10.4% (5/48) and crocidolite plus B(a)P group 46.3% (25/54). Besides, the survival curve and the mean survival time of rats with carcinoma dropped distinctly. Significant difference was shown among the crocidolite plus B(a)P group, crocidolite and B(a)P groups (P < 0.005). The result indicated the synergetic effect of crocidolite and B(a)P on the development of
lung carcinoma
in rats and the importance of giving up smoking in the people exposed to asbestos. A model of
lung carcinoma
in rat was successfully prepared for further study in experiment on treatment, prevention and mechanism.

Molecular weight of human hepatocellular carcinoma (HCC)-associated antigen-HL2 antigen was detected to be about 50,000 dalton by SDS-PAGE and Western blot. One hundred and twenty-seven paraffin sections of cancers and pathological liver tissue were tests by ABC immunohistochemical staining. MAb HL2 reacted with 62.7% HCC and also with 8 of the 10 stomach carcinomas but only with a few other digestive system carcinomas (pancreas carcinoma, rectum carcinoma and duodenum carcinoma) and hepatitis. MAb HL2 was negative in hepatocirrhosis and other tumors (vesica carcinoma, skin carcinoma, breast carcinoma, neurogliocytoma and
carcinoma of the lung
). The results suggest that MAb HL2 has values in diagnosis of HCC and differential diagnosis between HCC and other tumours. HL2 antigen might be a new tumor-associated antigen (TAA). Further study of HL2 antigen will significantly show the actions of TAA in the occurrence and development of tumor.

A novel inhibitor of angiogenesis named SCAIF80 (shark cartilage-derived angiogenesis inhibitory factor) from shark cartilage has been isolated and characterized. SDS-PAGE analysis followed by silver staining revealed a single band with molecular weight (M(r)) of 80 kD. To determine whether this protein was capable of inhibiting angiogensis, it was assayed in endothelial cell (EC) proliferation and migration assay. The results showed that SCAIF80 significantly suppressed EC proliferation and migration in a dose-dependent manner. In the chick chorioallantoic membrane (CAM) assay, SCAIF80 also showed a potent inhibitory activity on angiogenesis in vivo. In animal tests, the growth of tumor was potently suppressed by SCAIF80 therapy. Lewis

was inhibited by 93.83 % at a dose of 5 mg/(kg.d). These findings suggest that shark cartilage may produce a novel protein with anti-angiogenic and anti-tumor activity.

lung carcinoma
cells A-549 was studied. The expression of PKC-alpha PKC-betaII PKC-gamma PKC-delta and PKC-epsilon in A-549 cells was examined. No detectable PKC-zeta was observed. Short-term treatment of cells with PMA led to the translocation of these PKC isozymes, to different extent, from cytosol to cell membrane. Whereas, prolonged treatment of cells with PMA pronouncedly reduced the levels of PKC-alpha PKC-gamma PKC-delta and PKC-epsilon, but the intracellular level of PKC-betaII was not affected. Furthermore, PMA was observed to have differential effects on the down-regulation of PKC isozymes located in the cytosol and of those located in the membrane. Prolonged PMA treatment caused extensive decrease in the levels of cytosolic PKC-delta and PKC-gamma, and depleted cytosolic PKC-alpha and PKC-betaII. However, the amount levels of membrane PKC-alpha PKC-betaII PKC-gamma PKC-delta isozymes were not decreased. In contrast, PKC-epsilon in both cytosol and membrane fraction was obviously down-regulated by prolonged PMA treatment. This study provided novel evidence on the PMA-mediated activation and down-regulation of different PKC isozymes, which might be helpful in deepening our understanding on the roles of PKC activation and the alterations of their intracellular levels in processes of chemical carcinogenesis.

The relationship between PTEN expression and anoikis in 8 different human
lung carcinoma
cell (HLCC) lines, and PKB and FAK effects in the process were studied. Northern blots and Western blots were carried out to analyze PTEN expression at mRNA and protein levels and the phosphorylation level of PKB /FAK in HLCC lines, and anoikis was examined by using DNA Ladder analysis. It was shown that PTEN mRNA expression was at similar level in 8 HLCC lines. But protein expression did not accord with it's mRNA expression. There were no PTEN protein detectable in 95C 95D and A(1) cells; very low PTEN protein was expressed in A(549) and A(4) A(7) cells; and high PTEN protein expressed in H(460) cells. There were no deletion and point mutation in 95D and H(460) cells; and mRNA stability in 95D cells was much lower than that of H(460) cells. Anoikis was observed in PTEN high-expressed H(460) cells with serum lack and loss of adhesion, and 10% serum prevented it from anoikis. PTEN decreased the phosphorylation level of PKB and down-regulated FAK expression in HLCC. These results suggest that PTEN expression is different in 8 HLCC lines, and PTEN was involved in anoikis.

SCAIF-I, an inhibitor of angiogenesis from shark cartilage was purified to homogeneity. The 4 mol/L guanidinium chloride extract of shark cartilage was fractionally precipitated with 35%-65% acetone, then purified by Resource Q ion exchange chromatography, Sephacryl S-300 gel filtration, and reverse-phase high performance liquid chromatography. The pure inhibitor was homogeneous as a single band on a silver-stained 15% sodium dodecyl sulfate-polyacrylamide gel. SCAIF-I had an molecular weight of 18 kD. It specifically inhibited proliferation of endothelial cells, and strongly blocked endothelial cell movement and angiogenesis in the chorioallantoic membrane of chick embryos. Systemic administration of SCAIF-I at the dose of 5 mg/kg.d suppressed 87.93% of the growth of Lewis
lung carcinoma
implanted in C57BL/6 mice.
