Primary cultures of hamster embryo fibroblasts were transfected by purified, fragmented herpes simplex virus type 1 (HSV1) strain A44 DNA in the presence of carrier DNA. Control cultures were transfected with carrier DNA only. A cell line transformed by HSV1 DNA (EH/A44 cell line) was established after serial passages, without senescent crisis. A control cell line (EHT cell line), spontaneously transformed in vitro, was established after a 13th passage crisis. Both cell lines originated from the same primary culture thereby allowing the comparison of virus DNA induced and spontaneous transformation. Various parameters were studies for both cell lines and their clones: cell growth in vitro, presence of viral antigens, permissivity to superinfection and oncogenicity.

The efficiency of transfection, i.e., the number of plaques per microgram of DNA, was compared for herpes simplex virus (HSV) type 1 and type 2, and herpesvirus eidolon (antigenically unrelated to HSV) DNAs in HSV DNA-transformed cells (EH/A44) and control hamster cells (EHT). The efficiency of transfection of HSV-1 and HSV-2 DNAs was significantly higher in EH/A44 cells than in EHT cells, showing that an early step in HSV infection was involved in the partial resistance of HSV DNA-transformed cells to superinfection by various intact herpesviruses.

In acute experiments in the stomach of dogs electrocoagulation with simultaneous instillation of water (EHT-electrode i.e. electro-hydro-thermo-electrode) proved the most effective electrocoagulation method for controlling standardized bleeding: Optimal visibility at the bleeding site, extension of the therapeutic range as well as the avoidance of charring at the bleeding site are its major advantages. Disturbing adhesion of coagulated material on the electrode is also avoided. The electro-hydro-thermo-electrode is a cheap and portable means of controlling bleeding.

The study of a simple conformational problem, the rotation about the ester bond common to a series of R1COOR2 formula molecules, shows that CNDO/2, INDO and PCILO methods overestimate the interaction term between non bonded atoms. This leads to the prevision of very low barriers and amounts for the CNDO method to the failure in the prevision of some minima. Because of the exaggerated overestimation of this term, the EHT method seems not to be useful except when the steric interaction constitutes the dominant term in the conformation energy.

The cycle of Eimeria acervulina was grown in primary culture of cells of three-week-old-chicken kidneys. EHT medium (Eagle, Hydrolysate of Lactalbumine, Bacto-Tryptose phosphate) allowed the development of this coccidium. 44 hours after infestation of the cells with sporozoites, the first schizonts appeared. The merozoites of first generation were released after 54 hours. They invaded the neighbouring cells and developed in them into schizonts of second generation mature after 68 hours. After 72 and 93 hours, a third and a fourth generation of schizonts were noticed. Merozoites IV create the Gamogony. To obtain the gamogony in vitro, we inoculated the cultures with merozoites IV recovered from axenic animals. The oocysts were released into the medium 45 hours after inoculation.

Conformational energies of histamine and 4-methylhistamine monocations are calculated using the EHT molecular orbital procedure; the results are expressed as potential energy surfaces in which bond rotations (theta1 for ring-Cbeta, theta2 for Cbeta-Calpha) are measured along the axes, and energy variation is indicated by contours. Using the classical Boltzmann partition function and Simpson's rule for normalization, corresponding probability surfaces are generated which take account of the potential surface entropy. Comparing the two surfaces provides regions which are within a given probability contour of histamine but outside this contour for 4-methylhistamine. Thus, at the 99% probability level, three conformational regions defined by the bond rotation angles are indicated as possible "H1-essential" conformations of histamine: viz. trans (theta1=290-330 degrees, theta2=150-210 degrees) and gauche (theta1=260-280 degrees, theta2=30-90 degrees and theta1=290-320 degrees, theta2=270-320 degrees). This procedure provides a quantitative basis for comparison with other histamine derivatives and may have a general value for studying relationships between conformation and biological activity of closely related small molecules.

This study was conducted to elucidate renal uric acid metabolism in patients with primary aldosteronism (PA;16 cases) as compared with normotensive subjects (NT;25 cases) and essential hypertensives (EHT;51 cases). All subjects were hospitalized and received a regular diet(Na;120 mEq,K;75 mEq,daily) for more than two weeks, after which renal clearance tests were performed, and serum uric acid(SUA), fractional excretions of uric acid(FEUA), sodium(FENa), and inorganic phosphorus(FEP) were evaluated. Plasma aldosterone concentration(PAC) was measured in 16 patients with PA before treatment and in 8 patients after adrenalectomy. SUA was lower in PA than in either NT or EHT, and this lowering was more obvious in male subjects. In NT, PA and EHT, FEUA, an index of renal excretion of uric acid, correlated negatively with SUA and positively with FENa and FEP, which reflected sodium reabsorption at the renal total tubules and proximal tubules, respectively. Although FENa was nearly the same in all the three groups, FEUA and FEP were significantly higher in PA than in EHT or NT. However, no significant correlation was found between PAC and SUA or FEUA in PA. In PA a significant increase of SUA, and decreases of FEUA and FEP were observed after the removal of adenoma compared to before the surgery. These results suggest that uric acid transport might be closely related to sodium transport in the renal tubules, particularly at the proximal site, and also lead to the conclusion that the lower SUA in PA resulted from the suppression of reabsorption and/or an enhancement of secretion of uric acid in the proximal tubules, being related to the so-called escape phenomenon.

To investigate the circadian profiles of BP and heart rate in elderly patients with isolated systolic hypertension (ISH), the variability of BP and heart rate during day time (daytime) and the amplitudes of nocturnal fall were evaluated in ISH (n = 19) comparing with those of essential hypertensive patients (EHT, n = 18) and normotensive subjects (NT, n = 16) in the same age range. ISH showed a significantly wider BP variability during the day time and a greater amplitude of nocturnal fall compared with EHT and NT. However, the heart rate variability during the daytime and the amplitude of nocturnal fall were similar among the three groups. ISH showed a stronger correlation between BP and heart rate during 24 hours compared with EHT and NT. Approximately 80% of ISH showed a significant positive correlation between BP and heart rate. These results suggest that the BP of ISH patients is susceptible to fluctuations in autonomic nerve activity.

Plasma immunoreactive atrial natriuretic factor (ANF) and urinary sodium excretion were measured in elderly patients with isolated systolic hypertension (ISH) (n = 11), age-matched essential hypertensive patients (EHT; n = 16) and normotensive subjects (NT; n = 9) before and during a 60 min infusion of hypertonic saline (120 mEq of Na+). An exaggerated natriuresis during the sodium load was observed only in ISH. Baseline plasma ANF levels in ISH were significantly lower (P less than 0.05) than those of EHT and NT. There was no significant change in plasma ANF in EHT and NT subjects after the saline load. In contrast, there was a significant increase in plasma ANF (P less than 0.05) after the saline load in ISH. The change in urinary sodium excretion was significantly correlated with the change in plasma ANF (r = 0.75, P less than 0.01) in ISH. We conclude that an exaggerated natriuresis during a hypertonic saline infusion may be linked to an increase in plasma ANF in elderly ISH patients.
