Two certified blood pressure technicians and a computerized blood pressure monitor from Computerized Screening, Inc., model 2000 (CSI 2000) obtained 900 blood pressure (BP) readings from 100 subjects aged 15 to 85 years (mean = 44.1 +/- 1.8 SEM). Measurements were obtained simultaneously on the same limb by the observers and the CSI 2000 three times per subject within a 30-minute period). Pressures obtained from the CSI 2000, which uses a piezoelectric crystal microphone interfaced through a newly designed audio amplifier board, were compared with the results of indirect cuff mercury sphygmomanometry. Correlations between the CSI 2000 readings and simultaneous readings obtained by two certified BP technicians for systolic and diastolic BP were r = 0.96 (p less than 0.001) and r = 0.91 (p less than 0.001), respectively. For systolic BP readings, the estimating equation (least-squares) based on a linear relationship between measurements from the test and reference methods was: CSI 2000 systolic BP = -3.454 + [1.015 x (observers' systolic BP)]. For diastolic BP readings, the estimating equation (least squares) was: CSI 2000 = 5.997 + [0.906 x (observers' diastolic BP)]. Close agreement was found between the readings obtained by the CSI 2000 and the average of those obtained by the two certified BP technicians.

Cartilage sulfation (somatomedin) inhibitors (CSI) from rat liver produce reversible inhibition of cartilage growth. After gel filtration Sephadex G-200, CSI appear to have MW approximately 100,000 and they are urea- and trypsin-labile factors. To explore further the mechanism of CSI action, we used the chick pelvic rudiment bioassay and studied the effect of CSI on the incorporation on 35S-sulfate (proteoglycan synthesis), 14C-leucine (protein synthesis), 3H-uridine (RNA synthesis), and 3H-thymidine (DNA synthesis). Normal rat serum (NRS) significantly stimulated the incorporation of all four isotopes, as expected. After a 24-hour incubation, CSI significantly blunted cartilage stimulation by NRS regarding total isotope uptake (1), 35S-sulfate (NRS, 96 +/- 8 mcg/100 mg cartilage dry weight; NRS + CSI, 48 +/- 4, mean +/- SEM, n = 29, P less than .05); and (2) 14C-leucine (NRS, 2,089 +/- 172 cpm/mg dry weight; NRS + CSI, 1,102 +/- 141, n = 18, P less than .05); and (3) 3H-uridine (NRS, 6,711 +/- 832 cpm/mg; NRS + CSI, 3,227 +/- 425 cpm/mg, n = 18, P less than .05); but not (4) 3H-thymidine (NRS, 3,540 +/- 620 cpm/mg; NRS + CSI 3,249 +/- 285, n = 19). The inhibition of 35S-sulfate and 14C-leucine uptake by CSI was dose-dependent and reversible. For 35S-sulfate, uptake by cartilage incubated with CSI alone for 40 hours was 13 +/- 3 micrograms/100 mg; with CSI for 16 hours then fresh medium with NRS for 24 hours uptake was 39 +/- 12, P less than .05.(ABSTRACT TRUNCATED AT 250 WORDS)

The inter-mixed micellar/vesicular (non-phospholipid-associated) bile salt concentration (IMC) can be rapidly measured in model biles by centrifugal ultrafiltration, thus allowing reliable separation of vesicular and micellar cholesterol carriers by gel filtration with an elution buffer containing bile salts at the correct IMC (Donovan, J. M., and A. A. Jackson. 1993. J. Lipid Res. 34: 1121-1129). We adapted this method to the more complex human gallbladder bile and examined the relationship between cholesterol solubilization and crystallization in gallbladder biles from 10 cholesterol gallstone patients. The IMC (mean +/- SEM) was 9.67 +/- 1.97 (range 3.56-35.02) mM with significant enrichment with hydrophilic bile salt species. Upon gel filtration of these biles with an eluant buffer containing 10 major bile salts at concentrations according to their IMC, cholesterol was found to be solubilized mainly in mixed micelles. Vesicles were detected in all 10 biles after separation by KBr density gradient ultracentrifugation but in only 5 of these biles with the IMC method. Biles without vesicles had a lower CSI (1.15 +/- 0.12 vs. 1.90 +/- 0.28, P < 0.05), a higher total lipid concentration (11.9 +/- 2.3 vs. 5.9 +/- 1.1, P < 0.05), and a higher bile salt/ (bile salt + phospholipid) ratio (0.83 +/- 0.01 vs. 0.74 +/- 0.04, P = 0.07). For both IMC and ultracentrifugation methods, vesicular cholesterol concentration showed a negative correlation with crystal observation time and a positive correlation with cumulative crystal score during 21 days. Our data indicate that methods such as density gradient ultracentrifugation overestimate vesicular cholesterol solubilization in human biles.

Fifteen percent loss in the peak area of choline containing compounds (Cho) was recently observed in 1H MR spectra of contrast-enhancing tumor at 5-10 min after Gd-contrast administration [Magn. Reson. Med. 37:222-225, 1997]. In this study, chemical shift imaging (CSI, 1500/135 ms PRESS) was used to assess the spectral changes in 47 Gd-enhancing glial brain tumors and metastatic brain tumors measured at 0-5, 5-10, and/or 10-15 min after administration of Gd-contrast. Percent Cho peak area losses measured at these times, 3 +/- 3, 12 +/- 2, and 14 +/- 3 SEM, respectively, coincided with trends of line narrowing and up-field shift of the Cho peak. Significant changes in creatine and N-acetyl acetate signals were not observed. It is concluded that the Gd-induced loss of tumor Cho signal measured after 5 min, typically required for post contrast-MRI and the positioning of the CSI volume on tumor, shows little further change with time, if any.
