Culture conditions favouring the simultaneous formation of soluble protein and inclusion bodies (IBs) were chosen for producing the cytoplasmic protein beta-galactosidase or the periplasmic protein TEM-beta-lactamase. Soluble and insoluble cell fractions of Escherichia coli producing either beta-galactosidase or TEM-beta-lactamase were analyzed by one- and two-dimensional gel electrophoresis and subsequent silver staining or immunodetection of the recombinant protein. The results show that truncated fragments of the recombinant protein were not present in the soluble cell fraction but accumulate in the IB fraction. The presence of other cellular, non-plasmid-encoded proteins in IB preparations such as the outer membrane proteins OmpF, OmpC, and OmpA or the ribosomal subunit proteins L7/L12 was attributed to co-precipitation of cell-debris-associated components. Protein-folding enzymes were not detected in IB preparations. The specificity of in-vivo protein association in the formation of IBs and its implication on protein purification is discussed.

Alloys and intermetallic compounds of Au with the 3d transition metals Fe, Co, and Ni are nonequilibrium phases that have many useful potential applications as catalytic, magnetic, optic, and multifunctional magneto-optic materials. However, the atomically ordered Au-M (M = Fe, Co, Ni) intermetallics are particularly elusive from a synthetic standpoint. Here we report the low-temperature solution synthesis of the L12 (Cu3Au-type) intermetallic compounds Au3Fe, Au3Co, and Au3Ni using n-butyllithium as a reducing agent. Reaction pathway studies for the Au3Co system indicate that Au nucleates first, followed by Co incorporation to form the intermetallic. The nonequilibrium intermetallic nanocrystals have been characterized by powder XRD, TEM, EDS, selected area electron diffraction, and nanobeam electron diffraction, which collectively confirm the compositions and superlattice structures.

The magnetization configuration of a novel nano-chessboard structure consisting of L10 and L12 phases in a Co40Pt60 alloy is investigated using Lorentz transmission electron microscopy (LTEM) and micro-magnetic simulations. We show high-resolution LTEM images of nano-size magnetic features acquired through spherical aberration correction in Lorentz Fresnel mode. Phase reconstructions and LTEM image simulations are carried out to fully understand the magnetic microstructure. The experimental Fresnel images of the nano-chessboard structure show zig-zag shaped magnetic domain walls at the inter-phase boundaries between L10 and L12 phases. A circular magnetization distribution with vortex and anti-vortex type arrangement is evident in the phase reconstructed magnetic induction maps as well as simulated maps. The magnetic contrast in experimental LTEM images is interpreted with the help of magnetic induction maps simulated for various relative electron beam-sample orientations inside the TEM.
