In order to suggest a valid experimental model for correlated studies regarding the complex phenomena of reproductive biology in humans, light (LM), transmission (
TEM
) and scanning electron microscopic (SEM) observations were carried out on vaginae of sexually mature New-Zealand albino rabbits, both in estrus and at 5 h after mating. To avoid artifacts depending upon the operative times for specimen removal and, in particular, to well visualize the inner surface of this organ, vaginal irrigation with fixative was performed prior to the surgical removal. Our data demonstrate that the upper 2/3 of the vaginal duct form a kind of 'cervico-vaginal canal' which shares many histological features with the human endocervix. In fact, this portion is lined within by a monostratified epithelium provided with tall microvillous and ciliated cells and often invaginated in crypts. After mating, the microvillous cells showed evidence of secretory activity, exhibiting dome-like luminal surfaces. Secretory droplets often contacted cilia and
spermatozoa
present in the mucosal furrows and crypts. Because of these singular features and the morphofunctional interactions between vaginal epithelium and male gametes, we believe the rabbit to be a helpful model for comparative biology in humans, concerning sperm capacitation and the general reactivity of the female genital tract during the reproductive cycle.

Acrosome reactions were induced in seminal
spermatozoa
of the megachiropteran Pteropus poliocephalus, the grey-headed flying fox, using the ionophore A23187 (10 microM in Tyrodes solution with 1% w/v BSA). Samples were taken at 0, 10, and 120 minutes and fixed for electron microscopy. Initial motility of approximately 70% decreased to approximately 10% over 2 hours. The first motile

without acrosomal caps were seen after 6 minutes, and by 90 minutes approximately 60% of motile

had completed the acrosome reaction. The first sign of the reaction (by

) appears to be swelling and cavitation of the acrosomal matrix within the apical and principal segments, followed or possibly accompanied by fusion and fenestration of the outer acrosomal and plasma membranes and dispersion of the matrix except from the equatorial segment. The end-product of the reaction thus conforms to that seen in other eutherian mammals. However, the reaction exposes a uniquely moulded organization of subacrosomal material ("pseudoperforatorium"), shaped like an anvil over the rostral rim of the flattened nucleus and encased by the remaining inner acrosomal membrane. This has two "lateral" spikes on each side that extend approximately to the same width as the head in the region of the equatorial segment. While these barbs could serve to stabilize or anchor the apical segment of the acrosome, their exposure following the acrosome reaction suggests some specific mechanical role in penetration of the zona pellucida or in protecting more caudal structures from abrasive damage.

In this study we have examined the behaviour and the localization of the fucose-binding protein (FBP) in boar
spermatozoa
during ionophore induced acrosome reaction (AR) by means of normal
TEM
and specimen preparation in toto. During early stages of AR the FBP is first localized at the border between equatorial segment and anterior acrosome. With the propagation of the AR the FBP is dramatically expressed and visible over the entire surface of the acrosome and equatorial segment.

pictures of this stages show that the FBP is associated with the OAM. At later stages of AR, when acrosomal ghost formation occur, the FBP is associated with the acrosomal ghost, and equatorial segment and to a very low degree also with the IAM. It is concluded from this data that the FBP is responsible for the specific binding of the ghost-sperm unit to the zona pellucida.

by means of a specifically developed modified enzyme-linked-lectin-assay using glycosylated peroxidase derivatives. The distribution of the fucose binding protein was assessed by means of fluorescence microscopy with fluoresceinyl-glycosylated peroxidase. an intensive binding of fucosylperoxidase-colloidal gold was predominantly found at the apical part of the acrosome appearing as a crescent-like area.
