Two-dimensional electrophoresis of six fractions of split ejaculates from normal men (median age 23 years, n = 6) revealed large differences in the pattern of proteins found between the prostate-enriched fractions and secretions associated with the seminal vesicles. The glandular contributions were assessed using the concentrations of spermine, zinc and prostatic acid phosphatase (PAP) for prostatic secretion, and fructose and prostaglandin E for seminal vesicle secretion in the various fractions. Aside from PAP, four additional proteins were apparently associated with the prostatic fraction, one of which shared the biochemical characteristics of the specific ventral lobe protein of the rat prostate, prostatein (molecular mass 16 000, pI 4.8). The presumptive vesicular fractions contained a large number of low molecular mass proteins (10-20 000), with widely varying pI-values. The concentration of albumin and transferrin appeared to be highest in the sperm cell enriched fractions, indicating a major contribution to the ejaculate of testicular/epididymal origin.

A 7-year-old boy was brought to our clinic with the chief complaint of terminal pain on urination. Rectal examination revealed a prostate of almost normal size and consistency, but seminal vesicles were palpated to be hard and in a painful mass. Plain x-ray and urethrogram demonstrated several small stones in the posterior urethra and in seminal vesicles. In the first operation, the stones located in the posterior urethra were removed through the transvesical approach, and the seminal vesicular stones were extirpated in the next operation. The stones were found to consist of magnesium ammonium phosphate by infrared rays spectrum analysis. The histopathological findings of the partially resected seminal vesicles showed no inflammatory or tuberculous changes. Twenty cases of stones in seminal vesicle, in seminal canal, or in ejaculatory duct have been reported in the Japanese literature. Etiology, pathology, clinical problems and therapy of the stones of seminal tract are discussed in relation to the cases reported in Japan including the present case.

During the years 1971 to 1983 24 patients underwent radical prostatectomy for localised carcinoma of the prostate. There was one post-operative death from a pulmonary embolism, but all other patients were available for follow-up. All tumours were confirmed on palpation to be clinically localised to the prostate. Patients with clinical stages T0, T1, and T2 were referred for radical surgery. Patients with incidental carcinoma (T0), with less than 10% of the total tissue involved with carcinoma, were not subjected to surgery, and were followed up by observation only. Two patients developed recurrence of tumour at 18 to 12 months, and 2 years. One patient died 1 year later from the disease; he had a pathological T3 tumour with microscopic extension into the seminal vesicles. All other patients are alive and tumour-free at follow-up and 20 of the 23 patients are fully continent.

Neuropeptide Y (NPY) was found in high concentrations in the male genital tract. NPY levels were highest in the seminal vesicles, prostate, corpus cavernosum and vas deferens, where large numbers of immunoreactive nerve fibres were detected. Considerable quantities were also found in the epididymis and spongiosum. Lower concentrations were found in the glans penis, testis and foreskin. The presence of a large number of nerves containing NPY suggest that this active neuropeptide may play a role in control of genital function.

Seminal lead concentrations in normal and vasectomized males with no history of lead exposure were measured by flameless atomic absorption spectrophotometry. Extremely low concentrations of 3.8 micrograms/dl were found in normal males versus 4.1 micrograms/dl in vasectomized males. No significant correlation between lead and sperm count or sperm motility existed. It is suggested that the major contribution of lead in human semen is from the prostate and/or seminal vesicles and that very low lead levels may not have measurable effect on fertility indices.

Effects of Kampo (Chinese) drugs on hypothalamo-pituitary-gonadal system were investigated in immature male rats of Holtzman strain. Hachimijiogan(H), its components and Ninjin(N) were administered orally to the animals for 28 days, and the weight of the various organs, pituitary content of FSH, LH and PRL, and serum concentration of FSH, LH, PRL and testosterone were measured. H caused a significant increase in the weight of seminal vesicles while H, Sanyaku(S) and N caused a significant increase in the weight of the prostate, respectively. The pituitary content of FSH was significantly increased by H and S, respectively, while the testosterone concentration was significantly decreased by Jio, S and N, respectively. No increase in the weight of the prostate or seminal vesicles followed the administration of H, S and N to orchiectomied immature rats. This result clearly shows that these three drugs have no androgenic activity. After the administration of H to immature male rats, the character of dihydrotestosterone(DHT) receptor was studied by the DCC method. The number of binding sites of DHT receptor was significantly increased by 32% (p less than 0.001) by H. These results demonstrate that H has a stimulatory effect on androgen target organs through the increase in androgen receptor in these organs.

PMID:[Effects of Hachimijiogan on hypothalamo-pituitary-testicular system]. 666 39

The present study constitutes a systematic search for biochemical parameters which could be used to monitor the isolated function of various prostate lobes and accessory sex glands in the rat. Various parameters were measured in the 105 000 X g supernatants of the ventral, lateral and dorsal prostate lobes, and in the coagulating glands and seminal vesicles. These included spermine, citric acid, acid phosphatase, fructose and zinc, as well as the pattern of proteins in both cytosols and secretions, which were determined using one dimensional SDS, polyacrylamide gel electrophoresis. The results suggest that several parameters can be used including: Ventral lobe--spermine or prostatein; Lateral lobe--zinc or a 15 000 dalton protein; Dorsal lobe--spermine or a 60 000 dalton protein; and coagulating gland--fructose.

Hypogonadal (hpg) mice lack hypothalamic luteinizing hormone releasing hormone (LH-RH); consequently the pituitary gonadotrophs, though differentiated, remain inactive. The acute and chronic responses of gonadotrophs in hpg mice to LH-RH to male hpg mice produced an eightfold increase in plasma LH after 5 min, but granule depletion was not detectable ultrastructurally. Vacuoles (300-600 nm) were present more frequently in gonadotrophs from hpg mice given LH-RH compared with control mice. Daily s.c. administration of 2 micrograms LH-RH to male hpg mice for 20 days stimulated a 63-fold increase in the pituitary content of FSH but only a twofold increase in the pituitary content of LH. Testicular weight increased fivefold but the weight of the seminal vesicles did not change. The frequency of cells immunoreactive for LH increased (x 1.6), the gonadotrophs hypertrophied (x 1.9) and the cell content of granules increased (x 2.3) to values close to the normal range. The rough endoplasmic reticulum and Golgi apparatus became more prominent. A striking result of long-term daily administration of LH-RH was the accumulation of large (1-2 micrometers) lipid droplets in about 40% of gonadotrophs. All these changes induced by LH-RH regressed towards the untreated hpg state when LH-RH administration wad discontinued for 5 and 10 days. These results show that the gonadotrophs of hpg mice can be stimulated trophically by LH-RH, and that long-term administration stimulates the synthesis and release of the gonadotrophins but the effect on FSH is much greater than on LH.

Testis proteins of Drosophila melanogaster deficient for six different Y-chromosome regions were fractionated by means of a sodium dodecyl sulfate/polyacrylamide gel system designed to separate high molecular weight polypeptides (Mr, greater than 200,000). Analysis of the banding patterns indicates that the three regions containing fertility genes kl-2, kl-3, and kl-5 are responsible for three different high molecular weight polypeptides. Several observations indicate that these polypeptides are structural components of the sperm axoneme. They are present in seminal vesicles, which are highly enriched for mature sperm. They are first detected during development at a time when the first spermatids are elongating. Finally, deletion of either kl-5 or kl-3 leads to the absence of the outer dynein arm of the peripheral doublets of the axoneme. Although absence of the kl-2 region eliminates the third polypeptide, an associated structural defect in the axoneme has yet to be identified. The three polypeptides are in the Mr 300,000-350,000 range, and their mobilities are similar to those of dynein polypeptides from Chlamydomonas axonemes. Experiments using dosage variation and a temperature-sensitive sterile mutation in kl-5 suggest that the Y-chromosome regions contain the coding sequences for the polypeptides.

The relative binding affinities of testosterone (T), 19-nortestosterone (N) and their 5 alpha-reduced derivatives: 5 alpha-dihydrotestosterone (DHT) and 5 alpha-dihydro-19-nortestosterone (DHN) to the androgen receptor of the rat seminal vesicle was studied using competition experiments. In cell-free extracts incubated at +10 degrees C for 18 h the relative binding affinities of these steroids (DHT greater than T = DHN = N) proved to be specific for the androgen receptor, in the sense that only prostatic extracts gave a similar result while three other androgen binding proteins (human sex steroid binding globulin, rat epididymal androgen binding protein and an antibody raised against T) exhibited quite different binding specificities. In minced seminal vesicles incubated at 37 degrees C for 1 h the binding affinities showed marked differences (DHT greater than N greater than T greater than or equal to DHN) and similar patterns were observed with both the cytoplasmic and the nu clear receptors. Our findings suggest that (I) the simultaneous presence of a 4-ene double bond and 19-methyl group in T does not favor the tight binding of T to the androgen receptor; therefore, either saturation of this double bond or elimination of the 19-methyl group leads to increased binding and (II) while 5 alpha-reduction of T increases the affinity of this steroid to the receptor, that of N does not influence or rather tends to decrease the binding affinity. The opposite changes observed in the binding affinities of T and N after their 5 alpha-reduction may account for the lower androgenicity of N. On the other hand, the relative myotropic activity in vivo of these steroids is apparently determined by the ratio of their affinities (N/T approximately 3 at 37 degrees C) to the androgen receptor.
