We report a case of large granular lymphocytic leukaemia (LGLL) with mixed T-cell/B-cell phenotypes. The LGLL cells expressed T-cell markers such as CD1, CD2, CD3, CD5, CD7, CD8 and CD57. The CD8+ LGLL cells coexpressed B-cell markers including CD20 and PCA-1, and a fraction of purified CD8+ LGLL cells secreted double isotypes of immunoglobulins (IgG-kappa and IgA-kappa). Both TCRB and IGH genes were clonally rearranged. The LGLL cells could be divided into at least three subpopulations that were cytogenetically distinct, and all subpopulations involved the 11q23. The expression of both T- and B-cell markers on the LGLL cells suggests the involvement of a putative common lymphoid progenitor in leukaemic transformation.

Due to its dependence on IL-4 and IL-13 production, IgE production is frequently used to assess the type 2 character of an immune effector response. It is particularly relevant to measure IgE in murine models of immediate hypersensitivity, as allergen specific IgE is a critical effector molecule in this process. Given the complexity of developing ELISAs to measure specific IgE, total IgE levels are often reported with the implicit assumption that this provides an accurate gauge of specific IgE responses. Here, we rigorously test this assumption by examining the relationship between total and Ag-specific IgE levels in mice immunized to elicit a wide range of serum IgE responses. We identify a strong, consistent relationship between total and Ag-specific IgE, regardless of the phenotype of the immune response (type 1 vs. type 2 biased), the nature of the immune response (primary vs. recall), the genetic background of mouse strain examined (C57Bl/6, BALB/c or outbred CD1 mice), or the intensity of the initial immunological stimulus (0.2, 2.0 or 100 microg OVA). These findings indicate that measurement of total IgE levels through straightforward, easy to develop, total IgE ELISAs offers an appropriate surrogate for measurement of Ag-specific IgE levels, usually measured through the use of subjective PCA assays or Ag-specific IgE ELISAs.

The morphological development of in vitro single cerebellar Purkinje cells obtained from wild type P1 CD1 mice was assessed through a dedicated non-invasive technique based on image processing algorithms and multivariate analysis. Image processing algorithms were implemented to extract metrical features characterizing cell structure and dendritic arborization from sequential optical micrographs. Quantitative morphological features were analyzed in order to identify relevant metrical characteristics common to Purkinje cells in wild type P1 CD1 mice. Cell arborization was found to be characterized by a high fractal dimension and the directionality and level of complexity were shown to be key features for cell morphology classification, as underlined using a three-way PCA analysis.
