revealed it to contain one helix-loop-helix motif and one leucine zipper domain. Using yeast two-hybrid screening, five zinc-finger proteins were identified as
HCAP1
-interacting proteins. Among them, NDP52 (nuclear dot protein 52) appeared most frequently in positive clones and was the most strongly interacting protein. Then, the interaction between
HCAP1
and NDP52 was confirmed by GST pull-down assay and a coimmunoprecipitation experiment. Moreover, an immunofluorescent staining assay indicated that NDP52 colocalizes with
HCAP1
in the cytoplasm. By deletion analysis, the leucine zipper domain of
HCAP1
and the zinc finger domain of NDP52 were identified as important regions responsible for the interaction.

gene products on cell proliferation of T lymphoma Jurkat cell line,

gene! was transfected into Jurkat cells mediated by liposome, and the cells stably expressing exogenous

were screened with G418. The effects of
HCAP1
products on cell proliferation were assessed by viable cell count, cell growth curve and colony formation assay in soft agar. The results showed that the

transgenic Jurkat cells displayed slow growth rate, extended doubling time and reduced colony formation capability, as compared with the cells transfected with pBK/CMV empty vector (P < 0.01). It is concluded that exogenous
HCAP1
gene products could inhibit the proliferation of Jurkat cells.
