The failure of infrarenal aortic open reconstruction due to sterile sovranastomotic abdominal aortic aneurysm (SS-AAA) is a rare and complex long-term complication. Even if they undergo the same treatment, is necessary to distinguish between true aneurysmal degeneration of proximal aorta and chronic proximal aortic anastomosis sterile rupture with consequent false aneurysm formation: we call proximal para-anastomotic abdominal aortic aneurysm (PPA-AAA) the first and proximal anastomotic false abdominal aortic aneurysm (PAF-AAA) the latter. The etiology of this complication is exclusively degenerative and it occurs in the absence of infection, which has totally different features. SS-AAA have been reported in 1 to 4% patients, but the available studies differ about patient selection and diagnostic methods. According to these considerations we can suppose the real incidence greater and near to 25% in over 10 years follow-up patients. Clinical findings of PPA and PAF-AAA before rupture are poor and this consideration emphasizes the necessity of a long term ultrasound follow-up. Best diagnostic tools after echographic detection of SS-AAA are spiral TC scan and MR imaging. Due to image accuracy, the short time necessary to take the images and availability spiral TC has taken the place of standard TC and arteriography. Scar tissue field and visceral vessels involvement with consequent proximal clamping are the main problems in open repair of SS-AAA. Elective open repair mortality rate varies from 0 to 17% and increases dramatically after rupture. Endovascular repair at the present is suitable only for hardly selected cases, because of frequent visceral involvement. We report our 17 patients series (8 PPA and 9 PAF-AAA), which we have observed friom 1991 to 2003 in a total amount of 1363 abdominal aortic aneurysms treated. All the patients have been treated with elective open repair with a global perioperative mortality of 6% (1/17).

In each of three experiments, in vitro-matured and -fertilised zygotes were cultured to Day 7 post insemination in synthetic oviductal fluid (SOF). In Experiment 1, zygotes were cultured in groups in either SOF plus albumin (SOFA) or serum (SOFS) and then blastocysts were cultured individually for a further 24 h without a change of media. In Experiment 2, zygotes were cultured in groups using a 2 x 2 factorial design in SOFA or SOFS, with or without recombinant ovine granulocyte-macrophage colony stimulating factor (GM-CSF; 5 ng mL(-1)). Blastocysts were then cultured individually using a split-plot design in SOFA or SOFS with or without GM-CSF. In Experiment 3, zygotes were cultured in SOFA in which GM-CSF was absent (A) or present (P) during Days 1-3, Days 3-5 or Days 5-7 of IVC in six combinations as follows: AAA, AAP, APP, PPP, PPA and PAA. Serum or GM-CSF increased secretion of interferon (IFN)-tau in Experiments 1 and 2 both between Days 5 and 7 of group culture and during individual culture. Secretion of IFN-tau during individual culture was determined by the medium in which embryos were group cultured and the effects of GM-CSF and serum were not additive. In Experiment 3, the presence of GM-CSF between Days 1 and 3 of culture was responsible for stimulation of secretion of IFN-tau between Days 5 and 7; IFN-tau secretion was detected as early as Day 3 post insemination.

To clarify the mechanism for substrate recognition of alpha-aminoadipate aminotransferase (AAA-AT) from Thermus thermophilus, the crystal structure of AAA-AT complexed with N-(5'-phosphopyridoxyl)-l-glutamate (PPE) was determined at 1.67 A resolution. The crystal structure revealed that PPE is recognized by amino acid residues the same as those seen in N-(5'-phosphopyridoxyl)-l-alpha-aminoadipate (PPA) recognition; however, to bind the gamma-carboxyl group of Glu at a fixed position, the Calpha atom of the Glu moiety moves 0.80 A toward the gamma-carboxyl group in the PPE complex. Markedly decreased activity for Asp can be explained by the shortness of the aspartyl side chain to be recognized by Arg23 and further dislocation of the Calpha atom of bound Asp. Site-directed mutagenesis revealed that Arg23 has dual functions for reaction, (i) recognition of gamma (delta)-carboxyl group of Glu (AAA) and (ii) rearrangement of alpha2 helix by changing the interacting partners to place the hydrophobic substrate at the suitable position.

The yellow-green alga Trachydiscus minutus (Eustigmatophyceae, Heterocontophyta) was cultivated in a standard medium and under nitrogen- and phosphorus-starvation and its triacylglycerols were analyzed by RP-HPLC/MS-APCI. The molecular species of triacylglycerols included a total of 74 triacylglycerols having at least one polyunsaturated fatty acid. Polyunsaturated triacylglycerols were identified for the first time in a yellow-green alga. N-starvation brought about a nearly 50% drop in TAGs containing EPA, and also decreased TAGs containing ARA, while P-starvation had a sizable effect on those TAGs that contain two or three arachidonic acids. In four TAGs containing PUFA, i.e. EEE, EEA, EAA and AAA, N-starvation caused a rapid fivefold increase in ARA content and the ratio of TAGs containing ARA, i.e. AEE to AAA increased tenfold relative to control. Regioisomeric characterization of triacylglycerols containing palmitic, arachidonic (ARA) and eicosapentaenoic acids (EPA) showed that the proportion of positional isomers is affected by N- and P-starvation. N- and P-starvation also changed the ratio of symmetrical to asymmetrical TAGs. Positional isomers exhibited identical ratios of symmetrical and asymmetrical TAGs irrespective of the type of FAs. In control cultivation the major TAGs with a single PUFA were symmetrical ones (PEP or PAP) whose ratio to asymmetrical counterparts (PPE or PPA) was about 3:1, whereas N- and P-starvation yielded opposite ratios, 1:3-1:5. The control cultivation yielded ~90% asymmetrical TAGs with two PUFAs (i.e. PEE and PAA), whereas with N- and P-starvation the ratio of symmetrical to asymmetrical TAGs increased to 2:1 and 3:2, respectively.
