The efficiency of
autoantigen
fragments (Fa and Fc) to trigger a memory autoimmune response in rats primed with chemically modified rat male accessory glands (MRAG) was assayed by reimmunizing them with the fragments coupled to human serum albumin (Fa-
HSA
and Fc-
HSA
). The humoral immune response assay by ELISA and passive hemagglutination revealed that a high level of antibodies reactive with the
autoantigen
of accessory glands is triggered by Fa-
HSA
in male and by Fc-
HSA
in female rats, whereas Fc in male and Fa in female rats did not stimulate appreciable levels of antibodies. Furthermore, the specificities of male and female antibodies were directed mainly to Fa and Fc epitopes, respectively. Therefore, Fa behaved like a more immunogenic fragment for male rats and Fc for female rats.

The effect of aging on the immune response to
autoantigen
of rat male accessory glands (RAG) was studied in Wistar rats. Male and female rats, 3 and 12 months old, were immunized with chemically modified RAG and heterologous antigen (human serum albumin,
HSA
). The study of delayed type hypersensitivity (DTH) and antibodies against RAG revealed a higher response in 12-month-old animals than in 3-month-old animals (P less than 0.005), regardless of their sexes. No differences in DTH and humoral responses to
HSA
were observed. Experiments on the transfer of spleen cells showed an increase in response elicited by RAG immunization in young recipients of cells from normal or immunized old syngeneic donors. On the contrary, old recipients of spleen cells from normal or immunized young donors maintained their high response the same as non-transferred old rats. Therefore, both the lymphoid cells and the environment in which the response was elicited seem to be involved in the increase of the autoimmune response.

Sequence homology was searched among the nine cDNAs/deduced amino acid sequences encoding for the eight fertilization-related sperm antigens: namely, lactate dehydrogenase (LDH-C4), galactosyltransferase (GT), SP-10, rabbit sperm
autoantigen
(RSA), guinea pig (g)PH-20, cleavage signal protein (CS-1),
HSA
-63, human (h)PH-20, and AgX-1, respectively. Most significant identity (> 50%) was found between
HSA
-63 and SP-10 (59.8%), and between gPH-20 and hPH-20 (61.1%); followed by identity between SP-10 and GT (34.7%); and then between AgX-1 and hPH-20 (39.4%). All others had identity < 25%. The significance of these sequence homologies among the sperm antigens in the development of an antisperm contraceptive vaccine is discussed.
