Neuroblastoma
x glioma hybrid cells (NG108-15), differentiated by treatment with 1.5% dimethyl sulfoxide (DMSO) and 0.5% fetal bovine serum, were used to measure the effect of angiotensin II and III (ANG II and ANG III) on the generation of inositol polyphosphates. ANG II increased the synthesis of inositol monophosphates (IP1), inositol diphosphates (IP2), and inositol trisphosphates (IP3) with maximal responses observed at 300, 120, and 30 sec, respectively. The percent increases above basal values at the maximal responses were 140% +/- 9% (IP1), 142% +/- 4% (IP2), and 132% +/- 4% (IP3). This effect was not attenuated by pretreatment of the cells with
pertussis
toxin. Furthermore, both ANG II and ANG III increased the production of inositol polyphosphates in a dose-dependent manner with ED50 values of 145 nM and 11 nM, respectively. We conclude that differentiated NG108-15 cells express an ANG III selective receptor that mediates phosphatidylinositol breakdown through a
pertussis
toxin insensitive G-protein.

Neuroblastoma
X glioma hybrid cells NG108-15 were treated with a toxin derived from Bordetella
pertussis
. As compared to control cells grown in the absence of toxin, the inhibitory effects of opioid agonists upon cAMP formation were dose-dependently impaired by a non-competitive mechanism. Radioligand binding studies revealed that opioid agonist binding was dramatically reduced in toxin-treated membranes when tested in the presence of Na+/Mg++/GMP-PNP. Further, the potencies of guanine nucleotides to decrease opioid agonist binding were differentially modulated. These studies may facilitate our understanding of the mechanisms responsible for acute and chronic opiate effects.

The presence of specific AII receptors in 6 different human neuroblastoma cell lines was investigated using binding, cAMP and [Ca2+]i studies. We found high affinity (0.1 nM), low capacity ((1-2).10(3) sites/cell) binding sites for [125I](Sar-1,Ile-8)AII in one half of the cell lines studied. In the positive cell lines mathematical modeling of multiple competition curves among AII and analogs strongly indicated the presence of a homogenous class of sites, i.e., AT1 receptors. The presence of AT1 receptors was further substantiated by AII-induced inhibition of VIP-stimulated cAMP levels and by AII-evoked [Ca2+]i transient. The density of AT1 receptors in neuroblastoma cells was not affected by treatment with
pertussis
toxin and retinoic acid but was significantly increased by subacute treatment with VIP. In neuroblastoma cells, AII does not stimulate DNA synthesis, suggesting other roles rather than mitogenesis.

NS20Y cells possess a high density of stereoselective delta opioid receptors as determined by competition binding with 3H-diprenorphine and various opioid ligands. Scatchard analysis of [3H]diprenorphine saturation binding data revealed a Kd = 0.79 +/- 0.17 nM and Bmax = 370 +/- 50 fmol/mg protein. These opioid binding sites have highest affinity for delta opioid receptor selective agonists and lowest affinity for mu opioid receptor selective agonists. Agonist binding was sensitive to the presence of the monovalent cation, Na+. Activation of receptor with D-Ala2, D-Leu5-enkephalin (DADLE) resulted in dose-dependent inhibition of forskolin-stimulated intracellular [3H]cAMP accumulation, which was antagonized by (-)-naloxone but not (+)-naloxone. Relative potencies of various opioid agonists to inhibit intracellular cAMP production paralleled those observed in neuroblastoma x glioma NG108-15 hybrid cells. Pretreatment of NS20Y cells with
pertussis
toxin (PTX) eliminated opioid agonist inhibition of adenylyl cyclase activity. Chronic DADLE treatment resulted in desensitization and down-regulation of opioid receptor. An increase in intracellular [3H]cAMP level above the control was observed in the presence of naloxone after chronic DADLE treatment. Therefore, opioid binding sites in neuroblastoma NS20Y cells possess properties of the classical delta opioid receptor type. After neuroblastoma NS20Y was growth arrested by culturing the cells in serum-free medium for 72 hr, proliferation was reinitiated by addition of fetal calf serum (FCS), 0.01% to 12%, and was monitored by either [3H]thymidine incorporation or by dye viability assay. It was demonstrated that naloxone and naltriben but not Met5-enkephalin could attenuate FCS-induced proliferation in a dose-dependent manner. Naltriben was 54-fold more potent than naloxone to attenuate NS20Y proliferation. The maximal level of viable cells per well was reduced (35.2 +/- 1.9%) with no alteration in FCS concentration-dependent stimulation of growth. Similar inhibition by naloxone (37.3 +/- 2.7%) was observed with [3H]thymidine incorporation studies. This naloxone effect was serum concentration-dependent and could be blocked by culturing NS20Y cells in the presence of both naloxone and Met5-enkephalin. Although pretreatment of NS20Y cells with
pertussis
toxin could attenuate FCS-stimulated proliferation, naloxone effect on growth was not affected by
pertussis
toxin pretreatment. Furthermore, the naloxone effect was not NS20Y specific. A similar naloxone effect was observed with neuroblastoma N1E115, although not with neuroblastoma x glioma NG108-15, nor human neuroblastoma SHSY5Y, cell lines that have been reported to contain delta opioid receptors. Therefore, activation of delta opioid receptor could modulate FCS-induced growth in some but not all neuroblastoma cell lines.

tumors derive from the neural crest and synthesize neurotransmitters including the neuropeptide somatostatin. This study was designed to characterize somatostatin receptors both in primary neuroblastoma tumors and in two neuroblastoma cell lines, SKNSH and IMR32. Somatostatin receptors were identified in 6 of 7 Stage I and II compared to 7 of 19 Stage III and IV tumors. Down-regulation of somatostatin receptor binding was observed in five tumors during disease progression. A lack of high affinity binding of somatostatin was identified as a poor prognostic indicator; negative binding correlated with advanced disease and death. Somatostatin receptor binding was observed in the IMR32 cell line, but not in the SKNSH cell line. IMR32 cells demonstrated a single class of high affinity binding sites for both somatostatin and a synthetic analogue, octreotide (Kd 0.16 +/- 0.05 nM and 0.89 +/- 0.23 nM, respectively). Somatostatin and octreotide inhibited both vasoactive intestinal peptide-mediated and forskolin-mediated cyclic AMP accumulation in IMR32 cells. Somatostatin and octreotide inhibition of signal transduction was attenuated by pretreatment of the cells with
pertussis
toxin. Octreotide inhibited proliferation of IMR32 cells by 70% in a 6-day culture. In contrast, octreotide did not exhibit high affinity binding in SKNSH cells and had no effect on cyclic AMP accumulation or on proliferation in SKNSH cells. Together, these data indicate that octreotide interacts with high affinity somatostatin receptors to modulate signal transduction and regulate proliferation in neuroblastoma cell lines. These data also suggest that somatostatin receptor expression may be an independent prognostic factor in primary neuroblastoma tumors.

x glioma hybrid NG 108-15 and neuroblastoma x fibroblast hybrid NL308 cells possess endogenous bradykinin B2 receptors and m4 muscarinic acetylcholine receptors (mAChRs), which couple to phospholipase C and adenylate cyclase, respectively. Four genetic subtypes of mAChRs differed in their effects when stimulated in NG108-15 and NL308 cells overexpressing mAChRs. Broadly speaking, the principal effects fell into two categories: the odd-numbered receptors (m1 and m3) activated phospholipase C and increased inositol trisphosphate/Ca2+, as bradykinin did, whereas the even-numbered receptors (m2 and m4) inhibited adenylate cyclase via a
pertussis
toxin (PTx)-sensitive G-protein in NG108-15 cells. But all four types of NL308 cells overexpressing each m1, m2, m3 and m4 receptor activated phospholipase C, while keeping the PTx-sensitivity in m2/m4, but not in m1/m3 receptors. Coupling to ion channel effectors showed a comparable dichotomy in NG108-15 cells, while cross-activation occurred in NL308 cells.

x glioma NG108-15 hybrid cells have been examined for the expression of opioid receptor-like receptor (ORL1). [3H]Nociceptin/orphanin FQ (OFQ) bound to the cell membrane specifically (Kd = 3.6 +/- 0.6 nM) and inhibited forskolin-stimulated cAMP accumulation (EC50 = 0.72 +/- 0.3 nM). The responsiveness of NG108-15 cells to nociceptin/OFQ was blocked by
pertussis
toxin but not by naltrindole. The inhibitory activity of nociceptin/OFQ was significantly reduced after a prechallenge with the same peptide but was not influenced by DPDPE pretreatment, indicating acute and homologous desensitization of ORL1 receptors. Naltrindole caused the overshoot of cAMP in DPDPE-pretreated cells but not in nociceptin/OFQ-pretreated cells. The results indicate that ORL1 is functionally expressed and does not cross-interact with specific ligands of the delta opioid receptor in NG108-15 cells.
