Drugs which reduce central catecholamine activity increased the tremorigenic action physostigmine (0.25 mg/kg) in the rat. The potentiation of physostigmine tremor induced by these neuroleptic drugs was quantitated measuring the ratio of post- to prephysostigmine motor activity; this ratio may be useful index of catecholamine/cholinergic balance. Of the compounds tested, chlorpromazine HC1 (15 mg/kg) and reserpine (10 mg/kg) had the greatest potentiating effect on the tremor-producing action of physostigmine. LDOPA (500 mg/kg) reduced the tremor induced by physostigmine in rats pre-treated with reserpine, but not in rats pre-treated with chlorpromazine.

Effects of MKC-242 (5-[3-[((2S)-1,4-benzodioxan-2-ylmethyl)amino]propoxy]-1,3-b enzodioxole HC1), a novel 5-HT1A-receptor agonist, and reference compounds on wrap restraint stress-induced defecation were evaluated in rats. Wrapping restraint stress increased defecation in rats. The increase was attenuated by putative 5-HT1A-receptor agonists, MKC-242 and 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT). The suppressive effect of MKC-242 on wrap stress-induced defecation was antagonized by prior administration of a 5-HT1A-receptor antagonist, WAY100135. MKC-242 did not affect spontaneous defecation and 5-HT-induced defecation. Diazepam and amitriptyline also significantly reduced the stress-induced defecation. However, amitriptyline showed a potent anti-cholinergic effect in the oxotremorine-induced tremor test and reduced spontaneous defecation. In contrast to MKC-242 and 8-OH-DPAT, buspirone and tandospirone tended to suppress the increase at high doses. A major metabolite of buspirone and tandospirone, 1-(2-pyrimidinyl)piperazine, antagonized the suppressive effect of MKC-242. These findings suggest that stimulation of 5-HT1A receptors reduces stress-induced defecation but not spontaneous and 5-HT-induced defecation and that MKC-242 may be useful for the treatment of irritable bowel syndrome.

Two extraction reagents, hydrochloric acid (HCl) and acid ammonium oxalate solution (Tamm's reagent), were used to evaluate the redox state of iron in municipal solid waste (MSW) with different deposit ages. Orthogonal experiments were conducted to optimize the extraction conditions for extractable iron speciation (ferric and ferrous) in MSW. The optimal extraction conditions for HCl were determined as follows: the liquid-to-solid ratio was set at 100, and then the samples were extracted at the shaking speed of 200 rpm at 35 degrees C for 60 min by 1.00 M HCl. For Tamm's reagent, the optimal extraction conditions were extracted at the shaking speed of 175 rpm at 30 degrees C for 12 h with the same liquid-to-solid ratio. However, Tamm's reagent extraction is much more laborious and time-consuming. Thus the HC1 extraction might be a better choice for the evaluation of the redox state of iron in MSW. The results also showed that the yield of extractable iron increased with deposited age. About 60-83% of extractable iron was presented as ferrous in the MSW deposited for 1-8 years. This study supplied a tool for investigating the role of iron on the fate of pollutants in the landfill.
