Using IFAT, it has been shown that isolated egg-shells and uterine fluid of Dipetalonema viteae are the most potent antigens in heterologous systems using human sera from patients infected with
Wuchereria
bancrofti, Onchocerca
volvulus
and Loa loa, as well as in homologous systems using sera from animals infected with D. viteae. It is suggested that these antigens are unlikely to be highly species-specific, and that anatomical isolation of antigens is a necessary prerequisite to immunochemical analysis aimed at the preparation of a "pure" antigen.

Systematic lymphographic studies in 13 cases of infection with Onchocerca
volvulus
have enabled a "lymphographic diagram" to be made for each infected patient. The images obtained are similar to those observed in
wuchereriasis
. The microfilariae in onchocerciasis, however, are not transmitted in the lymphatic system, and the lesions, which vary according to the degree of infestation, show little progression. They are found mainly in the inguino-crural ganglia which are the first main ganglionic relay system for the lower limbs.

Sequential extraction of Onchocerca

from skin nodules with buffers of different ionic strength resulted in a mixture of antigens which reacted in double immunodiffusion with sera from proven cases of onchocerciasis, the number of precipitation lines varied from 1 to 5. Triton X-100 detached mainly cationic antigen proteins. It reduced the molecular size of many antigenic proteins, but did not effect their antigenicity. Labelling of the extracted proteins with high specific activity 125I did not destroy much antigenicity. Strong cross-reactivity between Onchocera

bancrofti, Mansonella perstans/Loa loa and of Sudanese control sera. Sera of all groups contained IgM reacting with Ov33-GST and with OvAg. There was no IgG1 response to Ov33-GST. IgG1 responses to OvAg were only detected in filariasis sera. IgG2 and IgG3 responses were not detectable or marginal in all groups. The IgG4 reaction of onchocerciasis patients to Ov33-GST and to OvAg was high, whereas few other filariasis sera contained IgG4 antibodies to Ov33-GST and to OvAg. A serodiagnostic test for onchocerciasis based on detection of IgG4 to Ov33-GST had a sensitivity of 93.3% and a specificity of 96%. An epitope common to Ov33 and to the homologous proteins of other filarial species was demonstrated with a monoclonal antibody. Purified Ov33-MBP fusion protein was used to follow the development of the antibody response of four chimpanzees experimentally infected with O.
volvulus
. The data indicates that antibodies to Ov33 are induced by developing worms and later parasite stages.

A recombinant clone, WbN1, isolated from a genomic expression library of

bancrofti and showing restricted specificity at the DNA level (Southern and PCR analyses) for
Wuchereria
bancrofti and Brugia malayi has been previously described. Sequence analysis of WbN1 indicated that it had notable similarity to myosin. Further characterization using in situ hybridization has localized the mRNA in the muscle of the adult parasite and in the microfilariae. Rabbit polyclonal antiserum, raised against the recombinant WbN1 fused to the maltose-binding protein, recognized a 200-kDa polypeptide in immunoblots containing B. malayi antigen extracts. The same antibody also recognized myosin extracted from Brugia pahangi, Onchocerca
volvulus
, and Caenorhabditis elegans. Localization using the rabbit antiserum revealed the presence of the antigen in the adult muscle tissue and in the microfilariae; the same antibody inhibited the binding of a monoclonal antibody 28.2 (directed toward MHC B of C. elegans myosin) to the recombinant WbN1 antigen and also to purified C. elegans myosin. Based on homology data, structural location, competitive ELISA, and immunoblot we conclude that WbN1 is related to myosin or a similar myofibrillar protein.

A PEG-ELISA was used to demonstrate parasite specific immune complexes in a significant proportion (25/26) of Onchocerca volvulus infection sera from Sierra Leone. The parasite antigen was detected using a peroxidase-conjugated rabbit serum raised to the bovine parasite O. gibsoni. Controls including European control serum, endemic control serum and Rh+ sera gave consistently low readings. Characterization of the parasite component in the immune complexes by Western blotting demonstrated a heat stable antigen of M(r) 46,000. This antigen was not present in the circulating immune complexes (CIC) prepared from patients with
Wuchereria
bancrofti infection, but a cross-reactive molecule of the same size was weakly recognized in the CIC of Loa loa and Mansonella perstans infected patients. No association between the level of parasite specific CIC and clinical disease was observed in the O.
volvulus

Pulmonary infection with D. immitis is an infrequently reported cause of lung disease in humans. Approximately 80 cases have been reported in the United States, with increasing numbers reported in recent years. The largest single series from one institution is a series of 6 patients from Ochsner Clinic in New Orleans, Louisiana. Although our series consisted of 3 females and no males, the disease is detected and diagnosed in males twice as often. The usual age range is 40 to 60 years. No case has yet been seen in children. Unfortunately, it is essentially impossible to diagnose by clinical means. The definitive diagnosis can be made quite easily, however, by histologic examination which reveals the worm within the lesion. It should be noted that the parasites are seen in a minority of sections; therefore, diagnosis may be missed. As a result, given the setting of pulmonary infarction with granulomatous response, multiple sections of the lesion should be examined. Other larval worms also occurring in the lung which might be confused with D. immitis include Enterobius vermicularis,
Wuchereria
bancrofti, Brugia malayi, and Onchocerca
volvulus
. These, however, are much less common and are morphologically distinct from D. immitis. Also, it is easily distinguished from ascarids, strongyloids, and hookworm larvae which are much smaller than the dirofilarial worms and do not contain reproductive organs. In North American, the occurrence of fragments of nematode within a pulmonary infarct is essentially pathognomonic for PD.
