Inbred CFW/L1 or BALB/c male mice were injected intraperitoneally with 1 X 10(5)
EAC
cells. They died in 98.6 or 100%, respectively, within 100 days of observation. Intraperitoneal injection of 6 X 10(7) syngeneic spleen cells from 6--9-week-old CFW/L1 or BALB/c donors 2.5--4 h after tumour inoculation significantly prolonged MST and 16.7 or 23.5% of mice, respectively, survived for 100 days. Smaller amounts of spleen cells, thymus cells, mixed spleen and thymus cells, or spleen cells from older donors did not show this effect. Homogenized spleen cells were also inactive. Antitumour activity of spleen cells was still evident when they were isolated from irradiated (2 000R) donors or were given 24 h after tumour inoculation.
Spleen
cells used for in vivo studies showed spontaneous cytotoxic activity in vitro against
EAC
cells in the 51Cr-release cell-mediated cytotoxicity test. The possible participation of NK cells in the observed in vivo antitumour activity of normal spleen cells is discussed.

The normal human lymphocyte population which exhibits "spontaneous" cytolysis of EB2 Burkitt's lymphoma cells has been characterized. The effector cell has EA and
EAC
' receptors but lacks E receptors and probably surface Ig. "Spontaneous" anti-EB2 cytotoxicity was not reduced by preincubation of the effector cells with plastic or iron carbonyl or by passage through cotton wool or agarose columns but was reduced by passage through nylon wool columns. Thymocytes were not cytotoxic to EB2 cells, and chronic lymphocytic leukaemia cells (of B cell characteristics) had reduced cytotoxicity compared with normal lymphocytes. Cells from various lymphoid organs of rats and guinea-pigs were also cytotoxic to EB2 cells with reactivity in spleen greater than or equal to blood greater than lymph nodes.
Spleen
cells from neonatally thymectomized rats had increased cytotoxicity compared with normal rat spleen cells, suggesting that T lymphocytes are not essential. The effector cell in rat spleen did not adhere to cotton wool or agarose columns, indicating some resemblance to its counterpart in human peripheral blood.

Lymphoid cell populations (spleen, lymph node, peripheral blood, thymus, and bone marrow) from LHC inbred hamsters were studied in order to characterize further the immune response of this species. The direct PFC response to several thymic-dependent or thymic-independent antigens was evaluated. A specific direct PFC response occurred 4 days after immunization with SRBC, DNP-BSA, DNP-lys-Ficoll, TNP-LPS, TNP-BA, and SSS-III. Attempts to induce a polyclonal antibody response with LPS, TNP-LPS, SSS-III, and DNP-lys-Ficoll were unsuccessful. A weak polyclonal response was induced with TNP-BA.
Spleen
cells and PBL responded strongly in vitro to the T-cell mitogens Con A and PHA-P, but gave weak and inconsistent responses to the B-cell mitogens LPS and PI-PC. LHC hamster lymphoid cell populations bore sIg and receptors for C3 (
EAC
rosettes) in approximately the same ratio as various murine species. However, the profile of the number of cells bearing low-to-intermediate densities of sIg differed significantly from those of murine species when analysed with the FACS. There was a sharp reduction in the number of cells with low-to-intermediate densities of sIg. These data suggest that B cells in this strain and species lack the ability to translate signals which lead to polyclonal antibody synthesis or lack the appropriate populations of B cells that have membrane receptors for mitogens which are thought to induce such activity in murine systems and provide evidence for separate signals that induce thymus-independent and mitogenic responses. The importance of this model for studying mechanisms involved in B-cell activation is discussed.
