Evidence suggests that the helminth antioxidant enzyme
superoxide dismutase
(
SOD
) may play a role in parasite's defense against the cellular immune mechanisms of the host. In order to investigate this for the human parasite Onchocerca
volvulus
, the enzyme activity was characterized, the release of
SOD
by the parasite was examined, and a complete cDNA encoding the O.
volvulus
SOD
was identified. The
SOD
activity in adult O.
volvulus
was found to be 8.1 +/- 4.2 U/mg of protein. A Cu/Zn-containing enzyme was demonstrated by its sensitivity towards cyanide, azide, and hydrogen peroxide. Isoelectric focusing, combined with an enzyme activity assay, revealed two activities at pI 6.8 and 7.6, with both activities inhibited by KCN. Adult parasites, maintained in vitro, released
SOD
into the culture medium, which was detected by enzyme activity. In parallel, lactate production was measured to ensure the viability of the parasite. Oligonucleotides (based upon conserved sequences in the
SOD
genes of other organisms) and the polymerase chain reaction were used to identify a portion of the
SOD
gene from O.
volvulus
genomic DNA. A cDNA library was constructed in lambda unizapII and screened with the genomic polymerase chain reaction fragment. A complete cDNA encoding the Cu/Zn
SOD
was identified, and its nucleotide sequence was determined. Southern blot hybridization experiments indicated that the Cu/Zn
SOD
is encoded by a single-copy gene with at least one intron.

. Treatment with three cytoprotective agents at the time of

release resulted in the following mortality rates:

, 89%; ibuprofen, 50%; prostaglandin E1 (PGE1, 11%. The predominant cause of death in all treatment groups was bowel infarction, with a smaller number succumbing to either sepsis or circulatory collapse. Concomitant administration of ephedrine or indomethacin to suppress prostaglandin E1's splanchnic vasodilatory activity did not cause any increase in mortality. A trial of aspirin, to simulate PGE's antiplatelet actions, showed no reduction in mortality when compared with detorsion alone. Prostaglandin E1 and, to a lesser extent, ibuprofen, appear to have cytoprotective effects during reperfusion of bowel compromised by

, independent of their influence on the mesenteric vasculature and thrombogenesis.

The Onchocerca
volvulus
superoxide dismutase
was expressed in Escherichia coli, using a protocol designed to produce the native enzyme rather than a fusion protein. The recombinant O.
volvulus
superoxide dismutase
(rOVSOD) was found in the cytosol of the disrupted bacteria and represented > 10% of the total bacterial protein. The enzyme was purified to homogeneity using DEAE-Sepharose chromatography, followed by phenyl-Sepharose chromatography. The rOVSOD was enzymatically active which was demonstrated by its reactivity with O2.- produced either by the xanthine-xanthine oxidase system or by stimulated eosinophils. The specific activity was determined to be 4668 U mg-1. This activity could be blocked by rabbit antiserum raised against the rOVSOD. The maximal activity was obtained upon supplementation of the bacterial growth media and enzyme buffer with copper and zinc ions. Activity characteristics in the presence of inhibitors was also characteristic of a Cu/Zn superoxide dismutase. The rOVSOD has an apparent subunit molecular mass of 16,000 in SDS-PAGE. The active enzyme behaves as a dimer of 32 kDa as determined by gel filtration.

The study describes the immunohistological localization of the extracellular CuZn
superoxide dismutase
(SOD2) in the parasitic nematode Onchocerca
volvulus
. Using specific antiserum raised against a 15-amino-acid peptide from the N-terminal region of the mature protein, this enzyme is detected primarily in the intestinal epithelium of the adult worms and to a lesser extent in the muscle cells of the uterine wall. A blocking experiment with the SOD2 peptide reduced the staining significantly, confirming specificity. The localization profile of SOD2 correlates extremely well with the localization of iron deposits in the gut and uterine muscle cells of adult O.
volvulus
. The detection of SOD2 in the functional intestine of O.
volvulus
, together with the evidence that it is a secreted protein, indicates that this enzyme in parasitic nematodes is in a position to interact with host molecules. It also demonstrates the accessibility of the parasite enzyme to an inhibitor or blocking antibody.

lambda fix II genomic library. Genomic Southern blot analyses indicate single-copy genes in the O.
volvulus
genome. The O.
volvulus
cytosolic and extracellular CuZnSOD genes (Ov-sod-1 and Ov-sod-2) are separated by 0.8 kb of sequence and are convergently transcribed. Since the transcripts from all three sod genes are trans-spliced, the transcription start point of each gene was determined in a heterologous system that lacks trans-splicing machinery by in vitro transcription using Drosophila embryo nuclear extracts, followed by primer extension experiments. The ability of the 5' flanking region of the genes encoding the three Ov-SODs to promote transcription was further examined in transient transfections of Chinese hamster ovary cells. In firefly luciferase reporter assays, the Ov-sod-1 and -2 and the MnSOD (Ov-sod-3) gene promoters showed minimal, strong, and moderate levels of activity in these cells, respectively. Both Ov-sod-2 and -3 gene promoter regions showed an initial increase in activity in response to 5' deletions. The results from the in vitro transcription experiments and the luciferase reporter assays were consistent and suggest the presence of Inr-like elements in the promoter regions of the Ov-sod genes.

) found in the excretory/secretory products of adult worms. Contrary to previous studies, the OvEC-
SOD
was found to have a 19 amino acid long signal peptide that is cleaved off during the process of maturation. To validate this result, we designed a novel method based on Caenorhabditis elegans cup5(ar465) mutants to specifically evaluate signal peptide-mediated secretion of nematodal proteins. Following purification, the recombinant OvEC-
SOD
was active as a dimer. Site-directed mutagenesis of the three cysteines present in the OvEC-

shows that enzyme activity is markedly reduced in the Cys-192 mutant. A homology model of the OvEC-
