A total of 56 strains of Haemophilus vaginalis were tested for their in vitro susceptibility to 21 antimicrobial agents by an agar dilution method. All strains were inhibited by 1 mug or less of penicillin, ampicillin, carbenicillin, and vancomycin per ml. The cephalosporins were less active; 4 mug of cefazolin per ml, 16 mug of cephalothin per ml, or 128 mug of cephalexin per ml was required to inhibit all strains. Kanamycin, gentamicin, tobramycin, and neomycin were relatively inactive against H. vaginalis. All strains were inhibited by 4 mug of streptomycin per ml and 2 mug of chloramphenicol per ml. Only 57% of the strains were inhibited by 4 mug of tetracycline per ml, whereas 43% were inhibited by 16 to 64 mug/ml. The combination sulfamethoxazole/trimethoprim was relatively inactive against H. vaginalis. All strains tested exhibited minimal inhibitory concentrations of >/=128 mug when tested against colistin, nalidixic acid, and sulfadiazine. Erythromycin and clindamycin were the most active of the antibiotics tested; for all strains the minimal inhibitory concentrations were </=0.06 mug/ml.

Kanamycin resistance was transferred by conjugation from a Haemophilus influenzae strain into Escherichia coli K12, and between E. coli K12 strains in subsequent transfers. Covalently closed molecules of DNA were isolated by sedimentation analysis of the DNA of the resistant E. coli K12 strains. These facts support the hypothesis that kanamycin resistance is mediated by a transferable plasmid in this strain of H. influenzae.

Sixty strains of Haemophilus influenzae were isolated from the genitourinary tracts of adults: 19 from cervicovaginal secretions, one from a woman with bartholinitis, 37 from urethral exudates, and three from urine. Non-capsulated strains were recovered predominantly, and biotype III accounted for 28 isolates and biotype IV for 25. Many of the H influenzae strains were found to be resistant to one or more of the antibiotics commonly used against sexually transmitted diseases. Resistance to tetracycline was prevalent and was found in 17 of the strains. Ten strains were ampicillin resistant and beta lactamase producing. Kanamycin resistance was less common (two strains). Trospectomycin (U-63366), a new spectinomycin analogue, was eight to 16 times more active than spectinomycin. All the quinolones tested were very active against all strains and may provide an effective alternative for the treatment of resistant H influenzae in genitourinary infections.

High-resolution two-dimensional gel electrophoresis of pulse-labeled Haemophilus influenzae extracts allows for the separation and quantification of more than five hundred protein spots. We have determined the changes in the protein synthesis patterns triggered by treatment with inhibitors of transcription, Rifampicin (Rif) and translation, Chloramphenicol (Chl), Erythromycin (Ery), Fusidate (Fus), Puromycin (Pur), Kanamycin (Kan), Streptomycin (Str), and Tetracycline (Tet) relative to the total protein synthesis rate. More than 200 spots changed in intensity under at least one condition. With the exception of the aminoglycosides, Kan and Str, all inhibitors triggered a clear increase in the synthesis rates of ribosomal proteins and RNA polymerase subunits. Northern analysis of rpoA, rpoB, rpoC, and six ribosomal protein genes indicated induction of transcription as well as antitermination as part of the mechanism of the regulation of gene expression. Total RNA synthesis was increased after exposure to Chl, Ery, Fus, and Tet, whereas Str had no effect. Rif led to an almost complete shutdown of RNA synthesis. Exposure to Chl, Ery, Fus, Rif, and Tet resulted in a decrease in the concentration of the stringent factor, guanosine 5',3'-bis-diphosphate (ppGpp) whereas Str again had no effect. Thus, as in Escherichia coli, the response of H. influenzae to translational inhibitors appears to be mediated by the regulatory nucleotide ppGpp.
