A 48-year-old man with small cell lung cancer developed ARDS, and massive pulmonary edema fluid was obtained with the fiberoptic bronchoscopy. The pulmonary edema fluid to serum ratios of total protein and albumin were 0.72 and 0.85 respectively. The ratio of
LDH
was higher (2.71), while that of cholesterol was lower (0.11) than that of total protein. Simultaneously, isopropyl N [I-123] p iodoamphetamine (I-123 IMP) and I-131 human serum albumin (I-131

) were injected into this patient. Samples of blood and pulmonary edema fluid were collected to measure the clearance through the pulmonary microvasculature. The time activity curves of I-123 IMP and I-131

in his blood samples revealed almost constant radioactivity from 5 minutes to 120 minutes after injection, while both radioactivity levels in pulmonary edema fluid samples increased with time. The clearance ratio of I-123 IMP to I-131

was constant at each sampling time (mean +/- SD, 1.51 +/- 0.32). The linear correlation between I-123 IMP clearance and I-131

clearance (r = 0.95, p less than 0.01) suggested that the clearance ratio of exudative plasma components may remain unchanged even if pulmonary microvasculature permeability has changed.

Sequence homology was searched among the nine cDNAs/deduced amino acid sequences encoding for the eight fertilization-related sperm antigens: namely, lactate dehydrogenase (
LDH
-C4), galactosyltransferase (GT), SP-10, rabbit sperm autoantigen (RSA), guinea pig (g)PH-20, cleavage signal protein (CS-1),
HSA
-63, human (h)PH-20, and AgX-1, respectively. Most significant identity (> 50%) was found between
HSA
-63 and SP-10 (59.8%), and between gPH-20 and hPH-20 (61.1%); followed by identity between SP-10 and GT (34.7%); and then between AgX-1 and hPH-20 (39.4%). All others had identity < 25%. The significance of these sequence homologies among the sperm antigens in the development of an antisperm contraceptive vaccine is discussed.

Two coumarin derived alpha-dicarbonyls, HOCGO and DMACGO, are presented as pH, polarity and quencher sensitive fluorescent reporters for proteins that can be targeted at reactive arginines. Both inactivate a number of enzymes that feature functionally critical and chemically susceptible arginyls. Both are chemoselective in responding towards Arg side chain but not towards Cys or Lys side chains under suitably dilute conditions. With pKapp approximately 6.7, HOCGO can serve as a pH sensor, while with pKapp << 4.0, DMACGO is better suited as a polarity sensor. A contrasting set of changes are manifest in the CGOs upon protein interaction that are either attributable to Arg modification or to the noncovalent probe associations with hydrophobic protein domains. DMACGO probes a single hydrophobic site on ovalbumin while HOCGO is largely unresponsive to this protein. Three to five arginyls are modified in
HSA
and BSA by HOCGO as well as DMACGO, while the latter also probes two hydrophobic sites on both these proteins. HOCGO modifies a single arginine in
LDH
active site, while its adducts with H4 and M4LDH isozymes titrate to the apparent pKa of 7.8. Other proteins labeled with HOCGO or DMACGO reveal a number of variations that can furnish information about the microenvironment in the sites probed. The CGOs are thus potentially useful reporters of protein domains that feature reactive arginines. Suitable experimental condition are defined based on mechanistic considerations that may be used in applying the CGOs as Arg modifiers and as fluorescent probes.

Development of a vaccine(s) based on sperm antigens represents a promising approach for contraception. The utility of an antigen in immunocontraception is contingent upon its tissue specificity, involvement in human fertility, and immunogenicity. A number of antigens have been characterized from the sperm surface. Notable among these are
LDH
-C4, RSA antigens, PH-20, SP 1U,
HSA
-63, FA-1, FA-2 and CS-1. These antigens have been proposed as potential candidates for the development of contraceptive vaccine(s). Their current status, application, relative merits, and immunogenicity in immunocontraception are discussed in this review.

Sperm proteins are known to be associated with normal fertilization as auto- or iso-antibodies to these proteins may cause infertility. Therefore, sperm proteins have been considered to be the potential candidate for the development of antifertility vaccine. Some of the sperm proteins proved to be promising antigens for contraceptive vaccine includes lactate dehydrogenase (
LDH
-C4), protein hyaluronidase (PH-20), and Eppin. Immunization with
LDH
-C4 reduced fertility in female baboons but not in female cynomolgus macaques. Active immunization with PH-20 resulted in 100 per cent inhibition of fertility in male guinea pigs but it induced autoimmune orchitis. Immunization with Eppin elicited high antibody titres in 78 per cent of immunized monkeys and induced infertility but the immunopathological effect of immunization was not examined. Peptides NT, 1, 2 and 4 were found to mimic immunobiological activity of native protein. Passive administration of antibodies to peptides NT, 1, 2 and 4 induced infertility in male and female rats and peptide 1 was found to be most effective in suppressing fertility. Active immunization with keyhole limpet haemocynin (KLH) conjugated synthetic peptide 1 impaired fertility in all the male rabbits and six of the seven male marmosets. Some organotin(IV) carboxylates of the general formula RnSn(L)m [n=3, m=1, R=Me, Pr, Bu and Ph; n=2, m=2, R=Me, Bu and Oct; L=anion of lauric (HLA), stearic (

) and myristic acid (HMA)] have been synthesized and characterized by various spectroscopic studies. Tri- and diorganotin(IV) carboxylates adopt trigonal-bipyramidal and octahedral geometry around tin atom, respectively. They have been screened in vitro for anti-tumor activity against cancer cell lines of human origin, viz. MCF-7 (mammary), HEK-293 (kidney), PC-3 (prostate), HCT-15 (colon) and HepG-2 (liver). Enzyme assays viz. lipid peroxidase, glutathione peroxidase, glutathione reductase and total glutathione assay have been carried out to explore the cause of their cytotoxiciy. The results indicate that ROS (reactive oxygen species) generation may be responsible for their cytotoxicity but elevation in
LDH
(lactate dehydrogenase) suggests that necrosis cannot be excluded. Further, DNA (deoxyribonucleic acid) fragmentation, acridine orange and comet assay support the fact that the apoptosis is the main cause of cytotoxicity of organotin(IV) carboxylates, whereas the necrosis plays a minor role. The anti-inflammatory activity evaluation shows that the complexes possess moderate activity. Results of acute toxicity of the complexes have also been discussed.
