Nine untrained women using low dosage monophasic oral contraceptives (OC) performed an intermittent treadmill test on two different occasions within one pill-cycle to determine the effect of OC on performance and some commonly used metabolic markers. The first test was performed after 5-8 days of resuming the OC agents after menstrual bleeding while the other test was performed after 19-21 days. Performance time on the final exhausting run of six intermittent high intensity 20 s runs was no different between trials [mean days 5-8: 22.3 (SEM 1.2) s vs days 19-21: 22.7 (SEM 1.1) s]. There was no difference in heart rate [peak heart rate days 5-8: 183 (SEM 3) beats.min-1 vs days 19-21: 186 (SEM 2) beats.min-1], oxygen consumption during any run [run 5 of days 5-8: 1,392 (SEM 51) ml.min-1 vs run 5 of days 19-21: 1,494 (SEM 3) ml.min-1] or in any of the metabolic variables measured at any time in venous blood [peak blood lactate concentration days 5-8: 8.4 (SEM 0.3) mmol.l-1 vs days 19-21: 8.1 (SEM 0.5) mmol.l-1; peak blood glycerol concentration days 5-8: 0.39 (SEM 0.02) mmol.l-1 vs days 19-21: 0.38 (SEM 0.02) mmol.l-1; resting free fatty acids concentration days 5-8: 0.25 (SEM 0.05) mmol.l-1 vs days 19-21: 0.29 (SEM 0.07) mmol.l-1; peak blood glucose concentration days 5-8: 6.7 (SEM 0.5) mmol.l-1 vs days 19-21: 6.6 (SEM 0.2) mmol.l-1; peak capillary blood ammonia concentration days 5-8: 139 (SEM 18.3) mumol.l-1 vs days 19-21: 170 (SEM 18.0) mumol.l-1]. These results suggest neither intermittent high intensity exercise performance nor energy metabolism change between days 5-8 and days 19-21 of a low dosage monophasic OC pill during one pill-cycle.

Crossbred wether goats (n = 24; 50% Boer, 6 per diet) initially averaging 27.4+/-0.4 kg were fed either wheat middlings (wheat midds), soybean hulls (soyhulls), or corn gluten feed at 1% BW (as-fed) along with orchardgrass hay (10.7% CP) offered to ad-libitum consumption for 72 d followed by 5 d total fecal collection. The Control (hay) diet was supplemented with 5.7% soybean meal to bring total dietary protein to 12.5%, by-products were brought to a higher Ca:P ratio with limestone or dicalcium phosphate to make total dietary Ca:P 1.5:1, and soybean meal was added to soyhulls to bring them up to 17% CP (wheat midds = 17% and corn gluten feed = 21% CP). Total DMI (916 g/d+/-57 or 3.2%+/-0.2 BW) did not differ (P > 0.92) among treatments. Initial BW (P = 0.25), final BW (P = 0.48), and ADG (P = 0.56) did not differ for the four treatments. Carcass weight was greater (P = 0.05) for goats fed soyhulls (16.0 kg) or wheat midds (15.6 kg) as compared with goats fed the hay diet (14.5 kg), with carcass weight from goats fed corn gluten feed being intermediate (15.3 kg, SEM = 0.3 kg). Carcass grade did not differ (P = 0.80) and averaged 5.42+/-0.4. Dressing percentage tended (P = 0.12) to be lower for goats fed the hay diet (46.4%) compared with soyhull (48.3%), corn gluten feed (48.3%), or wheat midd (48.8%) diets (SEM = 0.7). Ruminal pH was highest (P < 0.01) for goats fed the hay diet (6.52) and lowest for goats fed wheat midds (6.23) with soyhull (6.41) and corn gluten feed diets (6.35) being intermediate (SEM = 0.05). Digestibility of DM (70.1+/-2.5%), OM (70.3+/-2.6%,), CP (75.5+/-2.0%), GE (68.5+/-2.7%), NDF (68.1+/-3.0%), ADF (65.4+/-3.4%), cellulose (70.1+/-2.9%), and lignin (31.1+/-8.2%) did not differ (P > 0.15). Total ruminal VFA did not differ (86.0+/-6.1 mM, P = 0.59), but acetate:propionate ratio was higher (P < 0.01) for hay (3.1) and soyhull diets (3.3) than for corn gluten feed (2.4) and wheat midd diets (2.4, SEM = 0.11). Ruminal ammonia (mg/100 mL) was lower (P < 0.01) for goats fed hay (15.4) and soyhull diets (11.6) than those fed corn gluten feed (25.2) and wheat midd diets (23.0, SEM = 1.35). Ruminal pH was lower for goats fed the byproducts, but remained above 6. Serum urea nitrogen (mg/100 mL) averaged 21.0+/-1.0 (P = 0.11) with soyhulls tending to be lowest (19.3) and corn gluten feed tending to be highest (22.8). Soyhulls, corn gluten feed, and wheat midds appear to be viable feed ingredients for meat goat diets.

With the goal of shortening the postoperative healing phase and the minimization of the risk of peri-implantitis around dental implants, a polymer coating, with improved surface biocompatibility in the region of soft tissue penetration by the implant, was investigated. The polymer used was the relatively slowly resorbable poly-beta-hydroxybuterate (PHB) whose surface was further activated in a NH3--plasma. The influence of surface roughness of the substrate (commercially pure titanium) as well as the influence of the edge radii on the test samples was determined in prior studies. These studies formed the basis for an optimised coating process. In-vitro biocompatibility was determined using a human gingival primary cell culture. Surface morphology was determined with SEM and AFM to complement the cell culture studies.

Phenylalanine ammonia-lyase (PAL, EC 4.3.1.5.) activity, tanning ability, and polyphenols levels were measured in cherimoya (Annona cherimola Mill.) fruit treated with 20% CO(2) + 20% O(2) + 60% N(2) for 1, 3, or 6 days during chilling temperature (6 degrees C) storage. The residual effect of CO(2) after transfer to air was also studied. These observations were correlated with texture and cellular characteristics, visualized by cryo-SEM. Tanning ability and the early increase in tannin polyphenols induced by chilling temperature were reduced by CO(2) treatment. Conversely, high CO(2) atmosphere enhanced the nontannin polyphenol fraction as compared with fruit stored in air. Lignin accumulation and PAL activation observed in untreated fruit after prolonged storage at chilling temperature were prevented by high CO(2). Moreover, the restraining effect on lignification was less effective when the CO(2) treatment was prolonged for 6 days. In addition, fruits held at these conditions had greater firmness and the histological characterization of the separation between cells was similar to that in untreated fruits. We conclude that CO(2) treatment modulates the phenolic response that seems to regulate the strength of cell adhesion and so to prevent hardening caused by chilling temperature storage.

To identify the reserve of an individual's tolerance of the sensation of breathlessness and metabolic stress in maintaining intense intermittent exercise at exhaustion under conditions of normal breathing, the contribution of the effect of modest inspiratory load on these two responses to the change in the exercise sustainability (Ex(sus)) were examined. Seven men repeatedly performed 12 s exercise at 160% maximal aerobic power output followed by passive recovery for 18 s under normal and ventilatory muscle loaded (VML) breathing conditions until exhaustion. In the VML trial, ventilatory muscle work at exhaustion was double that of the normal control. The control Ex(sus) was reduced [mean (SEM)] [31.7 (6.6)%] while the slope of the time course for the rating of the perceived magnitude of breathing effort (RPMBE/Time), which reflected the intensity of breathlessness, was increased [164.8 (32.2)%] from control and the RPMBE at exhaustion was higher than corresponding control value [144.4 (21.8)%]. Moreover, increases in plasma ammonia and uric acid concentrations, which indicated metabolic stress, were increased [168.1 (28.0)% and 251.7 (57.4)%, respectively], with no change in total oxygen uptake from control when the control exercise was repeated with an identical duration of VML exercise. It was found that the reduction in Ex(sus) in the VML trial was correlated to the increase in their sensations of the intensity of breathlessness (RPMBE/Time: r=0.81; RPMBE at exhaustion: r=0.97, P<0.05). The reduction in Ex(sus), however, was not correlated to the increase in metabolite concentrations. These findings implied that there was no substantial reserve of tolerance of the sensation of breathlessness relative to that of metabolic stress in subjects maintaining intense intermittent exercise at exhaustion under normal conditions of breathing.

Corn stover was pretreated with aqueous ammonia in a flow-through column reactor, a process termed ammonia recycled percolation (ARP). This method was highly effective in delignifying of the biomass, reducing the lignin content by 70-85%. Most lignin removal occurred within the first 20 min of the process. Lignin removal by ARP was further confirmed by FTIR analysis and lignin staining. The ARP process solubilized 40-60% of the hemicellulose but left the cellulose intact. The solubilized carbohydrate existed in oligomeric form. Carbohydrate decomposition during the pretreatment was insignificant. Corn stover treated for 90 min exhibited enzymatic digestibility of 99% with 60 FPU/g of glucan enzyme loading, and 92.5% with 10 FPU/g of glucan. The digestibility of ARP treated corn stover was substantially higher than that of alpha-cellulose. The enzymatic digestibility was related with the removal of lignin and hemicellulose, perhaps due to increased surface area and porosity. The SEM pictures indicated that the biomass structure was deformed and its fibers exposed by the pretreatment. The crystallinity index increased with pretreatment reflecting removal of the amorphous portion of biomass. The crystalline structure of the cellulose in the biomass, however, was not changed by the ARP treatment.

Exhaled breath condensates have been widely used to detect inflammatory mediators in the fluid that covers airway surfaces of patients with inflammatory lung disorders. This approach is much less invasive than bronchoalveolar lavage, but respiratory droplets are markedly diluted by large and variable amounts of water vapor. We estimated the dilution of respiratory droplets by comparing concentrations of nonvolatile, reference indicators (total nonvolatile cations, urea or conductivity) in 18 normal subjects with normal plasma concentrations by assuming similar concentrations in the respiratory fluid and plasma. The volatile cation, NH4+ (most of which is delivered as NH3 gas from the mouth), represented 93 +/- 3% (SEM) of the condensate cations. More than 99% of the NH4+ was removed by lyophilization, making it possible to use conductivity to estimate total nonvolatile ionic concentrations and facilitating analysis of urea. Conductivity was significantly correlated with electrolyte and urea concentrations. Estimates of dilution based on total cations, conductivity, and urea were not significantly different (cations: 20,472 +/- 2,516; conductivity: 21,019 +/- 2,427; and urea: 18,818 +/- 2,402). These observations suggest that the conductivity of lyophilized samples can be used as an inexpensive, simple, and reliable method for estimating dilution of nonvolatile, hydrophilic mediators in condensates.

Collagen is often used in bioartificial livers as a biomimetic coating to promote liver cell adhesion and differentiation. Animal proteins are expensive and expose the host to risks of cross-species infection due to contamination with prions. Silk fibroin (SF) is a biocompatible protein produced by Bombyx mori silk worms and possibly an alternative to collagen. We prepared SF-collagen blend films with different SF content adherent to the bottom of standard tissue culture dishes, and characterized their surface morphology by SEM, their wettability and examined them for their capacity to support rat liver cell adhesion and metabolism. Cell metabolism was characterized by estimating the rate at which cells eliminated ammonia and synthesized urea for up to 48h of culture. SF-containing films were smooth, clear and more wettable than collagen. Cells readily adhered, formed junctions and small size aggregates on all films. As many cells adhered on SF as on collagen films. Cell adhesion to high collagen content blend films could not be reliably estimated because cells dwelt in the large cavities in the film. The effect of SF on cell metabolism differed with the investigated metabolic pathway. However, cells on SF-containing films eliminated ammonia and synthesized urea at rates generally comparable to, for urea synthesis at times higher than, that of cells on collagen. These results suggest that silk fibroin is a suitable substratum for liver cell attachment and culture, and a potential alternative to collagen as a biomimetic coating.

The effect of altering supplementation frequency on host N balance and key N transactions in the ruminal ecosystem were monitored. Four ruminally fistulated beef steers (BW = 513 kg; SEM = 6.5) were used in a 2 x 2 crossover design with two periods and two supplementation frequency treatments. Supplementation frequencies were 2 and 7 d/wk. Steers were fed tallgrass prairie hay (73.1% NDF, 5.3% CP) ad libitum. Supplement (42% CP; DM basis) was fed at 0.36% BW/d to steers supplemented 7 d/wk. Steers supplemented 2 d/wk received the same amount of supplement per week, but it was equally split among the two supplementation events. Steers supplemented 7 d/wk had higher forage (P < 0.02) and total digestible OM intake (P < 0.06), total N intake, fecal N excretion, and N retention. Although both supplementation frequencies were characterized by positive N balance, the decrease in N retention in the steers supplemented 2 d/wk was due to higher (P < 0.01) urinary N loss. Ruminal fluid was sampled at 0, 2, 4, 6, 12, 24, 48, and 72 h after supplementation beginning on a day when both treatments were supplemented. Frequency x hour interactions (P < 0.02) were observed for ruminal N metabolism criteria. Counts of peptide- and AA-fermenting bacteria peaked at 2 h and returned to nadir by 12 h for steers supplemented 7 d/wk. Steers supplemented 2 d/wk peaked at 6 h with a greater population and returned to nadir at 48 h. Ruminal ammonia concentrations followed a similar trend. Specific activity of ammonia production was lower (P < or = 0.05) immediately after supplementation for steers supplemented 2 d/wk, but by 12 h was the same as for 7 d/wk steers. Ruminal peptides and free AA peaked at 2 h for steers supplemented 2 d/wk and were generally higher (P < or = 0.05) during the first 6 h compared with steers supplemented 7 d/wk. Total VFA concentration was not different (P = 0.35) due to supplementation frequency. Frequency x hour interactions (P < 0.01) were observed for all molar proportions of VFA. The molar proportion of acetate and acetate:propionate ratio were lower (P < 0.01) and the molar proportions of propionate and butyrate were higher for steers supplemented 2 d/wk from 4 h to 24 h. In conclusion, forage use and N balance improved with supplementation 7 d/wk, but supplementation 2 d/wk was associated with some desirable shifts in select ruminal events that may contribute to moderating potential negative impacts of supplementing infrequently.

La(0.6)Sr(0.4)Co(0.2)Fe(0.8)O(3-delta)(LSCF) perovskite powders have been synthesised by solid-state reaction, co-precipitation, drip pyrolysis and citrate gel routes, and characterised using XRF, XRD, SEM and BET. Co-precipitation using oxalic acid or aqueous ammonia as precipitant failed to achieve the correct chemical composition. Perovskite structures were achieved in all other cases. Surface areas ranging from 0.6 to 17.4 m(2) g(-1) were obtained, which was reflected in the different microstructures observed. The citrate gel product exhibited a convoluted network morphology resulting in its large surface area. Thin-walled (approximately 200 microm) tubular membranes have been manufactured from the LSCF powders using viscous plastic processing. The tubes have been characterised using a custom-built gas analysis rig with on-line mass spectrometry. Porosity levels of the membranes were found to be very low (<0.1%). The spontaneous oxygen flux across the tubular membranes was determined as a function of temperature. Oxygen permeation rates ranged from 0.1 to 0.3 micromol cm(-2) s(-1) at 1273 K. The catalytic behaviour of the LSCF tubes towards methane oxidation has been studied using temperature programmed reaction and conventional catalytic measurements. The tubes favoured combustion reactions, with smaller amounts of partial oxidation and oxidative coupling products observed. Powder coatings have been incorporated to establish the effect of increasing surface area.
