The heterologous adoptive cutaneous anaphylaxis system was used to determine the kinetics of appearance of IgE-producing cells in various lymphoid tissues of mice following intratracheal (i.t.), intraperitoneal (i.p.), or subcutaneous (s.c.) immunization with tetanus toxoid and Bordetella pertussis organisms. Immunization, i.t. and i.p., produced similar patterns of response with the bronchial lymph nodes quantitatively exceeding the responses in other lymphoid tissues. In both cases the splenic lymphocyte response was second only to the bronchial and both appeared to parallel the serum PCA antibody. It is suggested that both responses represent draining lymph node responses since the bronchial lymph node drains both sites of immunization. After s.c. immunization a primary response of low order was found in the draining popliteal lymph node but not elsewhere. Although a dissociation was seen between responses obtained in various lymphoid tissues following s.c. and i.p. or i.t. immunization, no real evidence for a local mucosal response, such as has been reported for IgA, was obtained. These results lend experimental support to the observations that intratracheal and intraperitoneal immunization routes are most effective in production of IgE antibodies.

The effect of cultured thymic fragment (CTF) transplantation on the IgE response of nude mice was studied. Nude mice (BALB/c nu/nu) were immunized with a mixture of tetanus toxoid and aluminium gel intraperitoneally. Non-CTF transplanted nude mice could not regulate IgE production nor synthesize specific IgE antibody, and all died at 16 weeks of age. Nude mice that were transplanted with CTF from allogeneic low responder strains (C57BL/6, SJL), allogeneic high responder strain (ASW) and syngeneic high responder strain (BALB/c) could regulate IgE production, and these lived a normal life span. Additionally, the tetanus toxoid-specific IgE antibody response, which was estimated by PCA, paralleled that seen in the strain of the thymus donor, i.e. BALB/c and ASW thymus reconstitution produced the highest response, whereas SJL and C57BL/6 recipients' levels were significantly less (P less than 0.05). We postulate that the lesser responses were due to the determination of the phenotype response by the thymic microenvironment. The low responses were shown to be due to regulator T-cell imbalance. These data show that BALB/c T-cell precursors developing in non-BALB/c thymuses interact with BALB/c B cells to produce levels of IgE antibody that are more characteristic of the non-BALB/c differentiating microenvironment than of their own genetic background.

The composition of various isolated antibodies was determined by quantitative analyses for heavy chain subgroups and light chain types. Certain antibodies such as anti-tetanus toxoid and anti-A isoagglutinins were predominantly of the major gammaG1-type. However, a high preponderance of molecules of the minor gammaG2-subgroup was found for antibodies to dextran, levan, and teichoic acid. These findings explain some unusual features previously noted for anti-dextrans such as weak PCA reactions and lack of Gm antigens. Studies of several isolated antibodies from single heterozygous individuals showed a selective absence of genetic markers in certain antibodies and their presence in others. The "allelic exclusion" principle was clearly evident in the isolated antibodies of two different individuals. Large differences in the ratio of kappa to lambda light chains were observed for the same type of antibody from different individuals. Subfractionation of dextran antibodies by affinity for specific glycosidic linkage or combining site size produced marked changes in the ratios. The isomaltohexaose eluates of the dextran antibodies from two subjects were primarily kappa and the isomaltotriose eluates were predominantly lambda. The one anti-levan antibody studied was uniquely homogeneous, consisting exclusively of gammaG2-heavy chains and kappa light chains. By these criteria as well as others, it closely resembled myeloma proteins.
