Deoxynivalenol (DON) is a group B trichothecene and a common contaminant of crops worldwide. This toxin is known to cause a spectrum of diseases in animals and humans such as
vomiting
and gastroenteritis. Importantly, DON could inhibit the synthesis of protein and nucleonic acid and induce cell apoptosis in eukaryote cells. The transduction of signaling pathways is involved in the underlying mechanism of the cytotoxicity of DON. Mitogen-activated protein kinase and
Janus kinase
/signal transducer and activator of transcription seem to be two important signaling pathways and induce the inflammatory response by modulating the binding activates of specific transcription factors. This review mainly discussed the toxic mechanism of DON from the vantage point of signaling pathways and also assessed the profiles of DON and its metabolites in humans. Importantly, we conducted a human exposure risk assessment of DON from cereals, cereal-based foods, vegetables, water, and animal-derived foods in different countries. Some regular patterns of DON occurrence in these countries are suggested based on an analysis of global contamination with DON. This review should provide further insight for the toxic mechanism study of DON and human exposure risk assessment, thereby facilitating mycotoxin control strategies.

Peficitinib is an orally administered, once-daily
Janus kinase
inhibitor currently in development for the treatment of rheumatoid arthritis. It has been shown to be a P-glycoprotein (P-gp) substrate in vitro. The effects of verapamil, an inhibitor of the efflux pump P-gp, on the pharmacokinetic profile of peficitinib were assessed in this open-label, single-center, single-sequence, crossover drug-interaction study. Twenty-four healthy volunteers received a single 150-mg dose of peficitinib on days 1 and 12 of a 14-day treatment period and received verapamil 80 mg 3 times daily on days 5-14. Repeated-dose administration of verapamil increased mean peficitinib AUC

by 27%, 27%, and 39%, respectively, and also increased the mean AUC and C

of peficitinib metabolites H1, H2, and H4. Coadministration of verapamil with peficitinib 150 mg was generally well tolerated. Overall, the most commonly reported adverse event was headache, which occurred in 5 subjects (21%); all reported adverse events were grade 1 severity, with the exception of 1 grade 2 incident of
vomiting

Inflammatory bowel diseases are chronic diseases that develop on the genetic background. They are characterized by an idiopathic, chronic course and periods of activation and remission. However, genetic and environmental factors are thought to play a role in its pathogenesis. Significant improvements in treatment strategies have been witnessed. Depending on the severity of the disease, mesalamine, immunosuppressants, anti-TNF, anti-integrin,
Janus kinase
inhibitors, and thiopurines can be used for treatment. However, these treatments have side effects such as headache, dizziness, nausea, loss of appetite, hair loss, gas,
vomiting
, rash, fever, and decreased white blood cell count. The search for treatment that may be a safer alternative, immunomodulatory, and immunosuppressive therapy has gained importance nowadays. Herbal medicine is preferred to treat a wide range of acute and chronic gastrointestinal diseases, including ulcerative colitis. Preclinical and clinical studies show that plants are promising in terms of their use in treating pathological conditions. The effectiveness of plants in treating ulcerative colitis has been determined. However, more studies are needed to explore the long-term effects of these herbal medicines. The present review presents information on medicinal plants and phytochemicals reported for use or potential of application in ulcerative colitis, a type of inflammatory bowel diseases.
