E. coli HX88108 was isolated from a patient and found to produce plasmid-encode beta-lactamases with conferring highly resistance to ceforperazone(CPZ). The beta-lactamases of the E. coli HX88108 and transformants pFC, pFT1, pFT2 and pFT3 were studied. The beta-lactamases stability test among 11 beta-lactam antibiotics showed that beta-lactamases from E. coli HX88108. pFC, pFT1, readily hydrolyzed penicillins, the first, second-generation cephalosporins and CPZ. beta-lactamases of pFT2 and pFT3 hydrolyzed penicillins more strongly than cephalosporins. On the other hand, experiments of inhibiting enzyme were carried out. The results indicated that beta-lactamases of HX88108, pTF1, pFT2 and pFT3 were inhibited by clavulanic acid(CA) and sulbactam (SBT). Enzyme of pFC was inhibited poorly by CA and SBT. Through isoelectric focusing technique, the PIs were as follows: HX88108 contained three beta-lactamases, of which the PIs were 5.25, 5.3 and 5.6 respectively; the PIs of beta-lactamases from pFT2, pFT3, were 5.3 and 5.6. pFC and pFT1 were different plasmids encoded beta-lactamases with the same PI 5.25. The results indicate that the beta-lactamases of E. coli HX88108 may be a new member in
TEM
farmily.

In order to investigate the degeneration and regeneration of skeletal muscle fiber after tenotomy, the tenotomy were carried out on gastrocnemius muscles in 15 adult BALB/C mice. The tenotomized muscles were removed on 1, 2 and 4 weeks postoperatively. Specimens were processed for histological study with light microscope and

, and skeletal muscle specific protein expression analysis with myoactin (MA) and myoglobin (MG) immunohistochemical staining. The results showed that the regenerating muscle cells and the degenerating muscle fibre coexisted in tenotomized muscle. The internal structure and histochemical pattern of muscle fiber changed after tenotomy. The regenerating muscle cells fused with degenerating fibres.

Escherichia coli HX88108, which is resistant to cefoperazone(CPZ), was isolated from a severely infected patient. We studied genetical basis of beta-lactamase produced in E. coli HX88108 by pFL25, one of the recombinant plasmid of pFC. Largescale pFL25 plasmid was extracted, purified, and cleaved with restriction endonuclease EcoR I, Sal I, Pvu I, then subcloned into vector pUC19 as 1.9 kb, 0.9 kb, 0.65 kb fragments respectively. Recomminant plasmids were selected by alpha-complementation and determined by restriction endonuclease analysis. DNA sequencing was performed by the dideoxy polymerase chain termination method. Partial nucleotide sequence(1-78 nucleotide position) of the gene was found to be highly homologous (97%) with the gene coding for
TEM
-52 extended spectrum beta-lactamase of K. pneumonise, suggesting that the beta-lactamase coded by the cefoperazone resistant gene might be derived from
TEM
-type beta-lactamase.

Bacteriorhodopsin is a membrane protein of halobacteria and functions as a light-driven protein pump. After we isolated bR from cultured halobacteria, bR was mixed with amphiphilic DPPC under different pH. The liposomes were formed after sonication. The remaining biological activity of bR as a proton pump was then verified and pulsed-light-induced proton movement was detected, while liposomes were observed via
TEM
and light scattering. Although there was no noticeable difference in morphologies of both vesicles formed at pH=2.5 and pH=7.0, the orientations of bR in both liposomes were found to be opposite under these two conditions. This experiment confirmed that the protein bR, when self-assembling into liposomes under acid medium, kept the similar orientation as in the natural plasmid membrane. Such a normal-orientation was, however, different from most of reports in the literature about liposomes prepared under normal neutral conditions.
