Serum CA-50 antigen in the normal individuals and cancer patients was assayed by radioimmunoassay. CA-50, one type of carbohydrate antigens, consisting mainly of sialylated glycolipid (a ganglioside) and sialylated glycoprotein, can be isolated from the primary or metastatic tumors. Using the latest CA-50 antigen radioimmunoinhibition test kits prepared by Steana Diagnostics AB Company, Sweden, the serum CA-50 content was measured in 62 normal individuals, 9 primary hepatocellular carcinomas, 9 liver benign tumors, 23 ovarian or uterine carcinomas, 22 other malignancies and 21 benign tumors. The normal value was 6.05 +/- 4.93 U/ml. The test values in cancer patients were significantly higher than those in normals or
benign tumor
patients. Taking the normal mean value plus two times of standard deviation as positive, the positive rate was 77.7% for
liver cancer
, 65.2% for ovary and uterus carcinomas, 50% for the other cancers. There was only one positive in 62 normals with a false positive rate of 1.6%. Furthermore, 19 patients, being in the remission period or cured by effective treatment, gave normal CA-50 value. It is shown that this method can be used in cancer diagnosis, differential diagnosis and the monitor, prediction of prognosis.

Serum transforming growth factor beta 1 (TGF beta 1) concentration was determined in 47 patients with primary hepatic carcinoma (PHC) and 77 patients with various liver diseases and gastrointestinal tumors by means of enzyme-linked immunosorbent assay system. The result revealed that serum TGF beta 1 level of PHC patients were significantly higher (257.6 +/- 126.0 micrograms/L) than those in normal subjects (81.5 +/- 43.5 micrograms/L), in patients with hepatitis (152.4 +/- 98.4 micrograms/L), cirrhosis (191.8 +/- 96.3 micrograms/L), hepatic
benign tumor
(91.9 +/- 37.9 micrograms/L), metastatic
liver cancer
(146.4 +/- 73.7 micrograms/L) and gastrointestinal tumor (128.7 +/- 56.4 micrograms/L) (P < 0.01 or 0.05). Serum TGF beta 1 level was elevated in 34 of 47 (72.3%) PHC patients, including 5 of 7 (91.4%) small (< 5 cm) PHC patients. The sensitivity and specificity of diagnosis of TGF beta 1 in PHC were 72.3% and 77.9%, respectively. The diagnostic positivity was 78.5% in AFP-negative PHC patients. The combined assay of serum TGF beta 1 and AFP could further raise the detection rate of PHC up to 93.6%. Above results suggest that serum TGF beta 1 might be a candidate for a novel tumor marker for diagnosis and monitoring of PHC.

Previous work from our laboratory has implicated oxidative DNA damage and genetic instability in the etiology of transforming growth factor-alpha (TGFalpha)/c-myc-associated hepatocarcinogenesis. formation. We examined whether defects in the DNA repair pathways contribute to the acceleration of
liver cancer
in TGFalpha/c-myc mice. A cDNA expression array containing 140 known genes and multiplex RT-PCR were used to compare the basal levels of expression of DNA repair genes at the dysplastic stage. Thirty-five percent (8/23) and 43% (10/23) of DNA repair genes were constitutively up-regulated in 10-week-old TGFalpha/c-myc and c-myc transgenic livers, respectively, compared with wild-type controls. The commonly up-regulated genes were OGG1 and NTH1 of base excision repair; ERCC5, RAD23A, and RAD23B of nucleotide excision repair; and RAD50, RAD52, and RAD54 involved in DNA strand break repair. Additional treatment with a peroxisome proliferator, Wy-14,643, known to increase the level of oxidants in the liver, failed to induce a further increase in the expression level of DNA repair enzymes in TGFalpha/c-myc but not in c-myc or wild-type livers. Moreover, expression of several genes, including Ku80, PMS2, and ATM, was decreased in TGFalpha/c-myc livers, suggesting a fault or inefficient activation of the DNA repair pathway upon induction of oxidative stress. Together, the results show that DNA damage response is attenuated in TGFalpha/c-myc mice, creating a condition that may contribute to acceleration of

in this model.

The authors present the case of 68 year old patient in whom preoperative

with ulcers were diagnosed. Intraoperative macroscopic picture suggested malignant process. The patient underwent anterior rectal resection with regional lymphadenectomy. The postoperative histopathological evaluation with immunohistochemical studies angiosarcoma was diagnosed. A four year survival free of cancer was obtained, and the patient died due to myocardial infarct. Literature describes the short periods of survival of patients with vascular sarcoma of the colon. The most common reason for decease are metastases with massive haemorrhages. Bleeding tendency results from growing Kasabach-Merritt syndrome. It is characterized by profound thrombocytopenia and coagulopathy. The presented patient is a unique example of angiosarcoma with long period of survival.
