The in vivo localization of a polyclonal antibody (pAb) against a glycoprotein with a molecular mass of 68 kDa (GP68), which was found in developing mouse brain, was studied in murine tumor models to evaluate potential applications of this antibody for in vivo radioimmunodetection and/or therapy of cancer. The tissue distribution of 125I-labeled GP68 pAb 3 days after i.v. injection into mice bearing four different kinds of solid tumor revealed a high uptake ratio by adenocarcinoma 755 and Lewis 3LL
lung cancer
. In contrast, the uptake ratio was low in mice bearing Ehrlich solid tumor and
sarcoma
-180 (S-180). These uptake ratios accorded well with the in vitro binding activity of this antibody with the tumor cells. In an immunoscintigraphic study, adenocarcinoma 755 was successfully visualized with 67Ga-labeled GP68 pAb. The results of these biodistribution and in vivo radioimmunoscintigraphic studies suggest that GP68 antibody may be applicable to the diagnosis and/or therapy of cancer.

The expression of granulocyte colony-stimulating factor (G-CSF) mRNA was studied in human non-hematopoietic tumors, including 18 cases of lung cancers 10 cases of stomach cancers, three cases of glioblastomas, and one case each of breast phyllode
sarcoma
, thyroid cancer, and hepatocellular carcinoma. Northern blot analysis detected G-CSF mRNA in two of the
lung cancer
cases, in one of the glioblastoma cases, and in both the breast phyllode
sarcoma
and hepatocellular carcinoma cases. Since G-CSF receptors were not detected on the tumor cells by 125I-G-CSF binding assay, G-CSF autocrine loop are probably not involved in the growth of these G-CSF-producing tumors. Interestingly, granulocyte-macrophage colony-stimulating factor (GM-CSF) mRNA was concomitantly expressed in most of these G-CSF-producing tumors. No major gene deletions or rearrangements of G-CSF and GM-CSF genes were demonstrated by Southern blot analysis in the tumors expressing G-CSF and GM-CSF mRNAs except for one of the glioblastomas (G3) in which one chromosome 17 allele was deleted. Although the mechanism of the concomitant expression of G-CSF and GM-CSF mRNA is unknown, relatively high frequency of this phenomenon suggests the presence of common transcriptional factors acting on regulatory regions of G-CSF and GM-CSF genomes.

Twenty-six white male subjects, who worked with plutonium (239Pu) during World War II at Los Alamos, have been given medical examinations periodically over a period of 42 y to identify potential health effects. Inhalation was the primary mode of Pu exposures. The latest examinations, including urine bioassay and in-vivo measurements for radioactivity, were performed in late 1986 and 1987. The average age of the 22 living subjects in 1986 was 66 y. The diseases and physical changes noted in these persons are characteristic of a male population in their 60s. Estimates of individual Pu depositions, including lung burdens, as of 1987 or at time of death range from 52 to 3180 Bq (1.4 to 86 nCi) with a median value of 500 Bq (13.5 nCi). Four persons from the original group had died as of 1987. The causes of death were

From 47 probands, 7 families had 3 or more cancer cases at ages less than 45 years within the first generation; one satisfied Li's original criteria, two were acceptable because of multiple primary cancer in the probands, and others showed an aggregation of cancers with onsets at early ages, though no

To assess the possible carcinogenicity of phenoxy herbicides and related chlorophenols and dioxins, the International Agency for Research on Cancer is coordinating an international collaborative study of workers exposed to these compounds in their production or use. Four British cohorts of chemical manufacturers which have been recruited to the survey are described. They comprise a total of 2239 men employed during 1963-85. These subjects were traced to 31 December 1987 through the National Health Service Central Register and the National Insurance Index, and their mortality compared with that in the national population. Two deaths were from non-Hodgkin's lymphoma with 0.87 expected. Both deaths occurred more than 10 years after first exposure to phenoxy compounds. One further non-Hodgkin's lymphoma was registered in a living subject with probable exposure to phenoxy compounds. No cases of
soft tissue sarcoma
or Hodgkin's disease were recorded. A nonsignificant excess of
lung cancer
(19 deaths observed, 14.2 expected) is probably attributable to chance or a confounding effect of smoking. In one cohort only there was increased mortality from circulatory disease (34 deaths observed, 20.4 expected). A nested case-control study did not point to any occupational cause for this excess, but further evaluation will be needed during continued follow up.

Cobalt metal and cobalt compounds are extensively used for the production of high-temperature alloys, diamond tools, cemented carbides and hard metals, for the production of various salts used in electroplating and as catalysts, drying agents in paints, additives in animal feeds and pigments. Cobalt oxides are used not only in the enameling industry and for pigments, but also in catalytic applications. There is no indication that cobalt metal and cobalt compounds constitute a health risk for the general population. Allergic reactions (asthma, contact dermatitis) can be induced by certain cobalt compounds. Interstitial fibrosis has also been observed in workers exposed to high concentrations of dust containing cobalt, tungsten, iron, etc., mainly in the cemented carbides and the diamond-polishing industries. Several experiments have demonstrated that single or repeated injections of cobalt metal powder or some forms of cobalt salt and cobalt oxide may give rise to injection site
sarcoma
in rats and in rabbits but the human health significance of such data is questionable. Intratracheal administration of a high dose of one type of cobalt oxide induces lung tumors in rats but not in hamsters. In the latter long-term inhalation of cobalt oxide (10 mg/m3) did not increase the incidence of

. The human data are too limited to assess the potential carcinogenic risk for workers. Co2+ interacts with protein and nucleic acid synthesis and displays only weak mutagenic activity in microorganisms. Some cobalt salts have been reported to enhance morphological transformation of Syrian hamster embryo cells. Cobalt chloride displays some limited mutagenic activity in yeast and some cobalt compounds are able to produce numerical and structural chromosome aberrations in plant cells. Cobalt and its salts appear to be devoid of mutagenic and clastogenic activity in mammalian cells. Cobaltous acetate and cobaltous chloride have not been found to be teratogenic in hamsters and rats respectively.

Tumor-infiltrating lymphocytes (TILs) have been grown from a variety of human tumors. TILs from some patients with melanoma demonstrate lytic activity specific for autologous tumor, and can mediate tumor regression when adoptively transferred to select cancer patients. In this study, we have compared the in vitro properties of lymphocytes from peripheral blood (PBLs), from draining lymph nodes (DLNs), and from tumors (TILs) grown simultaneously from 10 patients: 2 with melanoma, 4 with breast cancer, 1 with gastric cancer, 1 with renal cancer, 1 with
sarcoma
and 1 with
lung cancer
. PBLs, TILs, and DLNs were cultured in RPMI 1640 + 10% human AB serum, 20% LAK cell culture supernatant, and 1,000 u/ml of recombinant interleukin-2. Half of each culture was restimulated with irradiated autologous tumor every 14 days. In all groups, tumor feeding enhanced lymphocyte proliferation, although TILs and DLNs consistently proliferated longer and more rapidly than PBLs. Eight of 10 early cultures of TILs and DLNs contained greater or equal proportions of CD8+ cells compared with CD4+ cells, but in long-term cultures an inversion of that ratio was seen (CD4+ greater than CD8+). In short-term chromium release assays, specific lysis of autologous tumor was seen in tumor-fed TILs and DLNs from one patient with melanoma, DLNs from one patient with breast cancer, and TILs from one patient with
lung cancer
. Other cultures had nonspecific lytic activity. Specific cytotoxicity against autologous tumor sometimes became apparent only after prolonged culture and repeated restimulation with autologous tumor.
