The component sterols in the lipid from a yeast, a species of Picia, grown on n-alkanes have been studied.
Ergosterol
and zymosterol in the 4-desmethylsterol fraction, 4alpha-methylzymosterol in the 4-monomethylsterol fraction and lanosterol in the 4,4-dimethylsterol fraction are present as the major components in the respective fractions. Among the minor components, the presence of 24-methylene-lanostenol, in addition to fecosterol and 4alpha-methylfecosterol, is indicated.

In order to investigate the effect of the different stereochemistry of C-24 on the microbial C-26 oxidation of sterol side-chain the genetically modified Mycobacterium sp. BCS 396 strain was used to transform erogsterol.
Ergosterol
was converted to 3-oxo-4,22-ergostadien-26-oic acid methyl ester, 3-oxo-1,4,22-ergostatrien-26-oic acid methyl ester, and 3-oxo-1,4,22-ergostatrien-26-oic acid, the structures of which have been determined by IR, 1H NMR, 13C NMR, and mass spectroscopy. The X-ray structure of 3-oxo-4,22-ergostadien-26-oic acid methyl ester revealed that oxidation at C-26 of the ergostane side-chain generates a chiral center with S-configuration at C-25 as a result of chiral induction of the C-24 center.

The sterol composition of Tuber melanosporum was examined by medium-pressure liquid and high-performance liquid chromatography, nuclear magnetic resonance spectroscopy, and mass spectrometry.
Ergosterol
(ergosta-5,7,22-trienol) and brassicasterol (ergosta-5,22-dienol) were identified as the major components (90%). Their quantities and their relative concentrations, probably good indicators for differentiating mature truffles from immature colored ones, were determined during the maturation of the fungus. The results show that the total quantity of sterols decreased with the age of the fungus, but that the ergosterol/brassicasterol ratio remained relatively constant.

The strain of Mycobacterium sp. VKM Ac-1815D was found to convert ergosterol and its 3-acetate mainly to androst-4-ene-3,17-dione (AD) thus demonstrating ability to reduce 7(8)-double bond and hydrolyze sterol ester in addition to oxidation of 3beta-hydroxy group, Delta(5)-Delta(4) isomerization and side-chain degradation.
Ergosterol
bioconversion in the presence of isoflavones and ions of some bivalent metals - known inhibitors of 3beta-hydroxysteroid dehydrogenase, did not alter products composition. Protection of ergosterol 3beta-hydroxyl with methoxymethyl group allowed the formation of bioconversion products retaining the Delta(5,7)-configuration. The major product was identified by mass-spectrometry and proton NMR as 3-methoxymethoxy-androsta-5,7-diene-17-one (MA). The MA producing activity was found to be inducible with sterols, cholestenone or lithocholic acid, but not with dehydroepiandrosterone, AD, androsta-1,4-ene-3,17-dione or organic acids. Under the optimized conditions, the yield of MA reached 5g/l from 10g/l O-methoxymethyl-ergosterol (approx. 60% molar conversion) for 120h. The results might be applied at the production of novel vitamin D derivatives.
