Indomethacin
has been suggested as a therapeutic tool to manage elevated intracranial pressure in patients with severe head injury and patients undergoing craniotomy for brain tumors.
Indomethacin
is a non-selective cyclooxygenase inhibitor. Compared to other cyclooxygenase inhibitors indomethacin has unique effects on cerebral blood flow. Administration of indomethacin causes cerebral vasoconstriction and decreases cerebral blood flow, which elicits a decrease in intracranial pressure. The mechanism of indomethacin-induced cerebral vasoconstriction is not completely understood and controversies exist whether indomethacin causes cerebral ischemia. Indomethacin
(IND) was incorporated into the materials and the kinetic of the release profiles was studied by applying the Pepas and Sahlin model. In this sense, materials modified by DI, particularly that with hydrothermal treatment, showed the higher adsorption capacity and slower release rate. This behavior could be associated to the synthesis method used that allowed a high percentage of silanol groups available in the solids surface, which can interact with the IND molecule. This feature coupled with the superparamagnetic behavior; make these materials very interesting for drug delivery systems.

Intracranial hypertension (IH) is one of the final pathways of acute brain injury. In severe traumatic brain injury (sTBI), it independently predicts poor outcomes. Its control represents a key aspect of the management. Lack of response to conventional therapies signals a state of ''refractory IH'', with an associated mortality rate of 80-100%. In such cases, hypothermia, barbiturates at high doses (BBT), decompressive craniectomy (DC), and extreme hyperventilation are utilized. However, none of them has proven efficacy.
