The reversal of vecuronium paralysis was studied in three series of anesthetized (methohexital, fentanyl, N2O/O2) informed adult patients receiving either 40 micrograms/kg neostigmine (NEO40) (n = 6), either 500 micrograms/kg edrophonium (EDRO500) (n = 6) or 1000 micrograms/kg edrophonium (EDRO1000) (n = 6). These drugs were given randomly once the adductor pollicis twitch height regained 10% of its initial value. The neuromuscular transmission recovery was assessed during 15 minutes after the antagonist administration, by recording twitch height (TH), train of four--2 Hz--every 3 minutes (
TOF
Ratio) and finally tetanic fade--50 Herz (TET50) and 100 Hz (TET100), 5 seconds duration, one minute apart--. At 15 minutes, the TH values (mean +/-
SEM
) were for EDRO500 92% +/- 7, for EDRO1000 93% +/- 3 and for NEO40 100% +/- 4 percent (n.s.). For the
TOF
Ratio, a statistical difference (p less than 0.05) was found between NEO40 86% +/- 4 and the two other groups: EDRO500 73% +/- 3, EDRO1000 69% +/- 4. For the TET50, the values were: EDRO500 93% +/- 3, EDRO1000 86% +/- 5 and NEO40 94% +/- 2 (n.s.). At 100 Hz, the values were: NEO40 61% +/- 8, EDRO500 43 +/- 151 and EDRO1000 31 +/- 12 (p less than 0.01). In conclusion, in the conditions studied, 40 micrograms/kg neostigmine restores the neuromuscular transmission of the adductor pollicis at a higher level than edrophonium 500 micrograms/kg does. Edrophonium, 1000 micrograms/kg instead of 500 micrograms/kg does not change the neuromuscular transmission recovery.

Doxacurium was administered by titrated infusion to 14 pediatric patients for 4.7-12.3 days after laryngotracheal reconstruction to produce minimum spontaneous movement and less than five posttetanic movements of the first toe after stimulation of the posterior tibial nerve. Recovery was documented by stimulation of the ulnar nerve with 2 Hz for 2 s (train-of-four [
TOF
]) at intervals of 1 min and measurement of the ratio of the fourth to the first response (
TOF
ratio) at the adductor pollicis. During spontaneous recovery, the
TOF
ratio was between 0.4 and 0.7 for 0.6-3.3 h, mean (
SEM
) 2.2 (0.31) h. The
TOF
ratio equaled 1 between 4.7 and 23.0 h, mean (
SEM
) 11.0 (2.1) h after termination of doxacurium infusion. In six of the patients, weakness and decreased coordination were noted for a few days to weeks postoperatively. There were no complications related to impairment of upper airway function or ventilation in those patients who had recovery of neuromuscular transmission to the extent of

ratio equal to 1 prior to extubation or in those patients in whom weakness or lack of coordination was noted after tracheal extubation.

An isozyme of pectinase from Aspergillus niger with polygalacturonase activity caused chitosanolysis at pH 3.5, resulting in low-molecular weight chitosan (86%), chitooligosaccharides (COs, 4.8%) and monomers (2.2%). HPLC showed the presence of COs with DP ranging from 2 to 6. Charcoal-Celite chromatography and re-N-acetylation of the COs followed by CD, IR, MALDI-

-MS and FAB-MS analyses revealed an abundance of chitobiose, chitotriose and chitotetraose. The COs-monomeric mixture showed a bactericidal effect towards Bacillus cereus and Escherichia coli more efficiently than native chitosan. Among the chitooligomers, the hexamer showed maximum antibacterial effect followed by the penta-, tetra-, tri- and dimers. Of the two monomers, only GlcN showed slight bacterial growth inhibition.

Proteins of a liver extract taken from a metabolically (13)C-labeled mouse were separated by 2D-PAGE and identified after tryptic digestion by MALDI-
TOF
MS peptide mass fingerprinting. (13)C-Labeling of proteins was achieved by an infusion of U-(13)C-glucose, which is metabolized to labeled nonessential amino acids. The labeling was analyzed using the relative isotopologue abundances of the measured isotope pattern of tryptic peptides and quantified by their increase in the average molecular mass (DeltaAVM). Fractional synthesis rates (FSR) of proteins were determined from corresponding peptides using measured DeltaAVM values as well as DeltaAVM values deduced from tRNA-precursor amino acid labeling, which in turn was derived from proteins showing high (13)C enrichments. The 8-h FSR values of 43 proteins were determined to range from 0 +/- 0.6 to 95 +/- 1%/8 h, with typical errors given as

values, which depend on the number of peptides of a specific protein usable for calculation. The method demonstrates that FSR values as an indicator for protein turnover in the liver proteome can be estimated within narrow error margins, providing baseline values from which treatment-dependent deviations could be detected with high statistical certainty.

A novel biologically relevant composite substrate has been prepared consisting of a calcium phosphate (CaP) layer formed by magnetron sputter-coating from a hydroxyapatite (HA) target onto a gold-coated silicon substrate. The CaP layer is intended to mimic tooth and bone surfaces and allows polymers used in oral care to be deposited in a procedure analogous to that used for dental surfaces. The polymer cetyl dimethicone copolyol (CDC) was deposited onto the CaP surface of the substrate by Langmuir Blodgett deposition, and the structure of the adsorbed layer was investigated by the surface specific technique of sum frequency generation (SFG) vibrational spectroscopy. The gold sublayer provides enhancement of the SFG signal arising from the polymer but plays no part in the adsorption of the polymer. The surface morphology of the substrate was investigated using
SEM
and AFM. The surface roughness was commensurate with that of the thermally evaporated gold sublayer and uniform over areas of at least 36 mum(2). The chemical composition of the CaP-coated surface was determined by FTIR and
TOF
-SIMS. It was concluded that the surface is primarily calcium phosphate present as a mixture of amorphous, non-hydroxylated phases rather than solely stoichiometric hydroxyapatite. The SFG spectra from CDC on CaP were closely similar, both in resonance wavenumbers and in their relative intensities, with spectra of thin films of CDC recorded directly on gold. Application of previous analysis of the spectra of CDC on gold therefore enabled interpretation of the polymer orientation and conformation on the CaP substrate.

Crosslinked gels (hylans) containing long-chain (MW>1 x 10(6)Da) hyaluronan (HA), a connective tissue GAG, show exceptional biocompatibility for vascular implantation but poorly interact with vascular endothelial cells (ECs). Previous studies showed in situ fragmentation of HA by UV light to bioactivate hylan gels and elicit enhanced EC responses. Since fragmented HA can be pro-inflammatory, it is important to define an optimal size distribution of HA fragments on the hylan surface that will recruit and support normally functional ECs and limit ulterior responses. Related studies have shown that exogenous models of HA do not necessarily replicate cell responses to HA scaffolds. Since scaffolds cannot be created based on fragmented HA alone, we sought to determine size-specific responses of ECs to HA substrates of defined fragment sizes by creation of HA-tethered culture surfaces. HA (1000, 200, 20 kDa) and an oligomer mixture were tethered onto an aminosilane (APTMS)-treated glass surfaces using a carbodiimide reaction. MALDI-

showed the HA digests to contain HA 4-8mers with a 75+/-0.4% w/w of 4mers. Immuno-fluorescence,
SEM
, AFM and XPS analysis revealed homogeneous amine and HA surfaces. An amine s-SDTB assay and HA fluorophore-assisted carbohydrate electrophoresis (FACE) indicated surface densities of 9+/-3 amine groups/nm(2) and 0.57+/-0.44 microg/cm(2), respectively. HA/HA fragments/oligomers were stable over 21 days of incubation in serum-free culture media. EC proliferation on these surfaces resulted was limited, a possible effect of smooth surface topography, high anionicity, and in case of 4mers, non-interaction with primary HA cell-surface receptors (CD44). This work is significant in that it allows testing of cell responses to substrates composed of single-sized fragments of HA that cannot by themselves be cross-linked into a gel. Future work in our lab will use this model to assess the effects of other HA oligomer sizes on EC behavior.
